Ocorrência de Beauveria bassiana (Bals.) Vuill. em broca-do-café (Hypothenemus hampei, ferrari) no estado de Rondônia, Brasil1 Acta Amazonica

Em trabalho efetuado visando o levantamento de inimigos naturais da broca-do-café (Hypothenemus hampei, Ferrari) no estado de Rondônia, verificou-se a ocorrência do fungo Beauveria bassiana (Bals.) Vuill. conforme identificação da Embrapa Rondônia. Os frutos de café foram coletados em cafezais da cultivar Conilon (Coffea canephora), nos municípios de Rolim de Moura, Ouro Preto do Oeste e Machadinho do Oeste, Rondônia, nos meses de fevereiro a março de 2000. A percentagem média de infecção de B. bassiana em frutos broqueados variou de 0,23% a 0,47%, sendo menor que em outras regiões do Brasil.

La mosca blanca de los invernaderos, Trialeurodes vaporariorum, es una de las principales plagas de cultivos de tomate bajo invernadero y es controlada principalmente con insecticidas químicos. Sin embargo, existen estrategias alternativas de control como hongos entomopatógenos (Beauveria bassiana) y enemigos naturales como parasitoides (Encarsia formosa). En el presente estudio se evaluó la compatibilidad de productos comerciales con adultos y pupas de E. formosa bajo condiciones de laboratorio. Vuill. se realiza en cultivo en medio sólido (CMS) utilizando productos agrícolas y sus residuos, como arroz (Oryza sativa) y bagazo de caña (Saccharum ojjicinarum), donde el CO2 puede acumularse como producto del metabolismo. Grooming capacity inhibition in Cornitermes cumulans (Kollar) (Isoptera: Termitidae) inoculated with entomopathogenic fungi and treated with imidacloprid Anais da SEB

This study was carried out to attest inhibition in C. cumulans grooming behavior when treated with entomopathogenic fungi and sub-doses of imidacloprid insecticide. Observations were made under the scanning electronic microscope. The termite workers showed an efficient grooming behavior which allowed the removal of all conidia from the cuticle of soldiers and other workers, in the first hours after fungal application. Foram selecionados 20 equídeos naturalmente infestados por Anocentor nitens, dos quais seus pavilhões auriculares foram submetidos a quatro tratamentos distintos: gel associado a Beauveria bassiana, apenas gel, apenas B. bassiana e um grupo-controle. Procederam-se à contagem e ao registro do número total de fêmeas adultas (>3mm) em cada pavilhão auricular, nos dias 0, 1, 4, 7, 11, 14, 18, 21, 25 e 28, para cálculo do percentual de controle. Foram coletadas e mantidas em laboratório 20 fêmeas pertencentes a cada tratamento, para cálculo dos períodos de pré-postura, postura, incubação e eclosão, e peso médio das posturas. O trabalho teve por objetivo verificar a mortalidade in vitro de ninfas e adultos de Amblyomma cajennense frente à ação de isolados dos fungos Beauveria bassiana e Metarhizium anisopliae. Foram avaliados três isolados de M. anisopliae (959, 319 e E9) e dois de B. bassiana (986 e 747). As suspensões de conídios foram preparadas a partir de fungos produzidos em meio de arroz, e cada bioensaio foi constituído de quatro tratamentos nas concentrações 10(5), 10(6), 10(7), 10(8) conídios/ml e um grupo-controle. The natural insect host range of the entomopathogenic fungi (EPF) Beauveria bassiana (Bb) and Metarhizium anisopliae (Ma) was investigated in Argentina during the winter of 2003 through spring of 2004. Fungi- infected insect samples (153) were collected from cornfields and the surrounding uncultivated areas in different localities of Buenos Aires (7), Tucumán (2), and Corrientes (3) provinces. The rates of Bb-infected host range varied among the Coleoptera (37%), Hemiptera (27%) and Dermaptera (1.3%). The biomass yield obtained in the first phase based on submerged cultivation of fungus was processed using methodology providing different contact with air space. Effect of moisture content and inoculum on the growth and conidia production by Beauveria bassiana on wheat bran BABT

The aim of the present work was to study the effect of moisture content and inoculum on the growth and conidia production by Beauveria bassiana on wheat bran (WB).The highest growth rate of B. bassiana 885.2 on WB media was obtained at a w=1.0, with no detected growth at a w < 0.97. Solid-state fermentation (SSF) using WB (66% moisture; a w=1.0) achieved a maximal yield of 1.18x10 10 conidia per gram of dry substrate (gds). This yield decreased one order of magnitude with higher moisture contents or the addition of sugarcane bagasse (SCB) as a texturiser. Immobilization of Beauveria bassiana Lipase on Silica Gel by Physical Adsorption BABT

