Mazzella, N.; Delmas, F.; Delest, B.; Mechin, B.; Madigou, C.; Allenou, Jean-pierre; Gabellec, Raoul; Caquet, Th..
In this work, the effects of matrix interferences on the analytical performance of a new multiresidue method based on off-line solid phase extraction followed by reversed-phase liquid chromatographic separation and electrospray triple quadrupole mass spectrometric detection were investigated. This technique allows the simultaneous determination of 30 triazines, phenylureas and chloroacetanilides, extracted from freshwaters, in 40 minutes. Quantifications were performed with the use of appropriate internal standards (i.e. atrazine D5, diuron D6 and metolachlor D6). The limits of quantification were from 1 to 32 ng L-1 for the triazines, from 5 to 59 ng L-1 for the phenylureas and from 13 to 54 ng L-1 for the chloroacetanilides. The matrix effects were...

Microalgal sensitivity varies between a diuron-resistant strain and two wild strains when exposed to diuron and irgarol, alone and in mixtures ArchiMer
Dupraz, Valentin; Coquille, Nathalie; Menard, Dominique; Sussarellu, Rossana; Haugarreau, Larissa; Stachowski-haberkorn, Sabine.
A wild strain of Chaetoceros calcitrans and wild and diuron-resistant strains of Tetraselmis suecica, were exposed to the PSII inhibitor herbicides diuron and irgarol, individually and in mixtures. The effects of three concentrations of diuron and irgarol and four binary mixtures were evaluated on doubling time, relative reactive oxygen species and lipid content by flow cytometry, and on photosynthetic efficiency by pulse amplitude modulated fluorescence. In both wild strains, significant effects were observed for each molecule at the highest concentration tested: at irgarol 0.5 μg L−1, C. calcitrans was shown to be more sensitive than T. suecica (+52% and +19% in doubling time, respectively), whereas at diuron 5 μg L−1, T. suecica was more affected...

Influence de la matière organique dissoute naturelle et d’herbicides, seuls et combinés, sur des microalgues marines et d’eau douce ArchiMer
Coquillé, Nathalie.
Microalgae are at the basis of aquatic food webs and may be the indirect target of herbicides from agricultural and urban uses. They also interact with other compounds from their environment such as natural dissolved organic matter (DOM), which can itself interact with herbicides. This thesis aimed to study, in laboratory controlled conditions, the influence of natural DOM on herbicide toxicity to microalgae by using monospecific, non-axenic cultures. The experiments included: natural freshwater and marine DOM, three herbicides (irgarol, diuron and S-metolachlor) singly and in mixtures, two freshwater microalgae (Gomphonema gracile and Sphaerellopsis sp.) and two marine microalgae (Chaetoceros calcitrans and Tetraselmis suecica). Effects were evaluated on...

Assessment of the quantities of herbicides and nutrients brought down by the river Charente to the coast and modelling of the dispersion of atrazine in the Marennes-Oleron bay ArchiMer
Munaron, Dominique; Dubernet, Jf; Delmas, F; Stanisiere, Jean-yves; Scribe, P.
Our study aims at making progress the estimate of herbicide and nutrient inputs to the Marennes-Oleron bay which could influence summer oysters mortalities, and to bring, a better understanding of the impact on coastal ecosystems by modelling the dynamics of these compounds in the shellfish-farming area. The development of a strategy adapted to the river Charente and its estuary reveals that agricultural activity on the watershed area is omnipresent, as well on the level of the nitrogen as on the one of herbicides contributions. It is thus necessary to specify Charente fluxes, which is the main tributary of the Marennes-Oleron bay, in order to better characterize its impact on shell fish-farming activity. Hydrodynamic modelling will constitute an...

Environmental stress in the pacific oyster Crassostrea gigas during gametogenesis : role of herbicides? ArchiMer
Burgeot, Thierry; Quirion, Sophie; Quiniou, Francoise; Knoery, Joel; Soletchnik, Patrick; Gagnaire, Beatrice; Renault, Tristan; Osada, M.; Geret, F.; Communal, P. Y.; Jadas-hecart, A.; Gagne, F..
In the Atlantic French coast, significant mortalities were observed in oyster communities during gametogenesis. These deaths possibly result from the cumulative effects of various stressors such as seasonal temperature variations, pesticide application and the release of contaminants from the resuspension of sediments. A field survey, based on aquaculture conditions at 15 and 70 cm from the sediment bed with genetically-resistant (R) and -sensitive (S) lines, showed a stress condition. Significant weight loss was observed during May 14th through June 10th for both R and S lines. On one hand, the levels of the egg yolk vitellin in females were decreased at May 14th for both R and S lines. On the other hand, a significant increase in vitellin production was...

