Molecular description and similarity relationships among native germplasm potatoes (Solanum tuberosum ssp. tuberosum L.) using morphological data and AFLP markers Electron. J. Biotechnol.
Solano Solis,Jaime; Morales Ulloa,Daniza; Anabalón Rodríguez,Leonardo.
Chile is considered to be a sub-center of origin for the cultivated potato, with native and introduced genetic material coexisting in the country. Thus, the different varieties present in Chiloe Island are characterized by a rich diversity of forms, sizes, colours and phenological characteristics. In the present work, the level of polymorphism and the genetic relationship were studied by means of molecular markers using the amplified fragment length polymorphism (AFLP) technique and twenty-seven morphological characters. Twenty varieties of potatoes from the Chiloe Island were analyzed. The commercial variety Desirée and one specie from the Etuberosa series, Solanum fernandezianum, collected in the Juan Fernandez Island were included as controls. A...

Immature embryo: A useful tool for oil palm (Elaeis guineensis Jacq.) genetic transformation studies Electron. J. Biotechnol.
Abdullah,Ruslan; Zainal,Alizah; Yew Heng,Wee; Chui Li,Leaw; Chee Beng,Yeap; Mei Phing,Lee; Abdullah Sirajuddin,Salwa; Soo Ping,Winnie Yap; Lourdes Joseph,Juanita; Azma Jusoh,Siti.
Oil palm (Elaeis guineensis Jacq.) is the highest yielding oil-bearing crop. However, being a perennial crop, genetic improvement of oil palm is extremely slow. Indeed, compared to other annual oil crops such as soybean and rapeseed, genetic manipulations remained less important. Therefore, to remain competitive, oil palm growers and breeders need new and novel approaches. In this report, the potential of immature embryos (IE) as a useful tool for oil palm genetic transformation studies was evaluated. It was evident that IEs were amenable to both direct and Agrobacterium-mediated gene transfer. Due to the abundant supply of IE, optimization of biolistic and Agrobacterium-mediated gene transfer into IEs were easily carried out. Transient transformation...

The upstream sequence of the phycocyanin b subunit gene from Arthrospira platensis regulates expression of gfp gene in response to light intensity Electron. J. Biotechnol.
Yongzhong,Lu; Zhang,Xuecheng.
In cyanobacteria, few details are known of the mechanisms through which the expression of the light-harvesting pigment c-phycocyanin is regulated. In the present study, a 419 bp upstream sequence of the phycocyanin b subunit (cpcB) gene from Arthrospira platensis FACHB341 was fused with green fluorescent protein (gfp) gene, and a heterologous reporting system was built up to investigate the influence of light intensity on the expression of gfp gene, and the regulation function of different region of the upstream sequence of cpcB gene. Results showed that the upstream sequence of cpcB gene could drive the expression of gfp gene in Synechococcus sp. strain PCC7942, and the expression was influenced by light intensity, the lower the light intensity, the...

Sarwat,Maryam; Singh Negi,Madan; Lakshmikumaran,Malathi; Kumar Tyagi,Akhilesh; Das,Sandip; Shankar Srivastava,Prem.
For studying genetic diversity in natural populations of Terminalia, a medicinal plant, our attempts to isolate high quality DNA using several previously reported protocols and even modifications were unsuccessful. We therefore combined CTAB based isolation, and column based purification step, to isolate DNA from Terminalia arjuna. The DNA isolated using this standardized protocol was high in quality and suitable for restriction digestion and generation of random amplification of polymorphic DNA (RAPD) and amplified fragment length polymorphism (AFLP).

Toxicity caused by para-substituted phenols on Tetrahymena pyriformis: The structure-activity relationships Electron. J. Biotechnol.
Jäntschi,Lorentz; Popescu,Violeta; Bolboaca,Sorana D.
The toxicity of thirty para-substituted phenols on Tetrahymena pyriformis was modelled using an original methodology that uses the complex structural information of the compounds. Two models were built. The methodology allows atomic properties to be assigned to toxicity based on the selection of pairs of descriptors from the entire family, which is called Molecular Descriptors Family (MDF). One model has two independent structural descriptors and the other has four. The model with four descriptors proved to have high estimated and predictive abilities (over 97% of toxicity could be explained by structural information). The partial charge distribution by bonds (molecular topology) and space (molecular geometry) interaction proved to be related with the...

Wang,Ping; Nong,Xu-Hua; Ge,Jie-Hong.
Nonylphenol ethoxylates (NPEOs), which are widely used for industrial and domestic purposes, exert adverse effects on wildlife after being used and discharged into the environment. However, their ultimate biodegradability and biodegradation pathway remains unclear. In this study, the aerobic degradability of nonylphenol ethoxylates (NPEOs) by the acclimated microorganisms in active sludge was examined using shaking-flask tests. The degradation of benzene rings in NPEOs was determined using UV spectroscopy and high performance liquid chromatography (HPLC). Results showed that more than 80% of benzene rings were removed after 8-10 days of degradation, and the majority of NPEOs were also removed after 9 days of degradation, indicating NPEOs and the benzene...

