Genetic improvement of hatchery propagated bivalve stocks : prospects and constraints ArchiMer
Boudry, Pierre.
To date, the most significant genetic improvement for the production of Pacific oyster (Crassostrea gigas) has been obtained through the production of triploids, especially since the development of tetraploids. Quantitative genetics studies suggest that significant gains, for disease resistance or other traits, could be obtained in diploids. However, the limited extent of hatchery-propagation (versus natural recruitment) and/or various technical difficulties and biological characteristics of the species have retarded the development of selective breeding programs. In the U.S.A., Australia and New Zealand, family-based selective breeding programs have recently been initiated to improve growth, disease tolerance and yield. In Europe, where both natural and...

Isolation of 12 microsatellite markers following a pyrosequencing procedure and cross-priming in two invasive cryptic species, Alexandrium catenella (group IV) and A. tamarense (group III) (Dinophyceae) ArchiMer
Laporte, Martin; Shao, Zhaojun; Berrebi, Patrick; Laabir, Mohamed; Abadie, Eric; Faivre, Nicolas; Rieuvilleneuve, Fabien; Masseret, Estelle.
Alexandrium catenella (group IV) and Alexandrium tamarense (group III) (Dinophyceae) are two cryptic invasive phytoplankton species belonging to the A. tamarense species complex. Their worldwide spread is favored by the human activities, transportation and climate change. In order to describe their diversity in the Mediterranean Sea and understand their settlements and maintenances in this area, new microsatellite markers were developed based on Thau lagoon (France) samples of A. catenella and A. tamarense strains. In this study twelve new microsatellite markers are proposed. Five of these microsatellite markers show amplifications on A. tamarense and ten on A. catenella. Three of these 12 microsatellite markers allowed amplifications on both cryptic...

Towards the development of clonal lines in the European sea bass (Dicentrarchus labrax L.): application of uniparental reproduction techniques with an insight into sea bass eggs ArchiMer
Colleter, Julie.
Clonal lines are a powerful scientific tool for improved genetic characterization of organisms used in research. Inbred fish lines can be produced in only two generations using uniparental reproduction techniques. Androgenesis, achieved with variable success in several freshwater species, has been attempted in the European sea bass (Dicentrarchus labrax L), a marine fish of commercial and scientific interest. The low yields of progenies inheriting only the paternal genome after UV-irradiation of eggs led to considerations on the occurrence of UV screening compounds in pelagic eggs. Mycosporine-like amino acids and gadusol were found in many marine and freshwater organisms, but their occurrence in fish eggs was not clearly related to a behavioral pattern...

Genetic impact of the reproduction dynamics in the European flat oyster Ostrea edulis ArchiMer
Lapegue, Sylvie; Taris, Nicolas; Lallias, Delphine; Bonhomme, Francois; Boudry, Pierre.
The European flat oyster (Ostrea edulis L.) is a marine bivalve whose natural geographical distribution ranges along the European Atlantic coast from Norway to Morocco, in addition to the Mediterranean and Black Sea. The latest results obtained on the genetic differentiation between these populations have led us to persue studies at a finer scale, in order to estimate the effective number of breeders and the temporal dynamics of reproduction and, more specially, recruitment. Several experiments were performed to document (1) the variance in allele frequencies during a natural settlement period, (2) the paternal contribution to fertilization by analyzing larvae sampled at the brooding stage within individual females, (3) the variance of individual...

Mitochondrial and microsatellite genetic differentiation of the flat oyster Ostrea edulis along the european coasts, and comparison with allozyme data ArchiMer
Lapegue, Sylvie; Diaz-almela, E.; Launey, Sophie; Ledu, Christophe; Boudry, Pierre; Naciri-graven, Yamama; Bonhomme, Francois.
Natural populations of the European flat oyster, Ostrea edulis, have been harvested since Roman times. This species is distributed along the European Atlantic coasts, from Norway to Morocco, as well as the Mediterranean Sea and Black sea coasts. Previous studies, based on allozymes, have concluded that isolation by distance is taking place in the Mediterranean sea and the Atlantic ocean. Because of the presumed non neutrality of these markers, another approach was made to examine the genetic differentiation of this species using nuclear microsatellite and mitochondrial markers, both considered as neutral, and to compare the results to those observed with allozymes.

Microsatellite markers as a tool to study reproductive success in the Pacific oyster, Crassostrea gigas (Thunberg), under controlled hatchery conditions ArchiMer
Boudry, Pierre; Collet, Bertrand; Cornette, Florence; Hervouet, Veronique; Bonhomme, Francois.
Oysters, like many marine species have a very high fecundity. Previous studies have shown that populations, from both hatcheries and the natural environment, have very low Ne/N ratios. These observations reveal high variation in reproductive success. In order to study individual reproductive success under controlled conditions, we used microsatellite markers to quantify parental contributions in in vitro crosses (5 males and 5 females) of Crassostrea gigas, the Pacific oyster. High polymorphism of the microsatellites (more that 50 alleles per locus) eased the parentage identifications. The results of a cross allowing gametic competition were compared with the results from a second cross where the gametes of the same parents were kept separate for each...

