Merchant ships discharging unwanted marine species in close proximity of a French aquaculture area: Risks involved ArchiMer
Masson, Daniel; Thomas, Gerard; Genauzeau, Sylvie; Le Moine, Olivier; Derrien, Annick.
The most important oyster farming area in Europe is in a close proximity of two medium size merchant ports. Cargo ships deballast in this area before loading, releasing unwanted or noxious marine species. During a sampling campaign aboard these arriving ships, we found in some ballast water samples a huge number of potentially toxic dinoflagellates and some potentially pathogenic bacteria. A model was applied to find the potential geographical spread of the discharged ballast water. This model predicts the water to reach highly vulnerable shellfish farmed areas in six to eight days.

Bacteries ichtyopathogenes en mariculture ArchiMer
Vigneulle, Martine.
Bacterial infections occurred during the development of marine fish farming of species such as turbot, sea-bass, sea-bream or salmon and trout. The main-specific bacteria involved in these diseases observed belong to 3 groups: the Vibrionaceae for Vibrio anguillarum (type 408) and Aeromonas salmonicida , the causative agents of respectively vibriosis and furinculosis; the Corynebacteriaceae for Renibacterium salmoninarum , responsible for bacterial kidney disease (BKD), the Enterobacteriaceae for Yersinia ruckeri , the causative agent of enteric redmouth disease. The clinical signs typical of those diseases always depend on the rearing and environmental conditions. The losses resulting from these bacterial diseases and the persistence of the bacteria in...

Minet, Jacques; Sauvager, F; Cormier, M.
Illnesses due to marine bivalvia consumption are mostly associated with viruses as causative agents. In more than the half of cases, this agent remains unidentifies, nevertheless. So far as bacteria are concerned, Salmonella are found firstly. Campylobacters represent a major cause of gastrointestinal disease, with Salmonella, among humans. Whether campylobacteriosis may be acquired from shellfishes remains unclear. We undertook to comparatively appreciate the survival of Campylobacter jejuni/coli and Salmonella typhimurium in seawater and contaminated mussels. Bacterial enumerations where performes using microbial cultures and specific (with immunofluorescence) direct viable counts (of substrate responsive bacteria). During laboratory contamination...

Ecologie des vibrions pathogenes ArchiMer
Dodin, A; Dosso, M; Guillou, M.
There exists no biochemical test allowing to define the pathogenicity of a bacterium, still less for a Vibrionaceae. Two essential characters are to be retained to hold responsible an isolated germ, for an illness: 1) the binding factor, the identification is difficult but a double diffusion of agar should simplify the method; 2) the search for the choleric toxin or a common part of the choleric toxin (toxin LT or ST of E. coli ). Here again, simplified techniques may be used. The acquirement of a plasmid responsible for the binding factor or for the toxin production of the non-pathogenic Vibrionaceae in the origin of certain epidemic diseases in France and in the world. Frequently, epidemics due to common germs (Aeromonas hydrophila of Africa) lead to...

A white spot disease-like syndrome in the Pacific blue shrimp (Litopenaeus stylirostris) as a form of bacterial shell disease ArchiMer
Goarant, Cyrille; Brizard, Raphael; Marteau, Anne-laure.
In May 1997, some white lesions evoking the white spot syndrome disease were observed in Litopenaeus stylirostris broodstock in New Caledonia. The occurrence of these lesions was neither associated with mortality, nor with histological evidence of white spot syndrome baculovirus (WSBV), The evidence suggests that these lesions result from a form of bacterial disease and are associated with an increased bacterial flora on the outer surface of the cuticle, as well as an increased incidence throughout the molt cycle. A microscopic observation of these lesions allows them to be differentiated from WSBV-associated lesions.

Etude preliminaire ArchiMer
Munro, P; Bianchi, M.
Cells of two serotypes of Escherichia coli (O26B6 and 0111B4) were counted using epifluorescence and immunofluorescence microscopy (nonavalent serum) and plate count on specific medium. Values from indirect counts were 10 fold smaller than those from both microscopic counts. In a sample of sea water from an area receiving a fecal pollution, the highest number of bacteria was obtained by epifluorescence microscopy. In that case, the number of cells counted by immunofluorescence microscopy was 10 fold smaller than the number of bacteria able to grow on the medium. It could be due to: the diversity of the natural community, the possibility to other bacteria than E. coli to grow on the used culture medium, the restrictiveness of the serum for "pathogenic...

