Evaluation of the average performance of topcrosses among food and grain type soybean BABT
Yokomizo,Gilberto K.; Vello,Natal A..
The food type soybean (exotic) are, in general, non-adapted to the Brazilian growing conditions. A breeding strategy to develop adapted food type soybean should involve crossing exotic with the available grain type genotypes. We evaluated the performance of topcrosses among food type and grain type soybeans. Best progenies showed different performance rates in each environment and they were not always the same in the different environments and joint analysis of lodging (L), plant height at maturity (PHM) and agronomic value (AV) was essential to avoid selection of plants with good performance for one trait and bad for another.) Piracicaba autumn environment (PA) caused low individual plant yield performance (IPY), and consequently, cultivation was not...

Bivalent RNA interference to increase isoflavone biosynthesis in soybean (Glycine max) BABT
Jiang,Yina; Hu,Yanlin; Wang,Biao; Wu,Tianlong.
In this work, a bivalent RNA interference (RNAi) plant-transformation vector was constructed to silence both the flavanone 3-hydroxylase (F3H) gene and the flavone synthase II (GmFNSII) gene in soybean (Glycine max). Two further unit RNAi vectors were constructed for each of these two genes. RNAi-mediated suppression of these genes effectively regulated flavone and isoflavone production in hairy roots that arose from soybean cotyledons transformed with Agrobacterium rhizogenes ATCC15834. Notably, the bivalent RNAi vector had a significantly higher effect for increasing isoflavone production compared with the two unit RNAi vectors. The study highlighted molecular methods that could be used to enhance isoflavone production in soybean and demonstrated the...

Soybean parent selection based on genetic diversity BABT
Carpentieri-Pípolo,Valéria; Pípolo,Antônio Eduardo; Silva,Flávio André Martins da; Petek,Marcos Rafael.
Thirty-four soybean lines were assessed for twelve traits. The genetic distances were estimates using multivariate techniques, to identify parents to be included in breeding programs for hybridization. Grouping by the Tocher method, from generalized Mahalanobis distances, divided the 34 lines into four groups. The most important agronomic traits, weight of seeds per plot, plant height, height of first pod and days to maturity were considered when recommending for crossing. The following crosses were recommended based on the genetic divergence and the key agronomic traits: lines 23, 10, 2, 27 and 25 (group I) with genotype 6 (group II) and genotype 16 (group III). Thus only ten crosses would be made, representing only 2% of the total crosses which could be...

Hydrothermal treatments in the development of isoflavone aglycones in soybean (Glycine max (L.) Merrill) grains BABT
Carrão-Panizzi,Mercedes C.; Goés-Favoni,Silvana Pedroso de; Kikuchi,Akio.
Studies were carried out to enhance the development of isoflavone aglycones in soybean. Grains of two soybean cultivars, BR 36 and IAS 5, 115 and 278 mg/100g of total isoflavone, respectively, were treated hydrothermalyl at 45, 60 and 85° C for 5, 30 and 60 minutes. Pre-treatments of grains at 60° C for 60 minutes allowed a considerable increase of the isoflavone aglycones. Non-treated grains of BR 36 and IAS 5 showed 1.2 mg/100g of genistein, after hydrothermal treatments, which increased to 12 and 53 mg/100g, in each variety, respectively. At higher temperature (85 ° C) there was a decrease of the aglycones due to inactivation of beta-glycosidases. Malonyl compounds were also reduced at higher temperatures. In processing functional soybean foods,...

Biological and physicochemical properties of cowpea severe mosaic comovirus isolated from soybean in the State of Paraná BABT
Bertacini,Paula V.; Almeida,Álvaro M.R.; Lima,J. Albérsio A.; Chagas,Cesar M..
Soybean plants with symptoms of bud blight were growing close to cowpea with severe symptoms of mosaic associated with blisters in the leaves. A group of plants of both species were collected and used for etiological studies. This kind of symptom in soybeans was common in certain areas of the State of Paraná, induced by tobacco streak ilarvirus. Host range, serological reaction, particle morphology and size, protein and nucleic acid analysis, and transmission by beetles from species Cerotoma arcuata Oliv. showed that the virus involved was cowpea severe mosaic comovirus. This is the first report on the occurrence of this virus in soybean plants in the State of Paraná. Results using indirect ELISA showed that in cowpea the relative virus concentration was...

Histology and ultrastructure of the fat body of Anticarsia gemmatalis (HÜBNER, 1818) (LEPIDOPTERA: NOCTUIDAE) BABT
Carvalho,Renata Baldo Rodrigues; Andrade,Fábio Goulart de; Levy,Sheila Michele; Moscardi,Flávio; Falleiros,Ângela Maria Ferreira.
The aim of this study was to analyze the morphologically the fatty body of fourth-instar Anticarsia gemmatalis larvae under light, transmission and scanning electron microscopy. Two distinct portions of the fat body were detected: the parietal (PA) and perivisceral (PV). The PA, the parietal portion, presented a long-stripped shape located below the tegument and lateral to the digestive tube. The PV, rarely observed, was in dorsal region, adhered to digestive wall. Both the portions were constituted of only one cellular type, the trophocytes. These cells in the PA were organized in one layer of thickness showing cylindrical contiguously morphology, whereas the PV was comprised by a mass of small cells, superposed as clusters. Both the portions were covered...

