Effect of the medium composition on formation of amylase by Bacillus sp BABT
Santos,Eliana de Oliveira.; Martins,Meire Lelis Leal.
Studies on the alpha -amylase synthesis was carried out with a moderately thermophilic, facultatively anaerobic Bacillus sp, isolated from soil samples. The cells were cultivated in a complex medium containing soluble starch or maltose as carbon source. The levels of the alpha -amylaseactivity detected in culture supernatants varied greatly with the type of carbon source used. Maltose, soluble starch and citrate stimulated alpha -amylaseformation. Addition of exogenous glucose repressed formation of alpha -amylase, demonstrating that a classical glucose effect was operative in this organism. The concentration of yeast extract was found to be important factor in the alpha -amylase synthesis bythe isolate. The activity of the enzyme increased between 2 and 5...

Oocyte surface in four teleost fish species postspawning and fertilization BABT
Rizzo,Elizete; Moura,Thais F.C.; Sato,Yoshimi; Bazzoli,Nilo.
Cytological and cytochemical studies were carried out to investigate the surface characteristics of oocytes of four teleost species from the São Francisco river. The fishes were submitted to hypophysation at the Três Marias Hybrobiology and Fishculture Station, Minas Gerais, Brazil, in January 1996. Postspawning, oocytes of the curimatãs Prochilodus affinis, Prochilodus marggravii and dourado Salminus brasiliensis were surrounded by a thick, three-layered zona pellucida with radial striae. The surface of spawned oocytes of the surubim, Pseudoplatystoma coruscans, was comprised of mucous coat located externally to a thin, two-layered and striated zona pellucida. Oocyte activation during fertilization, lead to cortical reaction, formation of a perivitelline...

Composition, functional properties and sensory characteristics of Mozzarella cheese manufactured from different somatic cell counts in milk BABT
Andreatta,Evelise; Fernandes,Andrezza Maria; Santos,Marcos Veiga; Mussarelli,Camila; Marques,Marina Célia; Oliveira,Carlos Augusto Fernandes de.
In the present study, composition, functional properties and sensory characteristics of Mozzarella cheese produced from milk with somatic cell counts (SCC) at low (<200,000 cells/mL), intermediate (≈400,000 cells/mL) and high (>800,000 cells/mL) levels were investigated. Three batches of cheese were produced for each SCC category. The cheeses were vacuum packed in plastic bags and analysed after 2, 9, 16, 23 and 30 days of storage at 4ºC. SCC level did not affect the moisture, fat, total protein and ash content, mesophilic and psychrotrophic bacteria, and sensory parameters of Mozzarella cheese. However, meltability increased in cheese manufactured from high SCC milk. Results indicated that raw milk used to produce Mozzarella cheese should not...

The seed bank of a gallery forest in Southeastern Brazil BABT
Grombone-Guaratini,Maria Tereza; Leitão Filho,Hermógenes de Freitas; Kageyama,Paulo Yoshio.
The soil seed bank was studied in a gallery forest in Southeastern Brazil. Samples were collected from edge to edge along transects perpendicular to the river during the wet (December, 1990) and dry (August, 1991) seasons. The number of seeds found in the seed bank samples was greater in the dry season than in the wet season indicating that there was important variation in the seed stocks in these two periods. The similarity values between the seed bank and the community composition were low. The high density of weed species might be reflecting forest fragmentation and indicating a degree of perturbation.

Duration of the pre-settlement period of the mangrove crab Ucides cordatus (Decapoda: Ocypodidae) under laboratory conditions BABT
Ventura,Robson; Silva,Ubiratã de Assis Teixeira da; Perbiche-Neves,Gilmar; Ostrensky,Antonio; Boeger,Walter Antonio Pereira; Pie,Marcio Roberto.
The goal of the present study was to determine the most appropriate time to release the immatures of Ucides cordatus (Linnaeus) produced in the laboratory into the natural environments. Specifically, the time when the megalopae sought the mangrove sediment to excavate the burrows was determined, as well as the time necessary for their metamorphosis into the first juvenile stage. Results indicated that the megalopae of U. cordatus reared in the laboratory took three to ten days (median = 6) after their molt to excavated burrows in the sediment. The average time for the megalopae to molt into juveniles was 12.6 days (SD = 2.3).

