Effects of cereal type, enzyme and sodium butyrate addition on growth performance, carcass traits and intestinal morphology of broilers Rev. Bras. Ciênc. Avic.
Shahir,MH; Moradi,S; Afsarian,O; Esmaeilipour,O.
A 2 × 2 × 2 factorial arrangement was conducted to evaluate the effects of cereal type (corn vs. wheat), enzyme supplementation (0 or 0.4 g/kg diet), and sodium butyrate addition (0 or 2 g/kg diet) on growth performance, carcass traits and intestinal morphology of male broilers from 10 to 42 d. A total of 600 male Arbor Acres chickens were assigned to eight dietary treatments in a completely randomized design, providing five replicate pens per treatment. Diets were formulated to meet or exceed NRC nutrient recommendations. When compared with birds fed corn-based diets, chickens fed wheat-based diets had inferior weight gain (1978 vs. 1807 g, p < 0.01) and feed per gain ratio (1.81 vs. 1.93; p < 0.01). Both weight gain (1870 vs. 1915 g; p < 0.01)...

Effect of ph and temperature on the activity of phytase products used in broiler nutrition Rev. Bras. Ciênc. Avic.
Naves,L de P; Corrêa,AD; Bertechini,AG; Gomide,EM; Santos,CD dos.
The activity of three commercial microbial phytase (Aspergillus oryzae, A. niger, and Saccharomyces cerevisae) products used in broiler nutrition was determined at different pH (2.0 to 9.0) and temperature (20 to 90°C) values. Enzymatic activity was determined according to the reaction of the phytase with its substrate (sodium phytate), in four replicates, and was expressed in units of phytase activity (FTU). A. oryzae phytase exhibited optimal activity at pH 4.0 and 40°C, but its absolute activity was the lowest of the three phytases evaluated. A. niger phytase exhibited maximal activity close to pH 5.0 and 45ºC, whereas S. cerevisae phytase presented its highest activity at pH close to 4.5 and temperatures ranging between 50 and 60°C. It was concluded...

ABSTRACT Two trials were performed to evaluate the effects of an enzyme complex (EC) on broiler performance, metabolizable energy, and phosphorus and nitrogen balance. In the first trial, 960 one-day-old male Cobb broilers were randomly distributed in a 3 x 2 factorial arrangement (three nutritional levels and the inclusion or not of EC), totaling six treatments with eight replicates of 20 birds per pen, and reared until 42 days of age. The control diet was formulated to meet the broilers' nutritional requirements. The other diets contained reduced available phosphorus (P), metabolizable energy, and amino acid levels, considering 100% and 150% of the EC nutritional matrix. The inclusion of the EC (200 g/ton) resulted in better broiler performance,...

Afonso,Esther Ramalho; Nacimento,Rafael Araujo; Palhares,Julio Cesar Pascale; Gameiro,Augusto Hauber.
ABSTRACT The objective with this study was to evaluate the impact of different swine nutritional strategies and technologies, as well as the land spread system, on logistical costs of pig manure distribution. For this, pigs were fed a conventional diet (C0) or a diet supplemented with 0.01% phytase (C1); organic minerals (40% substitution; C2); synthetic amino acids at reduced dietary crude protein levels (C3); or a combination of the three strategies (C4). All pig manure was weighed, then its chemical composition was determined. The mineral values and volume of manure produced were extrapolated to corresponding pig farm sizes in Sao Paulo state (300, 650, and 1000 sows) from real pig manure levels to produce simulations. To determine the logistical cost...

Effects of silicon and drought stress on biochemical characteristics of leaves of upland rice cultivars Rev. Ciênc. Agron.
Mauad,Munir; Crusciol,Carlos Alexandre Costa; Nascente,Adriano Stephan; Grassi Filho,Hélio; Lima,Giuseppina Pace Pereira.
ABSTRACT Silicon (Si) has beneficial effects on many crops, mainly under biotic and abiotic stress. Silicon can affect biochemical, physiological, and photosynthetic processes and, consequently, reduce drought stress. However, the effects of Si on rice (Oryza sativa L.) plants under drought stress are not well known. The objective of this study was to evaluate the effects of supplemental Si on proline content and peroxidase activity in upland rice plants in the presence and absence of drought stress. The experiment was conducted under greenhouse conditions and was arranged in completely randomized blocks in a 2 x 2 x 2 factorial scheme. Treatments comprised combinations of (1) presence or absence of Si supply (0 or 350 kg ha-1 of Si), (2) presence or...

