Herpes and herpes-like virus infections have been reported in various marine mollusc species associated with high mortality rates. Following the characterisation and genome sequencing of ostreid herpesvirus 1 (OsHV-1), specific diagnostic tools have been developed based on conventional PCR techniques or in situ hybridisation. We have now developed a real-time PCR assay for rapid, sensitive and quantitative detection of OsHV-1, and compared it with a conventional PCR technique described previously. The new assay utilised SYBR® Green chemistry with specific primers C9/C10 targeting the C region, which is present in two copies in the OsHV-1 genome and encodes two proteins of unknown function. Little information is available on viral infections that affect bivalve molluscs. Such a lack of data is due ta a certain inadequacy of the diagnosis methods that are employed when massive mortality events occur. Most laboratories involved in mollusc pathology still analyse samples through light microscopy. World-wide, there is thus currently a lack of information concerning the occurrence of bivalve herpesvituses. This is probably due to the lack of suitable diagnostic tools. The basic method for identification and examination of suspect samples is predominantly histopathology. To find biocatalyst allowing depolymerization of new polysaccharides, one needs to get a sensitive and well adapted method to a screening program. This led us to compare biochemical methods of detection of the depolymerization. Currently used methods such as reducing sugars assays, double bond monitoring or molecular weight determination were tested to follow the kinetic of depolymerization with different enzyme / polysaccharide couples. OsHV-1 was the first herpesvirus to be identified in an invertebrate host and is associated to mortalities in the Pacific oyster (Crassostrea gigas) and other economically important bivalve species. Vibrio nigripulchritudo was shown to contain saprophytic and pathogenic strains among New Caledonian isolates. We established a method to map the development of V. nigripulchritudo in pond sediments at three different genetic levels: the species level, then at the pathogenic cluster level and finally at the plasmid level, present only in all highly pathogenic isolates.