High connectivity across the fragmented chemosynthetic ecosystems of the deep Atlantic Equatorial Belt: efficient dispersal mechanisms or questionable endemism? ArchiMer
Teixeira, Sara; Olu, Karine; Decker, Carole; Cunha, Regina L.; Fuchs, Sandra; Hourdez, Stephane; Serrao, Ester A.; Arnaud-haond, Sophie.
Chemosynthetic ecosystems are distributed worldwide in fragmented habitats harbouring seemingly highly specialized communities. Yet, shared taxa have been reported from highly distant chemosynthetic communities. These habitats are distributed in distinct biogeographical regions, one of these being the so-called Atlantic Equatorial Belt (AEB). Here, we combined genetic data (COI) from several taxa to assess the possible existence of cryptic or synonymous species and to detect the possible occurrence of contemporary gene flow among populations of chemosynthetic species located on both sides of the Atlantic. Several Evolutionary Significant Units (ESUs) of Alvinocarididae shrimp and Vesicomyidae bivalves were found to be shared across seeps of the AEB. Some...

Evolutionary dynamics of Cucurbita argyrosperma from the Mesoamerican domestication center using SSR molecular markers PAB
Priori,Daniela; Villarreal,Daniel Zizumbo; Ek,Víctor Manuel de Jesús Canché; Limones-Briones,Verónica; Barbieri,Rosa Lía.
Abstract: The objective of this work was to evaluate the evolutionary dynamics of the wild-weedy-domestic gene pool of Cucurbita argyrosperma squash by estimating the levels of genetic diversity and gene flow in the putative area of its domestication. Nine populations were collected, and DNA was extracted from young leaves harvested separately from approximately 20 individuals in each population. The DNA fragments were amplified with eight pairs of SSR primers and separated by electrophoresis in 5% denaturing polyacrylamide gels. Genetic diversity and the amount of gene flow were estimated in the populations, and Bayesian grouping was used to determine the levels of gene infiltration and probability of ancestry. The ethnobotanical exploration indicated...

Genetic similarity in a hybrid population of 'Montenegrina' and 'King' mandarins PAB
Brugnara,Eduardo Cesar; Bastianel,Marinês; Weiler,Roberto Luis; Schwarz,Sergio Francisco.
The objective of this work was to confirm the hybrids obtained in plants originated from the crossing between the mandarins Citrus deliciosa 'Montenegrina' and C. nobilis 'King', and to estimate the genetic similarity among hybrids, and between each hybrid and its parents. Twenty‑three pairs of microsatellite primers were tested. Fourteen of these pairs showed polymorphic bands between parents. Primers CCSM 129 and CCSME 52 were sufficient to identify the 12 nucellar clones observed in the studied population. Genetic similarity analysis of the population (hybrids and parents) showed 0.56 average similarity. Besides the 12 clones of 'Montenegrina' identified, 25 hybrids were found of which D18, C32, D06, C05 and D09 are the more similar to 'Montenegrina'.

Interactions Between Non-Allelic Loci and Their Effects on Categorized Meat Performance Traits in Ducks Rev. Bras. Ciênc. Avic.
Dobek,A; Gornowicz,E; Moliński,IA; Grajewski,B; Lisowski,M; Szwaczkowski,T.
ABSTRACT Traits associated with meat performance are characterized by a complex inheritance model, including the effects of non-allelic interactions. The aim of this study was to estimate the significance of non-allelic interactions between pairs of loci in each of two alternative groups based on four important performance traits of ducks: body weight (BW), carcass yield calculated as the ratio of carcass weight to body weight (CY), breast and leg muscle yield as a fraction of carcass weight (BL-CW), and skin weight and abdominal fat, again as a fraction of carcass weight (SF-CW). The experimental material was based on an F2 cross between two parental lines: A-55 (Polish Peking) and GL-30 (French Peking). In total, 368 individuals were recorded. All of the...

Freitas,Suzane Fonseca; Moreira,Heden Luiz Marques; Souza,Daiane Machado; Moreira,Carla Giovane Ávila; Tavares,Rafael Aldrighi; Dionello,Nelson José Laurino.
ABSTRACT This study aimed to develop PCR assays, reaction combinations, and validation of tetranucleotide SSR loci for tilapia, to minimize the impact of erroneous allele inferences on genotype determination of this marker. Microsatellites containing tetranucleotide repeats were obtained from tilapia genome, version 2.1, avoiding loci in the same linking group. Primers were designed for different fragment sizes, and fluorescence added to each locus. A total of 10 loci were amplified, separately and in combination, and loaded into a single capillary sequencer panel. Alleles were amplified without stutters and easily interpreted. PCR amplifications of DNA repeatedly extracted from samples, and genotyping at different PCR rounds were performed to infer allele...