Genetic analysis of greenbug populations of Schizaphis graminum (Hemiptera: Aphididae) from Argentina and Chile based on enzyme variability Rev. Soc. Entomol. Argent.
Saldúa,Luciana; Tacaliti,María S.; Tocho,Erica; Dixon,Anthony F. G.; Castro,Ana M..
Twenty-nine Schizaphis graminum (Rondani) populations and sixty clones collected from very contrasting regions of Argentina and Chile were investigated electrophoretically. A high degree of enzymatic polymorphism was found. The enzymatic structure was described for the esterase system, finding nine different loci. Latitudinal stratification was determined and populations were associated into three groups, according to the latitude they were collected from. The 90% of loci resulted polymorphic in the first group and 100% of loci were polymorphic in the rest. Observed heterozygosity was lower than expected. No group had fixed alleles according to Fsr values; group one had a slight heterozygous excess, while the other groups showed slight positive Fsr values,...

Nitrate reductase activity in leaves and stems of tanner grass (Brachiaria radicans Napper) Scientia Agricola
Cazetta,Jairo Osvaldo; Villela,Luciana Cristine Vasques.
Tanner grass (Brachiaria radicans Napper) is a forage plant that is adapted to well-drained soils or wetlands, and responds well to nitrogen (N) fertilization. The assimilation of N involves the nitrate reductase (NR) enzyme, and its activity seems to be dependent on N supply. Molybdenum (Mo) is also important because it is a cofactor of NR. In this study, the variables of an in vivo assay were optimized for measuring nitrate reductase activity (NRA) in the leaves and stem tissues. This method was used to evaluate NO3- metabolism in plants fertilized with NaNO3, NH4Cl or urea, in association with or without application of H2MoO4, aiming to provide guidelines for N management of this species. The best conditions to determine NRA involved the incubation of...

Extraction, partial characterization and susceptibility to Hg2+ of acid phosphatase from the microalgae Pseudokirchneriella subcapitata Scientia Agricola
Jonsson,Claudio Martín; Aoyama,Hiroshi.
Pseudokirchneriella subcapitata is a unicellular green algae widely distributed in freshwater and soils. Due to its cosmopolitan characteristic, its use is recommended by national and international protocols in ecotoxicity studies. The alteration of phosphatase activities by agriculture pollutants like heavy metals has been extensively used as a biomarker in risk assessment and biomonitoring. In this study, we compared the extraction of acid phosphatase from P. subcapitata by different procedures and we studied the stability, substrates specificity, kinetics and the effect of Hg2+ in the crude extract. The freezing and thawing technique associated with probe sonication was the most suitable method of extraction. The enzyme was stable when frozen at -20ºC...

Low carbon/nitrogen ratio increases laccase production from basidiomycetes in solid substrate cultivation Scientia Agricola
D'Agostini,Érica Clarissa; Mantovani,Talita Rafaele D'Agostini; Valle,Juliana Silveira do; Paccola-Meirelles,Luzia Doretto; Colauto,Nelson Barros; Linde,Giani Andrea.
Basidiomycetes are laccase producers used for hydrolysis of lignocellulosic byproducts in fermentative processes and could be used on biofuel production or ruminant feeding. The objective of this study was to evaluate the effect of concentrations of non-protein nitrogen sources on laccase production and mycelial growth of Pleurotus ostreatus, Lentinula edodes and Agaricus blazei. The fungi were grown on soybean hulls to which urea (U), ammonium sulfate (AS) or mixture of AS:U (1:1) were added to achieve carbon/nitrogen (C/N) ratios of 5, 15, 20 or 30. The average longitudinal mycelial growth was measured and laccase activity was determined by the oxidation of 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid. Higher C/N ratios increased mycelial growth...