Evaluation of red cabbage anthocyanins after partial purification on clay BABT
Lopes,Toni Jefferson; Yaginuma,Sandra Regina; Quadri,Mara Gabriela Novy; Quadri,Marintho Bastos.
The aim of this work was to evaluate the red cabbage anthocyanins quality after purification by static adsorption assays on clay (Tonsil Terrana 580FF). Thin-layer chromatography analysis and scan spectrometry on UV-Visible showed a good dye stability of the pigment interacting with the clay. The use of pH 3.0 buffer during the batch assays provided a protective effect on the cabbage anthocyanins, limiting the dye acid hydrolyses. Analyses of the reducing sugars contents of the extract showed that clay retained 20% average of total free sugars under the test conditions.

Brustolin,Jean Carlos; Dal Pisol,Andreia; Steffens,Juliana; Toniazzo,Geciane; Valduga,Eunice; Di Luccio,Marco; Cansian,Rogério Luis.
The objective of this work was to evaluate the effect of different water pressures and concentrations of lactic acid on microbial counts (mesophilic bacteria, enterobacteria and Salmonella) on pig carcasses without contamination and contaminated carcasses, before and after the last shower and before being cooled. The tests were carried out using 4, 3 and 2 bar water pressure, and 2, 1 and 0% lactic acid concentration. In general, both the pressure in the shower and lactic acid had a positive effect by reducing the microbial count. The interaction between the pressure and lactic acid caused the largest reduction in carcasses surface count for mesophiles. With regard to enterobacteria on contaminated carcasses, the most important variable was the lactic acid...

Souza,Carlos Eduardo Alves de; Stolf,Aline Maria; Dreifuss,Arturo Alejandro; Lívero,Francislaine dos Reis; Gomes,Liana de Oliveira; Petiz,Lyvia; Beltrame,Olair; Dittrich,Rosangela Locatelli; Telles,José Ederaldo Queiroz; Cadena,Sílvia Maria; Acco,Alexandra.
Alcoholic liver disease is characterized by a wide spectrum of liver damage, which increases when ethanol is associated with high-fat diets (HFD). This work aimed to establish a model of alcoholic hepatic steatosis (AHS) by using a combination of 10% ethanol and sunflower seeds as the source of HFD. Male rats received water or 10% ethanol and regular chow diet and/or HFD, which consisted of sunflower seeds. The food consumption, liquid intake and body weight of the rats were monitored for 30 days. After this period, blood was collected for biochemical evaluation, and liver samples were collected for histological, mitochondrial enzyme activity and oxidative stress analyses. Our results indicated that the combination of 10% ethanol and HFD induced micro- and...

Preliminary analysis of the genetic variability of two natural beds of the Scallop Euvola ziczac (Linnaeus, 1758) in Brazil BABT
Wangüemert,Mercedes Gonzalez; Pezzuto,Paulo Ricardo; Borzone,Carlos Alberto.
Euvola ziczac (formerly Pecten ziczac), a simultaneous hermaphroditic scallop was heavily fished in Brazil between 1972 and 1980. The production peaked in 1980 with 8,800 tons and was followed by the total collapse of the resource. In order to investigate the possible loss of genetic variability of the stock associated to overfishing and self-fertilization, the polymorphism of phosphoglucomutase (PGM) and glucose phosphate isomerase (GPI) was analyzed by electrophoresis of the adductor muscle of scallops from São Francisco (26° 20.583’S; 48° 16.507’W) and Bom Abrigo (25° 28.735’S; 47° 37.621’W) beds; the southern and northern extremes of the scallop fishing ground, respectively. Optimization of Lipase-Catalyzed Transesterification of Cotton Seed Oil for Biodiesel Production Using Response Surface Methodology BABT
Li,Ying Xia; Dong,Bing Xue.
The aim of this work was to study the biodiesel production from cotton seed oil by lipase produced by Pichia guilliermondii lipase, which was immobilized onto hydrophobic magnetic particles (HMPs). The optimum reaction conditions were determined for lipase dosage, methanol-to-oil molar ratio, temperature and water content. Using response surface methodology, a quadratic polynomial equation was obtained for fatty acid methyl esters (FAMEs) content by multiple regression analysis. Verification experiments confirmed the validity of the predicted model. The optimal conditions for the enzymatic transesterification were temperature of 38.76℃, 31.3% immobilized lipase, 10.4% water content, and a methanol-to-oil molar ratio of 4.715:1. The gas chromatography- mass...