Effect of salt on the growth and metabolism of Glycine max BABT
Queiroz,Helena Müller; Sodek,Ladaslav; Haddad,Claudia Regina Baptista.
Soybean plants cultivated with 50, 100 and 200 mM of NaCl, revealed that root growth was less affected by salinity than shoots. Salinity led to a reduction in leaf area and an increase in water content of the roots. These factors could contribute to the adaptation of the plant, improving its hydration. Although nitrate and free amino acid levels were reduced by salt treatment in roots, protein content of leaves was not altered. Salinity led to alterations in xylem amino acid composition, with increases in Ser, Ala, Gaba and Pro and a decrease in Asn. Similar changes were seen for Asn and Ser in roots together with a much stronger increase in Gaba. It is suggested that the decline in Asn reflects its conversion to Ala and Gaba (via Glu) in the roots while...

Effects of a methanolic fraction of soybean seeds on the transcriptional activity of peroxisome proliferator-activated receptors (PPAR) BJMBR
Carrara,V.S.; Amato,A.A.; Neves,F.A.R.; Bazotte,R.B.; Mandarino,J.M.G.; Nakamura,C.V.; Filho,B.P.D.; Cortez,D.A.G..
Since the anti-inflammatory, antidiabetic and hypolipidemic effects of soy isoflavones may be mediated by activation of peroxisome proliferator-activated receptors (PPAR), the present study investigated whether the methanolic fractions obtained from soybean seeds (E1) and soybean seed coats with hypocotyls (E2) could influence PPARα, PPARγ and PPARβ/δ transcriptional activity. The isoflavones from E1 and E2 were quantified by HPLC analysis. E1 and E2 were rich in isoflavones (daidzin, glycitin, genistin, malonyldaidzin, malonylglycitin, malonylgenistin, daidzein, glycitein, and genistein). Moreover, E1 and E2 showed no evidence of genetically modified material containing the gene CP4 EPSPS. To investigate PPAR transcriptional activity, human promonocytic...

Rhizobia amylase production using various starchy substances as carbon substrates BJM
Oliveira,Arlem Nascimento de; Oliveira,Luiz Antonio de; Andrade,Jerusa Sousa; Chagas Júnior,Aloisio Freitas.
Six isolates of indigenous rhizobia of Central Amazonia were screened for the production of amylases in liquid media using various starchy substances as carbon sources. All rhizobia strains could produce more extracellular protein, biomass and amylases with the different kinds of carbon substrates. Among the carbon sources tested maltose was the best substrate for protein and amylase production. In general, peach palm flour and corn starch (maizena®) were also considered to be good carbon sources for rhizobia amylases. On the other hand, the biomass production by the rhizobia isolates was higher in the presence of oat flour. INPA strain R-926 was a good amylase producer in maltose (1.94 U) and corn starch (0.53 U) media. INPA strain R-991 was also a good...

Arbuscular mycorrhizal fungal communities in the rhizosphere of a continuous cropping soybean system at the seedling stage BJM
Cui,Jiaqi; Bai,Li; Liu,Xiaorui; Jie,Weiguang; Cai,Baiyan.
Abstract Arbuscular mycorrhizae (AM) fungi play a crucial role in the growth of soybean; however, the planting system employed is thought to have an effect on AM fungal communities in the rhizosphere. This study was performed to explore the influence of continuous soybean cropping on the diversity of Arbuscular mycorrhizal (AM) fungi, and to identify the dominant AM fungus during the seedling stage. Three soybean cultivars were planted under two and three years continuous cropping, respectively. The diversity of AM fungi in the rhizosphere soil at the seedling stage was subsequently analyzed using polymerase chain reaction (PCR)-denaturing gradient gel electrophoresis (DGGE). The results showed that an increase in cropping years improved the colonization...

Quality of beef burger with addition of wet okara along the storage Ciência e Agrotecnologia
Oliveira,Rafaela Bergmann Strada de; Lucia,Flávia Della; Ferreira,Eric Batista; Oliveira,Roseane Maria Evangelista de; Pimenta,Carlos José; Pimenta,Maria Emília de Sousa Gomes.
ABSTRACT Combining the consuming of agricultural residues with satisfying consumers is a challenge that may result in financial income for the frozen meat industries, besides generating products with different nutritional value and satisfactory technological quality. Thus, we aimed at elaborating and assessing the physical, chemical and microbiological characteristics of frozen raw and fried beef hamburgers with addition of okara in its wet form throughout their storage over a period of 120 days. The inclusion of okara was proportional to the reduction in the content of protein, and to the increase of the lipid and moisture amount in the fried and raw formulations. Okara increased the luminosity and dimmed along the storage period. In the raw formulations...
