Asparaginase is an enzyme used in clinical treatments as a chemotherapeutic agent and in food technology to prevent acrylamide formation in fried and baked foods. Asparaginase is industrially produced by microorganisms, mainly gram-negative bacteria. Zymomonas mobilis is a Gram-negative bacterium that utilizes glucose, fructose and sucrose as carbon source and has been known for its efficiency in producing ethanol, sorbitol, levan, gluconic acid and has recently aroused interest for asparaginase production. ABSTRACT We evaluated the influence of hesperidin and vitamin C (VitC) on glycemic parameters, lipid profile, and DNA damage in male Wistar rats treated with sucrose overload. Rats were divided into six experimental groups: I-water control; II-sucrose control; III-hesperidin control; IV-VitC control; V-co-treatment of sucrose plus hesperidin; VI-co-treatment of sucrose plus VitC. High consumption of sucrose induces DNA damage in male Wistar rats Anais da ABC (AABC)

ABSTRACT The purpose of this study was to determine the effects of the high consumption of sucrose on the levels of DNA damage in blood, hippocampus and bone marrow of rats. Male Wistar rats were treated for 4 months with sucrose (10% for 60 initial days and 34% for the following 60 days) in drinking water, and then, glycemia and glycated hemoglobin (A1C) were measured. Levels of DNA damage in blood and hippocampus were evaluated by the comet assay. The micronucleus test was used to evaluate chromosomal damages in the bone marrow. RESUMO O objetivo deste trabalho foi analisar as leituras de composição do leite cru por meio de espectrofotometria FTIR, utilizando-se curva de regressão PLS, bem como as contagens de células somáticas e bacteriana total por citometria de fluxo, após adição de amido e sacarose. The aim of the present study was to investigate the efficiency of the encapsulation-dehydration technique for cryopreservation of Cyrtopodium hastchbachii Pabst seeds. Immature seeds of this species were cryopreserved by an encapsulation-dehydration technique. Evaluation of the shelf-life of vegetable-type soybean pods BABT

The shelf-life of the vegetable-type soybean pods stored under different conditions was evaluated by chemical characteristics and color. The pods were harvested in the R6 stage and stored either at 30 or 7ºC for 9 d. After the storage period, the pods were blanched and threshed, and the immature green grains were used for the analysis. The protein content decreased after 6 d of storage at 7ºC. There was no difference in the lipid content after the storage at 30 and 7ºC for 9 d. The starch and sucrose contents decreased after the first day of storage at 30ºC. Effects of culture conditions on the production of inulinase by Kluyveromyces marxianus BABT

The present study was conducted to investigate the influence of initial sucrose concentration, pH and aeration rate on biomass and inulinase production by Kluyveromyces marxianus var. bulgaricus in a stirred batch reactor. Maximum inulinase activity (15.29 UmL-1) was obtained at a sucrose concentration of 10 g L-1, pH 5.0 and aeration rate of 1 vvm. The 20 g L-1 sucrose concentration was suitable for cell growth; however, enzymatic activity at this concentration was inhibited due to catabolic repression. The increase in aeration rate caused a reduction in enzyme activity with no relevant biomass increase.

Variation in phytate accumulation in common bean (Phaseolus vulgaris L.) fruit explants BABT

The in vitro synthesis of phytate was studied in common bean fruit explants. Different concentrations of sucrose; phosphorus (P); myo-inositol; abscisic acid (ABA); glutamine and methionine, were tested. Fixed concentrations of these compounds were tested at different periods (0, 3, 6 and 9 days). Variation in phytate coincided with different concentrations of sucrose, myo-inositol, P and ABA for the duration tested. These compounds caused an accumulation of phytate and were more effective in the presence of myo-inositol and P. The accumulation of P varied less than phytate for the different treatments tested in vitro. Morphological and physiological photon flux influence under in vitro culture of apple shoots BABT

The aim of the present study was to evaluate the growth and development of apple rootstock shoots submitted to different photon flux densities (7, 14, 21 and 60 µmol m-2 s-1) and three culture media. The best photon flux for maximizing growth, number of shoots and leaves was 14 µmol m-2 s-1, which also resulted in the highest values of chloroplastic pigments. On the other hand, the highest photon flux (60 µmol m-2 s-1) decreased significantly these parameters. Medium with reduced sucrose concentration did not affect the analyzed parameters, enabling an efficient multiplication of the cultivar with only 1% of sucrose.