Isolation and characterization of a pathogenic bacterium specific to Manila clam Tapes philippinarum larvae ArchiMer
Nicolas, Jean-louis; Ansquer, Dominique; Cochard, Jean-claude.
A bacterial strain belonging to the genus Vibrio was involved in a recurrent outbreak of disease in Manila clam Tapes philippinarum larvae at a commercial hatchery, This vibrio, named VTP for convenience, was not related to the known pathogenic vibrios and possessed unusual characteristics, Only Manila clam larvae and postlarvae were affected by the disease, Oyster and scallop larvae were not affected by vrp, Survival of vrp was found to be of short duration in seawater but even a very small number of cells (1 cell per 5 ml) could initiate the disease, VTP did not grow on thiosulfate-citratebile-sucrose agar and was resistant to chloramphenicol. An epidemiological study utilizing bioassays revealed the presence of VTP in adult bivalves and in the...

Fluorescent antibody enumeration of Vibrio cholerae in the marine environment. ArchiMer
Brayton, P; Roszak, D; Palmer, L; Huq, S; Gimes, D.
From microcosm studies, V. cholerae and related potential human pathogens enter a viable but nonculturable state. Direct viable counts by epifluorescent microscopy consistently remain higher than corresponding plate counts. Thus, the assumption that pathogens "die-off" or "decay" in the marine environment must be re-evaluated, since stressed or nutrient-starved cells are unable to grow and be enumerated by standard plate count methods. Indirect immunofluorescent microscopy offers a more sensitive detection system for environment sampling for human pathogens. Field indicate that cultures positive for V. cholerae O1 are also positive by fluorescent antibody staining; however, the reverse was not necessarily true. By adapting epifluorescent techniques for...

The dispersal of nitrogen-fixing Enterobacteriaceae from sewage into the waters and sediments of Morecambe Bay, UK. ArchiMer
Jones, K; Betaieb, M.
The sediments of Morecambe Bay contain large numbers of salinity-tolerant, nitrogen-fixing Enterobacteriaceae (NFEs) mainly Klebsiella pneumoniae ) which appear to be derived from sewage. Sewage effluents, regardless as to whether they have received primary or secondary treatment, contain huge numbers of NFEs with salinity-tolerant and freshwater strains present in roughly equal quantities. The bacteria are discharged into the rivers and estuaries and carried out to sea. Once in the sea the salinity-tolerant strains retain their viability better than the freshwater strains so that the incoming tide, which doses the sediments, contains mainly salinity-tolerant NFEs. It is probable that similar processes occur all round the U.K. coast, as inshore coastal...

Bonnefont, J; Lelong, P; Martin, Y.
The authors studied survival of faecal coliform (decrease rate + T90) in experimental devices (closed or continuous flow systems) using various dilutions of urban sewage in sea-water. Survival appears to be high (T90 between 12 to 81 h). Dilution level of sewage and organic matter contents seems to be important to explain the decrease of faecal coliforms observed in these experiments.

The effect of anaerobic digestion in biogas plants on survival of pathogenic bacteria OAK
Umetsu, K; Kikuchi, S; Nishida, T; Kida, K; Ihara, I; Yamashiro, T;
The paper deals with the hygienic advantages of sanitation to treat dairy manure in full-scale biogas plants. Slurry samples were collected from two thermophilic biogas plants (55ºC) and two mesophilic biogas plants (38ºC) in Hokkaido, Japan. The detectable number of Coli-aerogenes group and Enterococcus in the slurries after anaerobic digestion could not be found in both thermophilic biogas plants. In both mesophilic biogas plants, however, the viable numbers of Coli-aerogenes group and Enterococcus were detected in the slurries even after anaerobic digestion. The mean decimation reduction time (T90) values of Coli-aerogenes group and Enterococcus in the slurries during mesophilic digestion were 13.3 d and 16.7 d, respectively.