Persistence of atrazine impact on aneuploidy in the Pacific oyster, Crassostrea gigas ArchiMer
Bouilly, Karine; Mccombie, Helen; Leitao, Alexandra; Lapegue, Sylvie.
Aneuploidy is the alteration of the normal diploid chromosome number. In the Pacific oyster, Crassostrea gigas, hypodiploid aneuploid cells have regularly been reported as has a negative correlation between this phenomenon and growth and evidence for a genetic basis. We previously demonstrated a positive relationship between a pollutant, atrazine, and aneuploidy in Crassostrea gigas adults and juveniles. To evaluate the persistence of this impact, the present study focused on a sample of the same juveniles exposed to different atrazine treatments (0.01 mg/l which represents a peak value found in a polluted environment and 0.1 mg/l) for three and a half months and evaluated them for aneuploidy after another two and a half months in non polluted conditions....

Effects of twelve pesticides on larvae of oysters (Crassostrea gigas) and on two species of unicellular marine algae (Isochrysis galbana and Chaetoceros calcitrans) ArchiMer
His, Edouard; Seaman, Matthias.
The effects of seven herbicides, four insecticides and one molluscicide were tested at concentrations of up to 10 mg/l on larvae of oysters, Crassostrea gigas (9 days exposure), and on laboratory cultures of the algae Isochrysis galbana and Chaeloceros calcilrans (21 days exposure). Ali of the pesticides tested had significant toxic effects on at least one of the test organisms. The strongest effects were those of lindane and isoproturon on survival and growth of C.gigas larvae, and of isoproturon and carbetamide on growth of Isochrysis and Chaeloceros cultures. The log-probit model for the relationship between dosage and effect, and Haber's rule for the relationship between duration of exposure and effect, hardly ever applied to our data. In sorne cases...

Comportement géochimique des herbicides et de leurs produits de dégradation en milieu estuarien et marin côtier ArchiMer
Munschy, Catherine.
The presence and geochemical behaviour of herbicides were investigated in estuarine and marine waters of the French coasts (Seine, Charente, Gironde estuaries and Rhône delta). These organic contaminants were quantified by capillary Gas Chromatography (GC) using Nitrogen-Phosphorus Detecter and identified by GCIMS (Mass Spectrometry). A liquid-solid extraction method using XAD-2 resins was developed,  allowing the analysis of both chloro-triazines and their degradation products in the nanogram per litre (ng.l-1) detection range. Different types of chemicals were identified in the estuarine and coastal waters: triazine, amide and acetanilide herbicides. These compounds are mainly present in the water phase, with less than 1% associated with particulate...

Evaluation of the partial renewal of in situ phytoplankton microcosms and application to the impact assessment of bentazon and dimethenamid ArchiMer
De La Broise, Denis; Stachowski-haberkorn, Sabine.
Microcosms, each consisting of 2 L natural surface seawater maintained in 2.3-L glass bottles, were immersed at a depth of 6 metres. The renewal of ten percent of microcosm volumes was carried out every other day. Phytoplankton-containing seawater was used for renewal (previously filtered through 25-, 50- or 200-μm cut-off). Phytoplankton community pigment analysis (by HPLC) and flow cytometry analysis were performed. After 13 days, data exhibited phytoplankton characteristics in microcosms in the same range as that of the natural surrounding sea water over the same period. Furthermore, in these microcosms, a negative correlation was observed between the filtration cut-off used for renewal water, and the eukaryote cell count. Herbicides were tested as...

Decision criteria for optimizing postemergence johnsongrass control in soybean crops in Argentina PAB
Tuesca,Daniel; Puricelli,Eduardo; Nisensohn,Luisa; Faccini,Delma; Papa,Juan.
Field studies were established in Zavalla and Oliveros, Argentina, during four years in order to optimize Johnsongrass (Sorghum halepense (L.) Pers.) chemical control by means of the thermal calendar model in comparison with other criteria (weed height or days after sowing). The effect of three application dates of postemergence herbicides was determined by visual control, density of tillers originated from rhizome bud regrowth, and from crown and shoot bud regrowth, and soybean yield. Following the thermal calendar model criterion, applications during the second date afforded the best control. Weed height for the first date showed little variability between experiments but was highly variable in the second and third application dates, achieving in some...