Extracellular lipase from Beauveria bassianastrain CG481 was immobilized by using thirteen different immobilization protocols. Silica gel was chosen as the most suitable adsorbent with 94.8% of activity yield. The adsorption on silica gel did not change the optimum pH (8.5) and temperature (45ºC) values of the free lipase (FL) for lipolytic activity, and it showed higher activities in extreme conditions (pH 9.0 to 10.5, 60ºC). The lipase immobilized on silica gel (ILS) showed enhanced stability at pH 7.0 after 120 h incubation (69.0%) when compared to FL (33.3%). Production of extracellular protease by a Brazilian strain of Beauveria bassiana reactivated on coffee berry borer, Hypothenemus hampei BABT

Studies were carried out on extracellular protease production by Beauveria bassiana CG432 in liquid medium containing glucose and yeast extract. B. Bassiana presented active growth after lag period of 24 h., produced 80% of the total of the extracellular protease activity in 48 h which was maximum on the 5th culture day. The extracellular protease presented optimum activity at 60ºC, was stable up to 1M Cl-, maintained the stability during 15 day at 4ºC and -18ºC, but was not stable if frozen repeatedly.

Spore production of Beauveria bassiana from agro-industrial residues BABT

The purpose of this work was to produce Beauveria bassiana by Solid-State Fermentation using agro-industrial residues and optimizing the cultivation conditions. Refused potatoes, coffee husks and sugar-cane bagasse were tested. The blend of refused potatoes and sugar-cane bagasse (60-40%) with particle size in the range of 0.8-2 mm was used in the fermentation experiments. In Erlenmeyer flasks the best spore production was achieved with the following conditions: incubation temperature 26º C; initial pH 6.0; inoculum concentration 10(7) spores.g-1.dw and initial moisture 75%. Microorganisms as tools in modern chemistry: the biotransformation of 3-indolylacetonitrile and tryptamine by fungi BJM

Chemical reactions performed by microorganisms have been used as a modern tool in chemistry. In this work, the ability of the fungi Beauveria bassiana and Aspergillus niger to modify the chemical structure of indole compounds was studied. Beauveria bassiana was able to transform 3-indolylacetonitrile into 3-methylindole while Aspergillus niger transformed tryptamine into 5-hydroxyindole-3-acetamide. Interestingly, these fungi were able to perform both reduction and oxidation of the indole compounds fed, the oxidation occurring with improved levels of oxygen uptake. The results prove once more the versatility of microorganisms to perform a wide range of industrially attractive chemical reactions.

Effect of surfactants and temperature on germination and vegetative growth of Beauveria bassiana BJM

Three non-ionic surfactants: Tween20, Tween80 and Breakthru® were screened for their effects on spore germination and mycelial growth rates and for their influence on three isolates of Beauveria bassianaspore germination at various temperatures. Tween20 and Tween80 were compatible with all the B. bassiana isolates in the germination studies, but inhibited germination at higher surfactant concentrations, irrespective of the conidial concentrations. This work aimed at investigatimg whether the culture medium used in viability tests affects the conidial germination of Lecanicillium lecanii, Beauveria bassiana and Paecilomyces fumosoroseus isolates. The tests were performed on microscope slides containing one of the culture media: agar-water (AW), minimal medium (MM), potato-dextrose agar (PDA), potato-dextrose-1% yeast extract agar (PDAY), Sabouraud-dextrose-yeast extract agar (SDAY), and complete medium (CM). O objetivo deste trabalho foi avaliar a eficiência de Beauveria bassiana (Bb), da terra diatomácea (TD) e dos pós de rocha Protesyl e Rocksil, isoladamente ou em associações, para o controle de Alphitobius diaperinus. Os agentes de controle foram aplicados na forma sólida misturados à cama de frango. A concentração de conídios de Bb foi de 0,08% e, para os pós inertes, as concentrações foram de 0,5; 1,0 e 1,5%. Testaram-se 20 tratamentos, com cinco repetições de 200 insetos, por 10 dias. Os tratamentos que apresentaram maior eficiência foram TD 1,5% e Bb 0,08% + TD 1,5%, com 60,2 e 57,8% de mortalidade, respectivamente.
