Polymorphic DNA bands were identified between a near iso-isogenic line of wheat carrying both stem (Sr39) and leaf (Lr35) rust resistance genes and the recurrent line Thatcher (Tc) which lacks these genes. Both resistance genes are located on a translocated chromosomal segment derived from Aegilops speltoides and thus are genetically linked. The primers used to generate polymorphic bands were 3'-anchored inter-simple sequence repeat primers which identified genomic microsatellites with a repeated motif of 3 nucleotides in length. Short chain citronellyl esters were synthesized by a new microbial lipase from Rhizopus sp strain isolated and lipase produced at UNICAMP, Brazil. Direct esterification and transesterification reactions have been performed to produce citronellyl acetate and butyrate in a free-solvent system and with n-hexane in reaction medium. Reaction mixture for direct esterification and transesterification was carried out at 45ºC in equimolar concentration of substrates: acid or ester and alcohol. The protein lysozyme was deposited onto a permeable support comprising chemically functionalized glass fiber. The main objective of this study was to set a stable organic net with no effect on the medium bed permeability and a preliminary test the activity of this enzyme under immobilized conditions. The film formation is followed by atomic force microscopy (AFM) surface imaging. The effect on the bacteria Escherichia coli was tested using a simple microfiltration column. Background: The AdEasy system is a fast-track system for generating recombinant adenoviruses using the efficient homologous recombination machinery between shuttle and adenovirus backbone plasmids in Escherichia coli BJ5183 cells. The influences of water activity and solvent hydrophobicity on the kinetics of the lipase-catalyzed enantioselective esterification of flurbiprofen with n-butanol were investigated. The solvent effect was not similar for lipases from Candida rugosa (Crl), Mucor javanicus (Mjl), and porcine pancreas (Ppl). The lipase-catalyzed reaction rates in different solvents across a wide range of water activities revealed that the Ppl-catalyzed reaction exhibited no enantioselectivity and no substantial water activity or solvent dependence. Response surface methodology (RSM) was used for statistical optimization of jiean-peptide (JAA) production by Bacillus subtilis ZK8 cells adsorbed on wood chips to form a novel fermentation system. The Plackett-Burman design was used in the first step to evaluate the effects of eight factors, including six fermentation medium components and two cell adsorption conditions. Complementary DNA (cDNA) is valuable for investigating protein structure and function in the research of life science, but it is difficult to obtain by traditional reverse transcription. In this study, we employed a novel strategy to clone the human leukemia inhibitory factor (hLIF) gene cDNA from genomic DNA directly isolated from the mucous membrane of mouth. The hLIF sequence can be acquired within a few hours by means of amplification of each exon and splicing using overlap-PCR. Thus, the new approach developed in this study is simple, time- and cost-effective, and it is not limited to particular gene expression levels of each tissue. |

In the present report, citric acid production from raw glycerol in two fed-batch systems by acetate negative-mutants of Yarrowia lipolytica: Wratislavia 1.31 and Wratislavia AWG7 was compared. In the system, in which the total glycerol concentration was 200 g∙L-1, the substrate was added by pulsed additions, and in the other, in which the total glycerol concentration was 300 g∙L-1, the substrate was added at a constant feeding rate of 1.4 g∙h-1.
