The "CRABE" method is a crack detection technique based on the use of low electromagnetic frequencies. This (patented) method relies on a global injection of current within the structure under inspection. The presence of a crack shall cause a disruption in the current flow, and consequently of the magnetic field off the surface in the vicinity of a defect. It is this magnetic signature which is detected by the magnetic sensor used with "CRABE". The major advantage of this process, developed by TRAVOCEAN and the LETI Division of CEA with the help of IFREMER, appears clearly: No preliminary cleaning of the welds to be inspected is required, since no contact is necessary. Similarly, it shall be possible to inspect the whole of structures to a total cost all... |

In a recent report by risk assessment experts on the identification of food safety priorities using the Delphi technique, foodborne viruses were recognized among the top rated food safety priorities and have become a greater concern to the food industry over the past few years. Food safety experts agreed that control measures for viruses throughout the food chain are required. However, much still needs to be understood with regard to the effectiveness of these controls and how to properly validate their performance, whether it is personal hygiene of food handlers or the effects of processing of at risk foods or the interpretation and action required on positive virus test result. This manuscript provides a description of foodborne viruses and their... |

Here, we present a surface-enhanced Raman spectroscopy (SERS) nanosensor for environmental pollutants detection. This study was conducted on three polycyclic aromatic hydrocarbons (PAHs): benzo[a]pyrene (BaP), fluoranthene (FL), and naphthalene (NAP). SERS substrates were chemically functionalized using 4-dodecyl benzenediazonium-tetrafluoroborate and SERS analyses were conducted to detect the pollutants alone and in mixtures. Compounds were first measured in water-methanol (9:1 volume ratio) samples. Investigation on solutions containing concentrations ranging from 10−6 g L−1 to 10−3 g L−1 provided data to plot calibration curves and to determine the performance of the sensor. The calculated limit of detection (LOD) was 0.026 mg L−1 (10−7 mol L−1) for... |

Bonamia ostreae is an intracellular protozoan which is recognized as a cause of mortality in European populations of flat oysters (Ostrea edulis). Based on the recent characterization of actin genes of B. ostreae, specific primers were designed for real-time PCR using SYBR® Green chemistry. Specificity was demonstrated by the unique melting temperature peak observed in positive samples and by the lack of amplification in samples of oysters infected by closely related parasites, including Bonamia exitiosa. A calibration curve using a cloned template was defined to estimate copy number. The assay had a 6 log- dynamic range, mean inter- and intra-assay variation coefficients of <1% and a minimum detection limit of 50 gene copies per reaction. Using... |

Vibrio tapetis is the marine bacterium responsible for the brown ring disease (BRD) affecting die manila clam, Ruditapes philippinarum. Identification of V tapelis has been previously performed using biochemical criteria and serological procedures. All of these methods are time consuming and ill-adapted to individual screening. This study describes an oligonucleotidic probe (Vt446) and two PCR primers, deduced from the 16S rDNA sequence, allowing a fast and specific V tapetis identification using dot blot hybridisation and species-specific primed PCR (SSP-PCR). The probe and primers have been tested on 60 strains, including referenced Vibrio sp., Gram negative and positive bacteria, marine bacteria samples and isolated clam bacteria. For all the 19 V... |

Bonamiosis was first reported in association with mass mortality of flat oysters, Ostrea edulis, in June 1979 in L'Ile Tudy, Brittany. The disease rapidly spread to all of the oyster farming areas in France but also in other European countries. The French flat oyster production, which already suffered from another protozoan disease, marteiliosis, decreased from 5500 mt in 1979 to less than 2000 mt after 1980. In 2001, about 350 French farms sell 1650 mt of flat oysters. This production mainly relies on natural spat collection which specially occurs in Quiberon bay. One third of this spat is transferred from South to North Brittany for further growth. Quiberon bay constitutes an interesting site regarding the surveillance of bonamiosis because of the... |

Eight WEFTA laboratories participated in two series of intercomparison exercises on sulfite determination in tropical shrimps. Samples of tropical shrimps were spiked with sodium metabisulfite (Na2S2O5) and hydroxymethylsulfonate (HMS) at a level of 25-90 mg SO2/kg. Most of the laboratories determined the sulfite content with (modified) methods of Monier-Williams or De Vries et al. The overall mean recovery of sulfite was rather low (47-60%), which may be attributed to an irreversible reaction of sulfite with the tropical shrimps. The repeatability of the methods within the participating laboratories was good. However, reproducibility among the laboratories was... |

