Abstract: The objective of this work was to evaluate the response of an in vitro host range to Spodoptera frugiperda multiple nucleopolyhedrovirus (SfMNPV), a pathogenic virus to the fall armyworm (Spodoptera frugiperda, Lepidoptera: Noctuidae), for the further development of a biopesticide based on cell culture systems. The cell lines from Bombyx mori (BM-5), Lymantria dispar (IPLB-LD-625Y), Trichoplusia ni (BTI-Tn-5B1-4), Anticarsia gemmatalis (UFL-AG-286), and S. frugiperda (IPLB-SF-21AE and Sf9) were tested for their susceptibility to a highly-virulent Brazilian isolate of SfMNPV. ABSTRACT The Brazilian Poplar Moth, Condylorrhiza vestigialis (Guenée), compromises the wood productivity of poplar trees (Populus sp.), mainly affecting the matchstick industry in southern Brazil. Considering the lack of information on rearing techniques for this insect, the objective of this study was to develop an artificial diet to rear C. vestigialis with biological characteristics similar to the wild insects. Boric acid concentrations (0.02,0.03,0.045,0.067 and 0.101 g/100 ml of diet) were evaluated in combination with the Anticarsia gemmatalis Hüb. nuclear polyhedrosis virus (AgNPV) for enhanced virali activity against the insect. Seven days after inoculation, the median lethal concentration (LC50) was 1.52 x 10(5) for the AgNPV alone and 7.95 x 10² for the NPV mixed with 0.045g of boric acid/100 ml of diet. At subsequent evaluation dates (9,11 and 14 days after inoculation) LC50's for NPV+boric acid were ca. 4x lower than those observed for the NPV alone. The laboratory production of Anticarsia gemmatalis Hübner nuclear polyhedrosis virus (AgNPV) was evaluated for larvae inoculated at 1.0, 1.5, 2.0, and 2.5 cm and with two NPV dosages, 10(6) and 10(7) polyhedron inclusion bodies (PIB)/ml, sprayed on the insect diet surface. A fixed population of 25 larvae/300-ml cup was used. Highest virus yield was achieved for larvae inoculated with ca. 2.0 cm at the higher NPV dosage. Losses of NPV-inoculated larvae ranged from 42.4% to 64%, due to high incidence of canibalism, death by other causes, and pupation. Abstract Chrysodeixis includens has become the major Lepidopteran pest of soybean crops, especially in the Brazilian Cerrado (savanna) region. A native isolate of Chrysodeixis includens nucleopolyhedrovirus (ChinNPV) from this region, Buritis, MG, was assessed for its biological and molecular features. In addition, in vitro co-infection with Anticarsia gemmatalis multiple nucleopolyhedrovirus (AgMNPV), another virus of an important soybean pest, was tested. In the present study we evaluated different systems for the expression of mycobacterial antigen P36 secreted by Mycobacterium bovis. P36 was detected by Western blot using a specific antiserum. The P36 gene was initially expressed in E. coli, under the control of the T7 promoter, but severe proteolysis prevented its purification. We then tried to express P36 in M. smegmatis and insect cells. For M. smegmatis, we used three different plasmid vectors differing in copy number and in the presence of a promoter for expression of heterologous proteins.