Researches on genetic relationships among water buffalo populations are useful in supporting conservation decisions and designing of breeding schemes. The aim of this study was to assess the genetic relationships among Cuban Buffalypso/Carabao hybrids and Murrah and Jaffarabadi Brazilian buffaloes using nine cattle microsatellite markers. During the analysis, four loci were excluded due to the presence of null alleles, stuttering and gametic disequilibrium. Mean numbers of alleles and the heterozygosities observed and expected in the three populations were not significantly different. Heterozygosities found were above 60%. All five loci were in Hardy-Weinberg equilibrium. Wright´s index for estimating inbreeding were negative in the three populations. A...

ABSTRACT Some germplasm collections have a high number of accessions, which makes it difficult to explore the genetic variability present in the germplasm bank due to the redundancy and the difficulty of detailed analysis of all conserved accessions. Therefore, our study aimed to analyze the genetic diversity of 153 lima bean (Phaseolus lunatus) accessions for the purpose of constructing a core collection. Eleven SSRs were used for this purpose. The 153 lima bean accessions can be represented by low redundancy using a minimum of 34 accessions, thus representing 22 % of the size of the entire germplasm bank. The core collection had a higher Shannon diversity index and expected heterozygosity (1.906 and 0.811, respectively) than those presented by the entire...

Genetic linkage maps of chicken chromosomes 6, 7, 8, 11 and 13 from a Brazilian resource population Scientia Agricola
Ambo,Marcel; Campos,Raquel de Lello Rocha; Moura,Ana Sílvia Meira Tavares; Boschiero,Clarissa; Rosário,Millor Fernandes do; Ledur,Mônica Corrêa; Nones,Kátia; Coutinho,Luiz Lehmann.
A linkage map is essential not only for quantitative trait loci (QTL) mapping, but also for the organization and location of genes along the chromosomes. The present study is part of a project whose major objective is, besides from construction the linkage maps, the whole genome scan for mapping QTL for performance traits in the Brazilian experimental chicken population. Linkage maps of chicken chromosomes 6 to 8, 11 and 13 were constructed based on this population. The population was developed from two generations of crossbreeding between a broiler and a layer line. Fifty-one microsatellite markers were tested, from which 28 were informative: 4, 8, 7, 4 and 5 for chromosomes 6, 7, 8, 11 and 13, respectively. A SNP located in the leptin receptor gene was...

A NEW WORLD MONKEY MICROSATELLITE (AP74) HIGHLY CONSERVED IN PRIMATES Acta biol.Colomb.
OKLANDER,LUCIANA INÉS; STEINBERG3,ELIANA RUTH; DOLORES MUDRY,MARTA.
Given their great variability, microsatellites or STRs became the most commonly used genetic markers over the last 15 years. The analysis of these markers requires minimum quantities of DNA, allowing the use of non invasive samples, such as feces or hair. We amplified the microsatellite Ap74 in blood and hair samples in order to analyze the levels of genomic conservation among a wide range of primates including: Lemur catta, Alouatta caraya, Ateles belzebuth, Ateles chamek, Pan troglodytes, Papio sp., and Homo sapiens. In all cases we obtained amplification products that exhibited similar size both in monkeys and human (oscillating between 126 and 176 bp), except in the lemur where the detected fragment presented a size of approximately 1000 bp. The...

Population structure and impact of recurrent selection on popcorn using EST-SSR markers Agronomy
Guimarães, Amanda Gonçalves; Amaral Júnior, Antônio Teixeira do; Almeida Filho, Janeo Eustáquio de; Pena, Guilherme Ferreira; Vittorazzi, Cássio; Pereira, Messias Gonzaga.
The success of any recurrent selection program depends on the genetic variability of the evaluated population, which is used to refer to the diversity of existing alleles at many genetic loci. Thus, the goal of the present study was to investigate the impact of recurrent selection across nine cycles of a UENF-14 popcorn population through the analysis of genetic diversity and structure using microsatellite markers (EST-SSRs). Genomic DNA was extracted from young leaves of 25 individuals from each cycle (C0, C1 C2, C3, C4, C5, C6, C7, and C8), totaling 225 samples from the UENF-14 population. Fifty EST-SSR markers were used for the analysis of genetic diversity across the recurrent selection cycles, 16 of which were polymorphic. Thirty-four alleles were...

This study aimed to compare multiallelic distances to quantify genetic diversity in papaya. We evaluated forty-three individuals in the S2 generation, from the backcross between F1 (Cariflora x SS783) and Cariflora, and four accessions from the UENF/Caliman Germplasm Bank. Genetic distances used were Smouse and Peakall (1999), Kosman and Leonard (2005) and weighted index. Clustering among genotypes was performed using the hierarchical unweighted pair-group method with arithmetic mean analysis (UPGMA) and projection of the distance on the bidimensional plan. A high correlation between genetic distances was observed; however, through UPGMA group analysis, the distance determined by the weighted index provided the complete separation of 52BC1S2-08, 52BC1S2-29...
