Gene expression promoted by the SV40 DNA targeting sequence and the hypoxia-responsive element under normoxia and hypoxia BJMBR

The main objective of the present study was to find suitable DNA-targeting sequences (DTS) for the construction of plasmid vectors to be used to treat ischemic diseases. The well-known Simian virus 40 nuclear DTS (SV40-DTS) and hypoxia-responsive element (HRE) sequences were used to construct plasmid vectors to express the human vascular endothelial growth factor gene (hVEGF). The rate of plasmid nuclear transport and consequent gene expression under normoxia (20% O2) and hypoxia (less than 5% O2) were determined. Cardiovascular, respiratory and metabolic responses to temperature and hypoxia of the winter frog Rana catesbeiana BJMBR
The objective of the present study was to determine the effects of hypoxia and temperature on the cardiovascular and respiratory systems and plasma glucose levels of the winter bullfrog Rana catesbeiana. Body temperature was maintained at 10, 15, 25 and 35oC for measurements of breathing frequency, heart rate, arterial blood pressure, metabolic rate, plasma glucose levels, blood gases and acid-base status. Reducing body temperature from 35 to 10oC decreased (P<0.001) heart rate (bpm) from 64.0 ± 3.1 (N = 5) to 12.5 ± 2.5 (N = 6) and blood pressure (mmHg) (P<0.05) from 41.9 ± 2.1 (N = 5) to 33.1 ± 2.1 (N = 6), whereas no significant changes were observed under hypoxia. This review describes the ways in which the primary bradycardia and peripheral vasoconstriction evoked by selective stimulation of peripheral chemoreceptors can be modified by the secondary effects of a chemoreceptor-induced increase in ventilation. The evidence that strong stimulation of peripheral chemoreceptors can evoke the behavioural and cardiovascular components of the alerting or defence response which is characteristically evoked by novel or noxious stimuli is considered. The objective of this study was to observe possible interactions between the renin-angiotensin and nitrergic systems in chronic hypoxia-induced pulmonary hypertension in newborn piglets. Thirteen chronically instrumented newborn piglets (6.3 ± 0.9 days; 2369 ± 491 g) were randomly assigned to receive saline (placebo, P) or the AT1 receptor (AT1-R) blocker L-158,809 (L) during 6 days of hypoxia (FiO2 = 0.12). Hypoxia elicits hyperventilation and hypothermia, but the mechanisms involved are not well understood. The nitric oxide (NO) pathway is involved in hypoxia-induced hypothermia and hyperventilation, and works as a neuromodulator in the central nervous system, including the locus coeruleus (LC), which is a noradrenergic nucleus in the pons. The LC plays a role in a number of stress-induced responses, but its participation in the control of breathing and thermoregulation is unclear. Nitrate is reported to improve tolerance of plants towards oxygen deficiency enabled by waterlogging of the root system, but the mechanism underlying the phenomenon remains poorly understood. We studied the metabolism of nitrate in roots exposed to hypoxia, using soybean plants growing in a hydroponic system after suspending aeration and covering the surface of the nutrient solution with mineral oil. Nitrate depletion from the medium was more intense under hypoxia than normoxia, but in the presence of chloramphenicol, consumption under hypoxia was significantly reduced. The transport of organic N compounds to the shoot in the xylem sap of nodulated soybean plants was investigated in an attempt to better understand the changes in N metabolism under root hypoxia (first 5 days of flooding), with different sources of N in the medium. NO3- is beneficial for tolerance of plants to waterlogging, whereas other N sources such as NH4+ and NH4NO3, are not. Nevertheless, in the presence of NH4+ high levels of amino acids were transported in the xylem, consistent with its assimilation. Dissolved oxygen and ammonia levels in water that affect plasma ionic content and gallbladder bile in silver catfish Ciência Rural

Ionic contents (Na+, K+ and Cl-) of plasma and gallbladder bile (GB) of juveniles silver catfish, Rhamdia quelen (156.1±0.2g, 28.2±0.3cm), were determined in three different times (0, 6 and 24h) after exposure to: a) control or high dissolved oxygen (DO = 6.5mg L-1) + low NH3 (0.03mg L-1); b) low DO (3.5mg L-1) + low NH3; c) high DO + high NH3 (0.1mg L-1); and d) low DO + high NH3. High waterborne NH3 or low DO levels increased plasma and GB ion levels. These parameters might have followed different mechanisms to affect osmoregulation since a synergic effect of these variables was detected.
