ABSTRACT The present work explores in vivo physiological, morphological and chemical features during full hydration, desiccation and rehydration of the filmy fern Hymenophyllum dentatum with two main objectives: 1) to get further insight about the mechanisms underlying its desiccation tolerance, and 2) to understand how this plant manages mechanical stress induced by water loss and recovery. Abstract The naturally occurring wild barley mutant eibi1/hvabcg31 suffers from severe water loss due to the permeable leaf cuticle. Eibi1/HvABCG31 encodes a full ATP-binding cassette (ABC) transporter, HvABCG31, playing a role in cutin deposition in the elongation zone of growing barley leaves. The eibi1 allele has pleiotropic effects on the appearance of leaves, plant stature, fertility, spike and grain size, and rate of germination. Comparative transcriptome profile of the leaf elongation zone of the eibi1 mutant as well as its isogenic wild type showed that various pathogenesis-related genes were up-regulated in the eibi1 mutant. Seeds of Magnolia ovata were dried to different water contents to assess the viability and transcript abundance of genes related to seed development, cell cycle, cytoskeleton and desiccation tolerance.The expression of development, cell cycle and cytoskeleton relative genes (ABI3, CDC2-like and ACT2) alone could not explain the germination behaviour of M. ovata seeds in relation to drying damage. Irrespective of their initial water content, the seeds performed in the same way during the initial period of germination and the deleterious effects of desiccation only occurred in later stages. Some comments about seeds that show sensitivity to desiccation and short viability period, called recalcitrants, were made. Further studies showed that there was a gradient of sensitivity to desiccation and an intermediate class was proposed. The research demonstrated different factors related to desiccation tolerance such as ABA, proteins and sugars. It was analyzed the research of recalcitrant seeds in Brazil, that started around 1950 and nowadays the major aspects studied are recalcitrant seeds identification, seed low temperature and drying tolerance, storage capacity under different rooms and packages, chemical composition and viability during storage, fungicide treatment efficiency on seed conservation. |

The objective of this study was to evaluate the physiological quality of babassu (Orbignya phalerata Mart.) embryos as a function of desiccation. The fruits were placed in a convection oven at 37±2°C for 0, 3, 6, 9, or 12 days. After these periods, the moisture contents of the fruits and seeds were determined. A reduction in the water content occurred more rapidly in the fruits than in the seeds. The decrease in seed moisture content from 12.59% (on the basis of fresh weight) to 4.56% did not affect the percentage of germinated zygotic embryos; however, we did observe a decline in the vigor as the seed moisture content decreased. Embryos with a 4.56% moisture content developed into seedlings of shorter lengths after 30, 60, and 90 days of in vitro growth.... |

Considering the importance of water content for the conservation and storage of seeds, and the involvement of soluble carbohydrates and lipids for embryo development, a comparative study was carried out among the seeds of Inga vera (ingá), Eugenia uniflora (pitanga), both classified as recalcitrant, and Caesalpinia echinata (brazilwood) and Erythrina speciosa (mulungu), considered as orthodox seeds. Low concentrations of cyclitols (0.3-0.5%), raffinose family oligosaccharides (ca. 0.05%) and unsaturated fatty acids (0-19%) were found in the seeds of ingá and pitanga, while larger amounts of cyclitols (2-3%) and raffinose (4.6-13%) were found in brazilwood and mulungu, respectively. Desiccation tolerance in seeds depends on the species, development stage and drying conditions, especially the water removal rate. Coffea seeds are considered of intermediate performance, because they tolerate relative dehydration compared to orthodox seeds and are sensitive to low temperatures. The objective of this study was to verify the effect of different drying rates on the viability and storability of Coffea canephora seeds.
