A procedure is described to regenerate plants from protoplasts of Brazilian citrus cultivars, after isolation, fusion and culture. Protoplasts were isolated from embryogenic cell suspension cultures and from leaf mesophyll of seedlings germinated in vitro. The enzyme solution for protoplast isolation was composed of mannitol (0.7 M), CaCl2 (24.5 mM), NaH2PO4 (0.92 mM), MES (6.15 mM), cellulase (Onozuka RS - Yakult, 1%), macerase (Onozuka R10 - Yakult, 1%) and pectolyase Y-23 (Seishin, 0.2%). Protoplast culture in liquid medium after chemical fusion lead to the formation of callus colonies further adapted to solid medium. Somatic embryo formation occurred spontaneously after two subcultures, on modified MT medium supplemented with 500 mg/L of malt extract.... |

Abstract: The objective of this work was to evaluate how salt concentrations in media used for in vitro cultures affect the early development of “barueiro” (Dipteryx alata) seedlings obtained from the shoot apex. Two different in vitro culture media - Murashige & Skoog (MS) and wood plant medium (WPM) - in different salt concentrations (25, 50, 75, and 100%) were used. At 120 days, the D. alata seedlings in the MS medium, in its original concentration (100%), developed better than in other concentrations or in the WPM. Therefore, the MS medium was considered the most suitable for ensuring in vitro establishment because it provided greater root length (27.65 cm) and number of leaves per plant (26 leaves) than the other medium. |

Christ's thorn (Ziziphus spina-christi (L.) Desf.) is a cross-pollinated plant with a wide range of genetic variability in nature and, for this reason, vegetative propagation assumes importance for improvement programs. The objective of this work was to evaluate cutting, T budding and tissue culture methods for this species. Shoots of 22-25 cm length were treated by two culture media and three shoot diameters for cutting trial. The T budding treatments consisted of three and five collection dates in spring and autumn, respectively. Tissue culture nodal segments bearing axillary buds were removed from shoots of mature trees at different seasons. Experiments to determine the best disinfectant chemical, appropriate conditions and materials to prevent phenolic... |

A robust protocol for culturing meristems of Sorghum is required to assist with rapid genetic improvement of the genus. Through meristem culture, an efficient method for rapid micropropagation was developed for Sorghum bicolor (L.) Moench. hybrids, namely NC+262, NC+6C21 and NC+6B50. Complete plants were regenerated directly from shoot meristems without an intervening callus phase. Regeneration frequencies varied between the studied genotypes and according to the growth regulator combinations present in the medium. The combination of BAP and TDZ showed a synergistic effect on shoot multiplication. The highest number of shoots per meristem (68 ± 2) was recorded for the genotype NC+262, followed by NC+6C21 (31.3 ± 0.8), and NC+6B50 (16 ± 1.7). MS medium... |

The present work aimed at maximizing the number of plantlets obtained by the micropropagation of pineapple (Ananas comosus (L.) Merrill) cv. Pérola. Changes in benzylaminopurine (BAP) concentration, type of medium (liquid or solidified) and the type of explant in the proliferation phase were evaluated. Slips were used as the explant source, which consisted of axillary buds obtained after careful excision of the leaves. A Sterilization was done in the hood with ethanol (70%), for three minutes, followed by calcium hypochlorite (2%), for fifteen minutes, and three washes in sterile water. The explants were introduced in MS medium supplemented with 2mg L-1 BAP and maintained in a growth room at a 16h photoperiod (40 mmol.m-2.s-1), 27 ± 2ºC. After eight weeks,... |

The biotechnological techniques may help solve many problems of guava culture, such as the high perishability of fruits. Somatic embryogenesis can generate highly multiplicative cell cultures and with high regenerative potential, serving as basis for genetic transformation. The aim of this work was to obtain somatic embryogenesis of guava (Psidium guajava L.) cv. Paluma. Immature seeds were used, and they were inoculated in MS environment containing 400 mg L-1 of L-glutamine, 100 mg L-1 myo-inositol, 60 g L-1 sucrose, 100 mg L-1 ascorbic acid and supplemented with different types and concentrations of growth regulators. Embryogenic callus appeared after 37 days of culture in an environment containing 1.0 mg L-1 2.4-D + 2.0 mg L-1 2-ip, in 7% of the... |

Rangpur lime (Citrus limonia Osbeck) in vitro organogenesis was studied based on explant type and cytokinin culture media supplementation. Four explants types collected from epicotyl or hypocotyl regions of in vitro germinated seedlings were evaluated. The epicotyl-derived explants consisted of epicotyl segments and the hypocotyl-derived explants consisted of the entire hypocotyl segment, the hypocotyl segment attached to a cotyledon fragment, and the hypocotyl segment divided longitudinally. The explants were cultured on EME culture medium supplemented with benzylaminopurine (0, 0.5, 1.0, or 1.5 mg L-1). The evaluation was performed after 6 weeks. Best results considering both the explant responsiveness and number of shoots developed per explants were... |

ABSTRACT Guava (Psidium guajava L.) can be propagated by seed, layering, air layering, grafting (budding or grafting), cuttings (root or shoot) or tissue culture. Propagation by seed is used for rootstock production and for raising populations for screening at early phases in the breeding programs. Vegetative propagation methods are used to clone selected genotypes from these programs and commercial orchards because it perpetuates all characteristics of each cultivar. This review addresses different methods that can be used to propagate guava, the methods commercially adopted and the progress obtained in recent years. There are several propagation technologies available, however, the adoption levels are rather different between producing countries. Needs... |

Abstract The banana tree is one of the most cultivated fruit globally; however, some diseases significantly affect its production, such as Fusarium wilt. The most appropriate measure for controlling this disease in areas with inoculum pressure is the use of resistant cultivars. Therefore, this study aimed to generate banana somaclones of the cultivar ‘Prata-Anã’ resistant to Fusarium wilt by inducing somaclonal variation. ‘Prata-Anã’ stem apexes were established in vitro in MS culture medium and, on a monthly basis, subcultivated in AIA and adenine sulfate supplemented MS medium with added plant regulators: 6-benzylaminopurine (BAP, 4 ml L-1), Thidiazuron (TDZ, 1 ml L-1), and Paclobutrazol (PBZ, 10 ml L-1). The treatments were: T0: no regulator, T1: BAP,... |

ABSTRACT Nursery trees of stone fruits (Prunus spp.) are traditionally produced by union of two distinct genotypes - the rootstock and the scion - which, by grafting, form a composite plant that will be maintained throughout of all plant life. In Brazil, the rootstocks are predominantly seed propagated and therefore usually results in heterogeneous trees for vigor and edaphic adaptation. However, with advances in rootstock breeding programs that released cultivars and certification in several countries (notably in Europe), the system will come gradually evolving for vegetative propagation (cuttings and tissue culture) and use of seeds of selected rootstocks with specific characteristics and potted nursery trees production. For scion cultivar propagation,... |

ABSTRACT The aims of this study were to establish micropropagation for Chenopodium ambrosioides L. and analysis of volatile constituents in vitro as a function of different salt concentrations of MS medium and sucrose. The apical buds were established on MS medium added with 3% sucrose. The vegetative growth of nodal segments was tested in different salt concentrations of MS basic medium (0.25, 0.50, 1.00, 1.50, and 2.00) and of sucrose (5, 10, 20, 30, and 40 g L-1). Nodal and apical segments were tested in vertical and horizontal orientations. Different volumes of culture medium (20; 35, 50, 65, and 80 mL) were examined in the growth of nodal segments. Plants were acclimatized in three different substrates. The apical segments collected from matrices... |