Copper adsorption in tropical oxisols BABT
Silveira,Maria Lucia Azevedo; Alleoni,Luís Reynaldo Ferracciú.
Cu adsorption, at concentrations between 0 to 800 mg L-1, was evaluated in surface and subsurface samples of three Brazilian soils: a heavy clayey-textured Rhodic Hapludalf (RH), a heavy clayey-textured Anionic ''Rhodic'' Acrudox (RA) and a medium-textured Anionic ''Xanthic'' Acrudox (XA). After adsorption, two consecutive extractions were performed to the samples which received 100 mg L-1 copper. Surface samples adsorbed higher amounts of Cu than the subsurface, and exhibited lower Cu removed after the extractions, reinforcing the influence of the organic matter in the reactions. Cu adsorption was significant in the subsurface horizons of the Oxisols, despite the positive balance of charge, demonstrating the existence of mechanisms for specific...

Energy Potential of Animal Waste in the State of Paraná (Brazil) BABT
Mancini,Fabrízio Nicolai; Milano,Jackson; Araújo,Joilson Gonçalves de; Iastrenski,Karen Techy; Silveira,Nicolli Sperança; Pertile,Rodrigo Creuz.
Abstract Animal biomass is a source of energy commonly discarded without or with low utilization, but its availability draws attention to its reuse [1]. The utilization can be carried out in different ways, varying in terms of generation time and efficiency. The use of biodigestion requires a relatively small area compared to landfills or natural composting, and it provides other materials besides biogas, that can be recycled as fertilizer and has a short residence time of residues [2]. The energy supplied as biogas can be harnessed as heat or electricity in case of conversion. Considering the energy needs of animal husbandry and the ease of reinsertion of energy in the production cycle, the possibility of distributed generation of biogas was studied as a...

Characterization of the biomass of a hybrid anaerobic reactor (HAR) with two types of support material during the treatment of the coffee wastewater BABT
Silva,Vivian Galdino da; Campos,Cláudio Milton Montenegro; Pereira,Erlon Lopes; Silva,Júlia Ferreira da.
This study investigated the microbiology of a hybrid anaerobic reactor (HAR) in the removal of pollutant loads. This reactor had the same physical structure of an UASB reactor, however with minifilters inside containing two types of support material: expanded clay and gravel. Two hydraulic retention times (HRT) of 24h and 18h were evaluated at steady-state conditions, resulting in organic loading rates (OLR) of 0.032 and 0.018 kgDBO5m-3d-1 and biological organic loading rates (BOLR) of 0,0015 and 0.001 kgDBO5kgSVT- 1d¹, respectively. The decrease in concentration of organic matter in the influent resulted an endogenous state of the biomass in the reactor. The expanded clay was the best support material for biofilm attachment.

Influence of Prolonged Salting on the Physicochemical Properties of Duck Egg White BABT
Venkatachalam,Karthikeyan.
ABSTRACT Salted duck eggs are unique among the important authentic foods of Thailand. They are mainly produced in Chaiya district, Surat Thani province, in the southern part of Thailand. The egg whites of salted duck eggs are normally unusable due to heavily salty taste upon prolonged storage. The present study was aimed to examine the foaming characteristics of egg white after being cured for a prolonged period (30 days). At 5-day intervals the egg whites were measured for salt, moisture, water activity, hydrophobicity, pH, zeta potential, particle size, SH groups, surface tension and foaming properties (index of whipping, foam durability, specific density, overrun, air phase and yield stress). The results showed that prolonged curing significantly...