Development of pollen mediated activation tagging system for Phalaenopsis and Doritaenopsis Electron. J. Biotechnol.
Tsay,Hsin-Sheng; Ho,Hui-Min; Gupta,Sushim K; Wang,Chang-Sheng; Chen,Pei-Tsu; Chen,Emily Chin-Fun.
In the present study, a novel plant transformation system for Doritaenopsis and Phalaenopsis has been developed. The pollen-mediated activation tagging system was established by artificial pollination. The pollens, co-cultured with Agrobacterium tumefaciens strain EHA105 harbouring an activation tagging vector (pTAG-8), were used for pollination. In order to optimize the transformation efficiency, several factors (concentration of A. tumefaciens, concentration of acetosyringone during co-cultivation and the duration of co-cultivation) known to influence Agrobacterium-mediated DNA transfer were examined. A concentration of 0.5-1 x 10(8) CFU/ml for A. tumefaciens, 0.1 mM acetosyringone, and 6 hrs of co-culture period were found to be the optimal condition...

Resistance of diethylenetriaminepentaacetic acid to anaerobic biodegradation Electron. J. Biotechnol.
Alarcón,Evelyn; Decap,Jacqueline; Vidal,Gladys.
Kraft mills are responsible for the massive discharge of highly polluted effluents, and new bleaching processes (i.e. Total Chlorine Free (TCF)) is presented as a feasible option to reduce this environmental impact. However, increased TCF pulp production is accompanied by an increase in chelate use. The most commonly used chelates, ethylenediaminetetraacetic acid (EDTA) and diethylenetriaminepentaacetic acid (DPTA), are considered to be relatively persistent substances in water treatment plants, and consequently environmentally critical compounds. The purpose of this work is to investigate DPTA behaviour in an anaerobic system. An Anaerobic Filter (AF) was operated with three different DPTA load rates (LR DPTA = 0.07 - 0.28 gDPTA/L×d), and the...

Suppression subtractive hybridization PCR isolation of cDNAs from a Caribbean soft coral Electron. J. Biotechnol.
Lopez,Jose V; Ledger,Angela; Santiago-Vázquez,Lory Z; Pop,Mihai; Sommer,Dan D; Ranzer,Llanie K; Feldman,Robert A; Russell,G. Kerr.
Transcriptomic studies of marine organisms are still in their infancy. A partial, subtracted expressed sequence tag (EST) library of the Caribbean octocoral Erythropodium caribaeorum and the sea fan Gorgonia ventalina has been analyzed in order to find novel genes or differences in gene expression related to potential secondary metabolite production or symbioses. This approach entails enrichment for potential non-“housekeeping” genes using the suppression subtractive hybridization (SSH) polymerase chain reaction (PCR) method. More than 500 expressed sequence tags (ESTs) were generated after cloning SSH products, which yielded at least 53 orthologous groups of proteins (COGs) and Pfam clusters, including transcription factors (Drosophila...

Biosorption of uranium from aqueous solutions by nonliving biomass of marinealgae Cystoseira indica Electron. J. Biotechnol.
Hassan Khani,Mohammad; Reza Keshtkar,Ali; Meysami,Behrouz; Firouz Zarea,Mohammad; Jalali,Reza.
Biosorption of heavy metals can be an effective process for the removal and recovery of heavy metal ions from aqueous solutions. In this paper uranium biosorption by protonated, Ca-pretreated and non-pretreated Cystoseira indica algae biomass was investigated in a batch system. The results of the kinetic studies showed that the sorption of uranium on protonated and non-pretreated biomass followed pseudo-second order kinetics. The effect of pH on the equilibrium uranium sorption capacity of Cystoseira indica exhibited that highest uptake occurred at pH 4 at a solution with 350 mg/l uranium concentration. At various initial uranium concentrations from 50 to 1000 mg/l, batch sorption equilibrium at 30ºC was reached within 3 hrs and the sorption...

Treatment of low strength sewage with high suspended organic matter content in an anaerobic sequencing batch reactor and modeling application Electron. J. Biotechnol.
Donoso-Bravo,Andrés; Carballa,Marta; Ruiz- Filippi,Gonzalo; Chamy,Rolando.
In this work, an anaerobic sequencing batch reactor (ASBR) was operated for 8 months to treat low strength sewage with high suspended organic matter content. Three phases of operation with increasing organic loading rates (OLR) were performed: 0.4 kg COD/m³ x d (phase I), 0 .8 kg COD/m³ x d (phase II) and 1.2 kg COD/m³ x d (phase III). Adequate stability parameters (pH, total alkalinity) were obtained through all three experimental phases. During phases I and II, the removal efficiencies of organic matter (expressed as total chemical oxygen demand (COD) and total suspended solids ranged between 50-60%. However, these values decreased to 15-25% in phase III. In addition, a non-complex model, including hydrolysis, acidogenesis and methanogenesis, was applied...

Functional complementation in Saccharomyces cerevisiae under control of the natural yeast promoter Electron. J. Biotechnol.
Lavigne,Rob; Volckaert,Guido.
A novel approach for functional complementation of foreign genes in Saccharomyces cerevisiae is presented. This approach is based on the use of the widely available cognate gene plasmids (e.g. pRS416) of the European Functional Analysis Network (EUROFAN). The functional complementation of the human homolog of YOR159c (SME1 gene) shown here is the first demonstration of complementation using the original yeast promoter, theoretically offering a more natural regulation of protein expression
