Controlled release alginate-chitosan microspheres of tolmetin sodium prepared by internal gelation technique and characterized by response surface modeling BJPS
Elsayed,Mahmoud M. A..
Tolmetin sodium (TS) is a powerful non-steroidal mitigating drug for the treatment of rheumatoid joint inflammation, osteoarthritis, and adolescent rheumatoid joint pain. In addition to its gastrointestinal (GIT) problems, TS has a short biological half-life (1 hr). In a trial to overcome these side effects and control the rate of (TS) release, chitosan coated alginate microspheres are recommended. A Box-Behnken experimental design was employed to produce controlled release microspheres of TS in the sodium alginate and chitosan copolymers (Alg-Ch) by emulsification internal gelation methodology. The effect of critical formulation variables namely, drug to polymer ratio (D:P ratio), speed of rotation and span 80% on drug encapsulation efficiency (% EE),...

Targeting of temozolomide using magnetic nanobeads: an in vitro study BJPS
Gürten,Berna; Yenigül,Elçin; Sezer,Ali Demir; Altan,Cem; Malta,Seyda.
Temozolomide, a chemotherapeutic drug that is often administered for the treatment of brain cancer has severe side effects and a poor aqueous solubility. In order to decrease the detrimental effect of the drug over healthy cells, a novel drug delivery vehicle was developed where the therapeutic drug was encapsulated within the hydrophobic cavities of b-CD modified magnetite nanoparticles, which are embedded in chitosan nanobeads prepared by salt addition. In-vitro studies have shown that the magnetic properties of the novel delivery vehicle are adequate for targeted drug delivery applications under an external magnetic field. Additionally, an increase in the amount of chitosan was shown to exhibit a strong shielding effect over the magnetic properties of...

Gatti,Tiago Henrique Honorato; Eloy,Josimar Oliveira; Ferreira,Leonardo Miziara Barboza; Silva,Isabel Cristine da; Pavan,Fernando Rogério; Gremião,Maria Palmira Daflon; Chorilli,Marlus.
Abstract Mucoadhesive nanoparticles are particularly interesting for delivery through nasal or pulmonary routes, as an approach to overcome the mucociliary clearance. Moreover, these nanoparticles are attractive for peptide and protein delivery, particularly for insulin to treat diabetes, as an alternative to conventional parenteral administration. Thus, chitosan, a cationic mucoadhesive polysaccharide found in shells of crustaceans, and the negatively-charged dextran sulfate are able to form nanoparticles through ionic condensation, representing a potential insulin carrier. Herein, chitosan/dextran sulfate nanoparticles at various ratios were prepared for insulin loading. Formulations were characterized for particle size, zeta potential, encapsulation...

Scale-up in the synthesis of nanoparticles for encapsulation of agroindustrial active principles Ciência e Agrotecnologia
Duarte,Dirliane Santos; Nascimento,José Augusto de Almeida; Britto,Douglas de.
ABSTRACT Given the potentiality of nanoparticles (NP) to load substances as active principle of drugs, cosmetics and food, there is interest in increasing its production. Particularly in the agroindustrial area, the byproducts are source of functional compounds that must be stabilized by encapsulation, for example, to allow its application. Considering the amount of generated byproducts, it is necessary to understand the encapsulation synthesis in a high scale dimension. In this way, the active compounds vitamin C (from the byproduct of the acerola processing) and polyphenols (from the byproduct of grape processing) were nanoencapsulated into chitosan NP at three reactional volumes: 0.1; 1.0 and 10.0 dm3. The NP were characterized for yield, particle size,...

Partial substitution of nitrite by chitosan and the effect on the quality properties of pork sausages Ciênc. Tecnol. Aliment.
García,Mario; Beldarraín,Tatiana; Fornaris,Leonid; Díaz,Raúl.
The aim of the study was to evaluate the influence of partial nitrite replacement by chitosan on the quality of Ham Visking (a type of pork sausages). Five Ham Visking formulations were elaborated modifying the sodium nitrite (0.011; 0.016 or 0.0212%) and chitosan concentrations (0.25 or 0.5%) in the products. Sausages were stored at 4 ºC and physicochemical, microbiological, and sensorial evaluations were performed in order to estimate their shelf life. Chitosan can be used in pork sausages without affecting ensory attributes such as color although the panelists detected textural differences among the samples with chitosan, which suggests that there is some influence of deacetylation degree of chitosan on the textural behavior of sausages which still...

Proteomic analysis of the inhibitory effect of chitosan on Penicillium expansum Ciênc. Tecnol. Aliment.
LI,Mingyan; CHEN,Chi; XIA,Xiaoshuang; GARBA,Betchem; SHANG,Linlin; WANG,Yun.
Abstract The antimicrobial effect of chitosan on Penicillium expansum, a major postharvest pathogen of pome fruit, and the possible mechanisms involved in its effect were examined in this study. Chitosan strongly inhibited spore germination and hyphal growth of P. expansum. Light microscopy and transmission electron microscopy observations revealed that chitosan also caused morphological changes in hyphae and conidia, such as abnormal branching and vacuolation. Proteomic changes in P. expansum after chitosan treatment were analyzed by two-dimensional electrophoresis (2-DE) and mass spectrometry (MS) analysis, and 26 proteins were ambiguously identified and categorized based on their putative biological function. Proteins related to DNA or protein...
