Biodiversity of rhizobia associated with cowpea cultivars in soils of the lower half of the São Francisco River Valley Rev. Bras. Ciênc. Solo
Leite,Jakson; Seido,Sirando Lima; Passos,Samuel Ribeiro; Xavier,Gustavo Ribeiro; Rumjanek,Norma Gouvêa; Martins,Lindete Míria Vieira.
The biodiversity of rhizobium in soils of the São Francisco Valley is unknown and can be studied using cowpea as trap plants. The objective of this study was to verify the diversity of diazotrophic bacteria that nodulate cowpea in soils of the lower half of the São Francisco River Valley by morphological and genotypic characterization. Seven soil samples (A1, A2, A3, A4, C1, C2 and MC) were collected to capture bacteria associated to five cowpea cultivars (IPA 206, BRS Pujante, BRS Marataoã, Canapu Roxo, and Sempre Verde), in a 5x7 factorial design with three replications. Thirty days after plant emergence, the nodules were collected and the bacteria isolated and analyzed in relation to their growth characteristics in YMA medium. The 581 isolates were...

Soybean response to starter nitrogen and Bradyrhizobium inoculation on a Cerrado oxisol under no-tillage and conventional tillage systems Rev. Bras. Ciênc. Solo
Mendes,I. C.; Hungria,M.; Vargas,M. A. T..
In Brazil, Bradyrhizobium inoculation has successfully replaced the use of N fertilizer on soybean [Glycine max (L) Merr.] crops. However, with the expansion of no-tillage cropping systems in the Cerrados region, the idea that it is necessary to use small N rates at the sowing to overcome problems related with N immobilization has become widespread, mainly when soybean is cultivated after a non-legume crop. In this study we examined soybean response to small rates of N fertilizer under no-tillage (NT) and conventional tillage (CT) systems. Four experiments (a completely randomized block with five replicates) were carried out in a red yellow oxisol, during the periods of 1998/1999 and 1999/ 2000, under NT and CT. The treatments consisted of four urea rates...

Genetic Variability and Symbiotic Efficiency of Erythrina velutina Willd. Root Nodule Bacteria from the Semi-Arid Region in Northeastern Brazil Rev. Bras. Ciênc. Solo
Menezes,Kelly Alexsandra Souza; Escobar,Indra Elena Costa; Fraiz,Ana Carla Resende; Martins,Lindete Míria Vieira; Fernandes Júnior,Paulo Ivan.
ABSTRACT Legume-rhizobia symbiosis is a cross-kingdom association that results in large amounts of nitrogen incorporated in food webs. For the Brazilian semi-arid region, data on genetic variability and symbiotic efficiency of Papilionoidae rhizobial communities are very scarce. The aim of this study was to evaluate the genetic variability and the symbiotic efficiency of eight rhizobial isolates obtained from “mulungu” (Erythrina velutina Willd.) nodules. For 16S rRNA gene sequencing, the genomic DNA was extracted using a commercial kit, amplified with universal primers, and subjected to sequencing reactions. For the isolate ESA 71, PCR amplifications for nodC and nodA genes were attempted. Rhizobial efficiency was assessed by two greenhouse experiments....

Miyauchi,Marina Yumi Horta; Lima,Dáfila Santos; Nogueira,Marco Antonio; Lovato,Gisele Milani; Murate,Letícia Sayuri; Cruz,Márcio Ferreira; Ferreira,Josué Maldonado; Zangaro,Waldemar; Andrade,Galdino.
Some diazotrophic bacteria can fix nitrogen biologically in gramineous host plants. Generally, gramineous plants are also associated with mycorrhizal fungi, that can improve mainly plant P uptake. Among the factors affecting plant-microbe interactions, the plant genotype plays an important role. This study evaluates the effect of diazotrophic bacteria and an arbuscular mycorrhizal fungus (AMF), on five genotypes of maize (Zea mays L.), in relation to plant biomass, shoot N and P concentrations, and fine root morphological traits. The experimental design was entirely randomized in a factorial 5 × 4 × 2 arrangement, i.e., five maize genotypes (hybrids C333B, AS3466, and PREMIUM, and the inbreed lines lg40897-1 and lg40505-1), three diazotrophic bacteria...