ABSTRACT This study determined the presence of Tremovirus A as the possible agent related to Avian Encephalomyelitis in broiler chicks from the states of São Paulo (SP) Paraná (PR), Goiás (GO), Santa Catarina (SC) and Rio Grande do Sul (RS), between the years 2006 and 2015. Samples of the nervous, digestive, respiratory, immune, and renal systems, plus muscular organs from broiler chicks with neurological problems such as ataxia and muscle tremors, and four (4) commercial vaccines as positive control, were tested by reverse-transcriptase (RT-PCR) amplification and DNA sequencing. A highly conserved region (P1) of the viral genome, was used to amplify a segment which encodes a structural protein VP4. Out of 112 samples, 46 were positive (42%) for Tremovirus... |

ABSTRACT In 2006, the International Organization for Standardization (ISO) published two standard protocols for the detection and enumeration of Campylobacter spp. in foods: the ISO 10272-2(direct method) and the ISO 10272-1 (enrichment method). The aim of this study was to compare the efficiency of these two methods in the detection of Campylobacter spp. and propose a modification in the enrichment method. Compared with the enrichment method, the direct method yielded a higher number of positive results for Campylobacter spp. and, consequently, presented higher sensitivity percentage. The isolation of Campylobacter spp. was more difficult when 10 mL of rinse was used in the enrichment method, which is currently recommended by the ISO protocol. Therefore,... |

An immunization scheme for production of antiserum to staphylococcal enterotoxin A (SEA) is proposed. The reference method of Robbins and Bergdoll was modified to reduce the number of doses and the amount of toxin used per animal. The best immunization scheme used injections in days 0, 8, 24, 59, 62 and 67. The amount of toxin at each injection was 5, 6, 20, 50, 100 and 200<FONT FACE="Symbol">m</font>g, respectively. The total amount of toxin was 381<FONT FACE="Symbol">m</font>g, which corresponded to a reduction of 107<FONT FACE="Symbol">m</font>g in the amount of toxin for each animal when compared to the reference method. The average antiserum titer using the Optimum Sensitivity Plate - OSP was 1:60 and using ELISA... |

Potato yellow vein virus (PYVV) expresses yellowing symptoms on plants and it is transmitted by the whitefly vector Trialeurodes vaporariorum Westwood. No molecular viral detection from the vector extracts has been reported to date. The objectives of this study were I) amplification of the PYVV major coat protein (CP) gene from viruliferous vector extracts by RT-PCR using specific primers, II) cloning and III) sequencing and sequences analysis. Some Colombian Solanum phureja (Jus et Buk) field potato plants expressing yellowing symptoms and confirmed PYVV positive by RT-PCR were used as virus donor plants for feeding whiteflies. Viruliferous vectors were obtained by depositing aviruliferous whiteflies on symptomatic leaves and kept in a muslin... |

Advances in genetic engineering permit the modification of plants to be tolerant to certain herbicides that are usually not selective. For practical and commercial purposes, it is important to be able to detect the presence or absence of these traits in genotypes. The objective of this research was to develop a procedure for identifying genetically modified soybean (Glycine max L. Merr.) with resistance to the herbicide glyphosate. Two studies were conducted based on germination test. In the first study, soybean seeds were pre-imbibed in paper towel with the herbicide solutions, then transferred to moist paper towel for the germination test. In the second study, seeds were placed directly in herbicide solutions in plastic cups and tested for germination... |

Garlic viruses often occur in complex infections in nature. In this study, a garlic virus complex, collected in fields in Brazil, was purified. RT-PCR was performed using specific primers designed from the consensus regions of the coat protein genes of Onion yellow dwarf virus, a garlic strain (OYDV-G) and Leek yellow stripe virus (LYSV). cDNA of Garlic common latent virus (GCLV) was synthesized using oligo-dT and random primers. By these procedures individual garlic virus genomes were isolated and sequenced. The nucleotide sequence analysis associated with serological data reveals the presence of two Potyvirus OYDV-G and LYSV, and GCLV, a Carlavirus, simultaneously infecting garlic plants. Deduced amino acid sequences of the Brazilian isolates were... |

Diurnal variation in detection can influence bird census results. I measured variation in daily and seasonal detections of birds in a forest fragment at Barreiro Rico ranch, Anhembi, state of São Paulo. Birds were recorded on transects of unlimited distance, sampled one day per month, dawn to dusk, between December 2000 and March 2001 (rainy season) and between May and August 2001 (dry season). Significant daily variation in detection occurred during the rainy season, with most detections between dawn and 9 h. In the dry season detections declined for total numbers of individuals and species and most detections occurred during late morning and mid afternoon. Detection patterns might reflect seasonal variation in bird behavior rather than seasonal variation... |