The production of sorbitol by permeabilized and immobilized cells of Z. mobilis in sucrose BABT

The production of sorbitol by permeabilized and immobilized cells of Zymomonas mobilis in Luffa cylindrica was investigated in sucrose medium. A full 2³ factorial design was used to verify the influence of each factor and its interactions. The cell permeabilization showed a significant and negative effect upon the production of sorbitol, while the time of cultivation and the immobilization process were significant and positive. The results demonstrated that the cell immobilization and the time of cultivation of 36 h presented higher production of sorbitol.

Compacted mouse morulae were frozen at 0.3ºC/min. or 0.5ºC/min. from -6ºC to -24ºC or -32ºC in 10% of glycerol plus different sucrose concentrations with or without 0.1% of honeybee royal jelly. Embryos were thawed in water bath at 22ºC for 20 seconds and cryoprotectant dilution was done in three steps. Embryos were cultured in Whitten’s medium for 24, 48 and 72 hours at 37ºC, 5% of CO2 and 100% of humidity. The in vitro development ranged from 56.6% to 100% after 72 hours. Expanded blastocysts were transferred to pseudopregnant recipients on the third day of the estrous cycle. Changes in sodium appetite evoked by lesions of the commissural nucleus of the tractus solitarius BJMBR

Ablation of the area postrema/caudal nucleus of the tractus solitarius (NTS) complex increases sodium intake, but the effect of selective lesions of the caudal NTS is not known. We measured depletion-induced sodium intake in rats with electrolytic lesions of the commissural NTS that spared the area postrema. One day after the lesion, rats were depleted of sodium with furosemide (10 mg/kg body weight, sc) and then had access to water and a sodium-deficient diet for 24 h when 1.8% NaCl was offered. Water and saline intakes were measured for 2 h. Saline intake was higher in lesioned than in sham-lesioned rats (mean ± SEM: 20 ± 2 vs 11 ± 3 mL/2 h, P < 0.05, N = 6-7). Woronin bodies in Penicillium janczewskii Zaleski BJM

Penicillium janczewskii Zaleski is an efficient microorganism for the production of extracellular inulinases and grows rapidly on medium containing sucrose or inulin as carbon source. Maintenance of this filamentous fungus on inulin medium induces secretion of large amounts of inulinases, but the resulting mycelium has thinner cell walls that easily collapse and break. Woronin bodies in hyphae of P. janczewskii grown on sucrose and inulin substrates were observed. Invertase, whether adsorbed on styrene-divinylbenzene copolymers or otherwise, was used for continuous sucrose hydrolysis using a cell-type membrane reactor (CTMR), coupled with an ultra (UF-100kDa), or a microfiltration (MF- pore diameter of 5 µm) membrane. Sweet potato (Ipomoea batatas L.) plants were cultivated in vitro in Murashige and Skoog (MS) medium with 20 and 40 g L-1 of sucrose under two different photon flux densities (21 and 60 µmol m-2 s-1). Leaves developed in vitro mostly showed high variable to maximum fluorescence ratio (F V/F M) , indicating good development of photosynthetic apparatus. Gas exchanges, sucrose, starch and reducing sugar contents were analyzed in leaves of orange trees with and without Citrus Variegated Chlorosis (CVC), a disease caused by the bacteria Xylella fastidiosa. Healthy plants (HP) and diseased plants (DP) were evaluated 20 and 26 months after bacterial inoculation, in October 1998 and April 1999, respectively. On the first evaluation date, CO2 assimilation rates for HP were greater than DP in the early morning and midday, but on the second date they were greater in HP all along the day. The present research aimed to characterize some biochemical responses of Coffea canephora (clones 02 and 153) and C. arabica (Catucaí IPR 102) genotypes subjected to low positive temperatures, helping to elucidate the mechanisms involved in cold tolerance. For that, one year old plants were subjected successively to 1) a temperature decrease (0.5°C a day) from 25/20°C to 13/8°C (acclimation period), 2) a three day chilling cycle (3x13/4°C) and to 3) a recovery period of 14 days (25/20°C). In Catucaí (less cold sensitive when compared to clone 02) there was an increased activity in the respiratory enzymes malate dehydrogenase and pyruvate kinase.
