Isolation and purification of bromelain from waste peel of pineapple for therapeutic application BABT

The aim of this work was to isolate and purify bromelain extracted from the pineapple peel by ammonium sulfate precipitation (40-80%), followed by desalting and freeze-drying with a 75% activity recovery and 2.2 fold increased specific activity. Ion exchange chromatography on DEAE-Sepharose was able to separate the polysaccharides from the enzyme, which was recovered in the elution step, maintaining its enzymatic activity. The batch adsorption of bromelain was evaluated in terms of total protein and enzymatic activity using Langmuir and Langmuir-Freundlich models. Results showed that the process could be suitable for the recovery and purification of the enzyme, maintaining its specific activity.

Testicular morphology of adult wistar rats treated with Rudgea viburnoides (Cham.) Benth. leaf infusion BABT

The present study was carried out to investigate the effects of Rudgea viburnoides infusion on the body biometry and testicular morphometry and stereology of adult Wistar rats. Two groups received the infusion daily at the concentration of 3 or 6 mg/mL for 40 days. The control group received only water. Neither the biometrical parameters nor the tubular diameter and the height of the seminiferous epithelium showed any significant alterations in the treated animals. Leydig cells stereology did not show any significant alterations in the treated animals. These results indicated that R. viburnoides did not cause alterations in body biometry and testicular morphometry.

Evaluation of microorganisms with sulfidogenic metabolic potential under anaerobic conditions BABT

The aim of this work was to identify groups of microorganisms that are capable of degrading organic matter utilizing sulfate as an electron acceptor. The assay applied for this purpose consisted of running batch reactors and monitoring lactate consumption, sulfate reduction and sulfide production. A portion of the lactate added to the batch reactors was consumed, and the remainder was converted into acetic, propionic and butyric acid after 111 hours of operation These results indicate the presence of sulfate-reducing bacteria (SRB) catalyzing both complete and incomplete oxidation of organic substrates. A novel class of Tc-99m complexes able to cross the blood brain barrier has been investigated and described here. These compounds are formed by reacting the bis-substituted nitrido precusors [99mTc(N)(PS)2] (PS = phosphino-thiol ligand) with triethylborane (BEt3) under strictly anhydrous conditions and using non-aqueous solvents. The molecular structure of these tracers was not fully established, but preliminary experimental evidence suggests that they result from the interaction of the Lewis base [99mTc(N)(PS)2] with the Lewis acid BEt3, which leads to the formation of the novel -B-N<FONT FACE=Symbol>º</FONT> Tc- adduct. Seed germination in Miconia theaezans (Bonpl.) Cogniaux (Melastomataceae) BABT

The effects of light and temperature were studied on the seeds of Miconia theazeans by isothermic and alternating temperature incubations. The optimum temperature for seed germination was determined by final percentage and germination rates as located in the range of 27.5 to 30 °C and by germination kinetics at the range of 19.5 to 30 °C. The germination was dependent on diffusion processes. The minimum and maximum temperatures were 12.5-15°C and 32.5-35°C, respectively. Our study evaluated how the consumption of diets with low (LOW group - 0.4/1) or high (CON group - 13.6/1) omega-6/omega-3 ratio across generations (F1 and F2) can modulate liver fatty acid (FA) profile and blood biomarkers. Liver content of α-linolenic acid was higher in animals always fed with LOW diet than animals that changed from CON to LOW diet, which by your time was higher than animals always fed with CON diet. Liver saturated FA concentration decreased in both groups from F1 to F2. Effect of ivermectin on the life cycle and larval fat body of Culex quinquefasciatus BABT

The present work investigated the behavior, survival rate and lethal effect of 1.5 ppb ivermectin on Culex quinquefasciatus larvae, based on morphological and biological parameters, and possible alterations of their fat body. Changes in the number of eggs/egg mass and length of the larval stage were investigated. For this experiment, 600 larvae of 3rd and 4th instars of the mosquito were tested. The laid eggs were separated and the hatched larvae were counted. Some larvae submitted to 1.5 ppb ivermectin solution were used to prepare samples for histological study. This work investigated the genetic control of seed morphological traits and its correlations with grain yield in common bean. Three crossings among bean cultivars with different growth habit and seed characteristics were analyzed. F1 progenies gave origin to F2, RC1P1F1 and RC1P2F1 generations. Random samples of seeds from F2 generations and parents, F1 and backcrossings were sown during the season 2003/2004. Aflatoxin M1 contamination in grated parmesan cheese marketed in Rio de Janeiro - Brazil BABT