Bacterial strains from floodplain soils perform different plant-growth promoting processes and enhance cowpea growth Scientia Agricola
Costa,Elaine Martins da; Carvalho,Fernanda de; Nóbrega,Rafaela Simão Abrahão; Silva,Jacqueline Savana; Moreira,Fatima Maria de Souza.
ABSTRACT Certain nodulating nitrogen-fixing bacteria in legumes and other nodule endophytes perform different plant-growth promoting processes. The objective of this study was to evaluate 26 bacterial strains isolated from cowpea nodules grown in floodplain soils in the Brazilian savannas, regarding performance of plant-growth promoting processes and ability to enhance cowpea growth. We also identified these strains by 16S rRNA sequencing. The following processes were evaluated: free-living biological nitrogen fixation (BNF), solubilization of calcium, aluminum and iron phosphates and production of indole-3-acetic acid (IAA). The abilities to nodulate and promote cowpea growth were evaluated in Leonard jars. Partial sequencing of the 16S rRNA gene...

Plant densities and modulation of symbiotic nitrogen fixation in soybean Scientia Agricola
Luca,Marcos Javier de; Hungría,Mariangela.
Soybean nitrogen (N) demands can be supplied to a large extent via biological nitrogen fixation, but the mechanisms of source/sink regulating photosynthesis/nitrogen fixation in high yielding cultivars and current crop management arrangements need to be investigated. We investigated the modulation of symbiotic nitrogen fixation in soybean [Glycine max (L.) Merrill] at different plant densities. A field trial was performed in southern Brazil with six treatments, including non-inoculated controls without and with N-fertilizer, both at a density of 320,000 plants ha−1, and plants inoculated with Bradyrhizobium elkanii at four densities, ranging from 40,000 to 320,000 plants ha−1. Differences in nodulation, biomass production, N accumulation and partition were...

Alfalfa yield and quality as function of nitrogen fertilization and symbiosis with Sinorhizobium meliloti Scientia Agricola
Oliveira,Wladecir Salles de; Oliveira,Patrícia Perondi Anchão; Corsi,Moacyr; Duarte,Fábio Rodrigo Sanches; Tsai,Siu Mui.
The efficiency of the symbiotic process between alfalfa (Medicago sativa L.) and Sinorhizobium meliloti can be drastically affected by soil nitrogen. This research evaluates how mineral fertilization affects the contribution of symbiosis to the yield and quality of alfalfa in two experiments under controlled conditions, and a third one in the field. Under controlled conditions, positive effect of nitrogen fertilizer was observed, whereas, in the field, negative effect of the mineral nitrogen (450 kg ha-1 year-1) on the symbiotic process was observed, with reduction of nodulation and nitrogenase activity. However, there was no effect on yield, total nitrogen concentration, crude protein, non-protein nitrogen and digestibility in relation to the treatment...

Growth rate and nutritional status of an organic coffee cropping system Scientia Agricola
Ricci,Marta dos Santos Freire; Alves,Bruno José Rodrigues; Miranda,Simone Cordeiro de; Oliveira,Fabio Freire de.
In view of the low N concentration in organic fertilizers, it is necessary to use high rates of such fertilizers to attend coffee crop requirements. Hence, N is the most limiting nutrient for organic coffee production. The objective of this work was to evaluate the influence of sunn hemp (Crotalaria juncea) organic fertilization on the growth and nutritional status of coffee cultivars, as well as to quantify plant biomass and N input derived from biological nitrogen fixation, and their effect on soil chemical characteristics. The experiment consisted of six coffee (Coffea arabica) cultivars intercropped with and without sunn hemp sown in November 2001 and pruned at mid-height 76 days later. At 175 days, the standing biomass of the legume was cut, measuring...

Occurrence of diazotrophic bacteria in Araucaria angustifolia Scientia Agricola
Neroni,Rafaela de Fátima; Cardoso,Elke Jurandy Bran Nogueira.
Araucaria angustifolia is an environmentally threatened tree and the whole biota of the Araucaria Forest should be investigated with the aim of its preservation. Diazotrophic bacteria are extremely important for the maintenance of ecosystems, but they have never been studied in Araucaria Forests. In this study, diazotrophic bacteria were isolated from Araucaria roots and soil, when grown in semi-specific, semi-solid media. The diazotrophic character of some recovered isolates could be confirmed using the acetylene reduction assay. According to their 16S rRNA sequences, most of these isolates belong to the genus Burkholderia.
