Abstract The importance of glutathione (GSH) in alternative cellular roles to the canonically proposed, were analyzed in a model unable to synthesize GSH. Gene expression analysis shows that the regulation of the actin cytoskeleton pathway is strongly impacted by the absence of GSH. To test this hypothesis, we evaluate the effect of GSH depletion via buthionine sulfoximine (5 and 12.5 mM) in human neuroblastoma MSN cells. In the present study, 70% of GSH reduction did not induce reactive oxygen species, lipoperoxidation, or cytotoxicity, which enabled us to evaluate the effect of glutathione in the absence of oxidative stress. The cells with decreasing GSH levels acquired morphology changes that depended on the actin cytoskeleton and not on tubulin. We... |

Partial or total replacement of fish meal by a mixture of plant protein (PP) sources (corn gluten, wheat gluten, extruded peas, rapeseed meal and sweet white lupin) balanced with indispensable amino acids was examined in juvenile gilthead sea bream over the course of a 6-month growth trial. A diet with fish meal (FM) as the sole protein source was compared to diets with 50%, 75% and 100% of replacement (PP50, PP75, and PP100). The possible influence of diets on growth performance, plasma metabolites, gut integrity, liver structure, anti-oxidant and immune status was evaluated. Final body weight was progressively decreased with PP inclusion, but in PP50 and PP75-fed fish, feed efficiency (FE) was significantly improved and specific growth rates remained... |

This paper describes a method for the determination of reduced sulfur compounds in hydrothermal seawater and body fluids from the hydrothermal tube worm Riftia pachyptila. Sulfur is a hey component of the hydrothermal ecosystem based on chemosynthesis, Sulfur compounds were derivatized at pH 8 (4.3 for sulfide in biological fluid) with a fluorescent reagent, monobromobimane, and separated by reverse-phase HPLC. The eluted compounds were detected by spectrofluorimetry. This method allowed: the quantitative analysis of sulfide, sulfite, thiosulfate, cysteine and glutathione in seawater, vascular blood and coelomic fluids from R, pachyptila, The detection limits were in the 0.1 mu mol l(-1) range with a precision lower than 10%, The method has been applied to... |

Cadmium (Cd), is an environmental and industrial pollutant that affects the male reproductive system. Cd induces its effect by affecting tissue antioxidant enzyme systems. Green tea extract (GTE) is an antioxidant and free radicals scavenger and has a chelating property. The purpose of this study was to investigate the protective effect of GTE against testes damage induced by Cd. Four groups of male rats, were utilized as following: Controls, GTE treated, Cd treated and Cd + GTE, treated rats at the same doses. The rats received GTE and or Cd orally in drinking water. After 5 weeks, the animals were sacrificed and testes were removed for microscopic and Biochemical evaluation. The levels of lipid peroxides (LPO) and glutathione (GSH) were detected in the... |

Early detection of toxic events induced by xenobiotics is necessary for a proper assessment of human risk after the exposure to those agents. The aim of this work was to evaluate the cell line HEp-2 as an experimental model to determine the genotoxic effects of sodium arsenate. To this end, we determined the metabolic activity cells by the MTT test on seven concentrations of arsenate that range from 27 to 135,000 μM, obtaining the median lethal concentration (LC50), the lowest observed effect concentration (LOEC), and the not observed effect concentration (NOEC) of sodium arsenate at 24 h of exposition. According to the cytotoxic response obtained, we evaluated the genotoxic effect of the 27 and 270 μM concentrations by using the... |

Plants of Azolla caroliniana were treated with different selenium concentrations (0, 1, 2, 5, 7, 10 ppm) for seven days. Selenium (Se) content in Azolla plants increased significantly with increasing Se concentrations in the culture media up to 5 ppm. This indicated that Azolla plants were a good accumulator for Se. Selenium accumulation determined changes in Azolla biomass, doubling time and relative growth rates. Treatment of Azolla plants with low concentrations of Se (1 ppm) resulted in a significant increase in biomass. This was accompanied by a reduction in hydrogen peroxide and malondialdehyde (MDA) contents; the decrease percentages were 78% and 60%, respectively at 1 ppm Se in comparison with the control. At higher Se concentrations (>5 ppm),... |

ABSTRACT: In this study, the micropropagation of Impatiens balsamina was established from stem and shoot explants. The effects of GA3 and glutathione on the morphogenesis of this species were also investigated, in order to induce in vitro flowering. It was found that the optimum in vitro plant regeneration was achieved on MS medium supplemented with 1.0 mg L-1 GA3 and in vitro flowering was also obtained from the same medium after 4 weeks of culture. To understand cellular behavior during in vitro flowering, Mitotic Index (MI), chromosome counts, measurement of mean cell and nuclear areas, DNA measurements and ploidy levels were analyzed from in vivo plants, in vitro grown plants and plantlets that flowered in vitro. The chromosome count was the same for... |

ABSTRACT The aim of this study was to assess the effect of selenium-enriched Candida utilis with high contents of organic selenium (Se) and glutathione (GSH) on growth performance, antioxidant capacity and immune function of broiler chickens. A total of 100 healthy 7-day-old male broiler chickens were randomly divided into 5 groups, and fed diets supplemented with (a) Na2SeO3, (b) C. utilis, (c) Se-enriched Saccharomyces cerevisiae, (d) Se-enriched C. utilis, and (e) the control without any supplements. The experiment lasted for 6 weeks and parameters were recorded on day 42. No significant differences in average daily gain were found among the 5 groups. However, Se-enriched C. utilis supplemented in the diet increased activities of glutathione peroxidase... |