This study aimed to determine the occurrence of AFM1 contamination in the samples of grated parmesan cheese marketed in the Metropolitan Region of Rio de Janeiro -Brazil. Thirty samples representing 10 major brands marketed in the region were analyzed by High Performance Liquid Chromatography with fluorescence detection (HPLC-FLD) after purification with immunoaffinity column. The method showed recovery values within the range of 70-90%, with RSD lower than 15% and limits of detection and quantification below the maximum level allowed by the European Commission for the presence of AFM1 in cheeses. Broiler transportation conditions in a Brazilian commercial line and the occurrence of breast PSE (Pale, Soft, Exudative) meat and DFD-like (Dark, Firm, Dry) meat BABT

The objective of this work was to investigate the effect of road transportation conditions on the occurrence of broiler PSE-(Pale, Soft, Exudative) and DFD-like (Dark, Firm, Dry) meats in the Brazilian commercial slaughterhouse plant. Samples of Pectoralis major m from 47-day-old broilers of commercial lineage were analyzed. The results indicated that water-bathing birds just before journey over 3.0km promoted the occurrence of 46.0% of PSE and 4.0% of DFD-like meat, while birds under non-water bathing conditions presented 14.7 and 2.0%, respectively. Effect of alcoholic fermentation in the content of phenolic compounds in cider processing BABT

The objective of this work was to study the effect of alcoholic fermentation on the content of phenol compounds of five cider apple varieties. The initial content in the apple juice samples, as determined by HPLC, varied from 188.4 to 2776.17 m mg.L-1. In three of them (DC, PJ, GU), the total phenol compounds remained unaffected by fermentation. However, in two (DM, KE), the final values were lower (55 and 313 mg.L-1). In these apple cider, the values of caffeic acid increased from 6.6 mg.L-1 to 41.8 mg.L-1. The catechin content increased during the process, approximately 13 mg.L-1 independent of the variety. Diet and dietary overlap of three sympatric fish species in lakes of the upper Paraná River floodplain BABT

For a dietary study, specimens of Astyanax bimaculatus, Astyanax schubarti, and Moenkhausia intermedia were caught monthly with seines-nets in six lakes (Pau Veio, Porto Rico, Três Amigos, Mutum, Pontal, and Canal do Meio), on the floodplain of the Upper Paraná River, from March 1992 through February 1993. We analyzed stomach contents of 599 A. bimaculatus, 293 A. schubarti, and 394 M. intermedia, by the Occurrence and Points methods, combined in the Alimentary Index (AI). This analysis revealed that these species have broad dietary spectra. A. bimaculatus was an omnivore, with a tendency toward herbivory-insectivory. Thirty male Wistar rats (initial weight of 270 g ± 20 g) were used in the study, which were divided into six groups five each. Milk line as an indicator of the harvesting time of three hybrid seeds of corn (Zea mays L.) BABT

The experiment was carried out to evaluate the time for harvesting seeds of three hybrids of corn (Zea mays L.) at their best physiological quality and the use of the milk line as an indicator of the physiological seed maturity. Single hybrids CD 1723 and CD 5501 and double hybrid OC 705 seeds were collected for 59 days, every four days, starting 23 days after female flowering. Temperature Dependent Seed Germination of Dalbergia nigra Allem (Leguminosae) BABT

The germination of endangered species Dalbergia nigra was studied and 30.5° C was found as optimum temperature, although the species presented a broad temperature range where germination occurs and light had no effect. The analysis of kinetics of seed germination confirmed the asynchronized germination below and above the optimum temperature. The light insensitive seed and germination also at high temperatures indicated that D. nigra could occur both in understories and gaps where the mean temperature was high.
