Feasibility study of organic matter and Ammonium removal using loofa sponge as a supporting medium in an aerated submerged fixed-film reactor (ASFFR) Electron. J. Biotechnol.
Nabizadeh,Ramin; Naddafi,Kazem; Mesdaghinia,Alireza; Nafez,Amir Hosien.
Biofilm systems are efficient in the removal of organic matter and ammonium from wastewaters. In this study, loofa sponge, a natural product, was used as a supporting medium in an aerated submerged fixed-film reactor to evaluate its performance in removing organic matter and nitrogen from wastewater. Four pilot runs were performed with chemical oxygen demand (COD) concentrations of 100, 200, 300 and 400 mg l-1 to provide an organic loading rate of 0.6, 1.2, 1.8, and 2.4 kg m-3d-1 respectively. In these pilot runs, the influent ammonium nitrogen concentrations were justified to 5, 10, 15 and 20 mg l-1 as N to provide an influent nitrogen loading of 30, 60, 90 and 120 g m-3.d-1 respectively. Although soluble COD removal efficiency greater than 80 percent was...

Mahendhiran,Muhilan; Ramirez-Prado,Jorge Humberto; Escobedo-Gracia Medrano,Rosa Maria; Canto-Canché,Blondy; Tzec-Simá,Miguel; Grijalva-Arango,Rosa; James-Kay,Andrew.
Background ADP-glucose pyrophosphorylase (AGPase) is a rate-limiting enzyme catalyzing the first step in the starch biosynthesis pathway in higher plants. To date, there are no reported variants or isoforms of the AGPase enzyme in bananas (Musa spp. family Musaceae) as is the case of other plants. In this study, genomic DNA sequences homologous to the gene encoding one of the large subunits of the enzyme were amplified from 10 accessions of the genus Musa, including representatives of wild ancestors (AA and BB genomes), dessert bananas (AA, AAA, AB and AAB genomes), plantains (AAB genome) and cooking bananas (ABB and AAA genomes), and studied in order to find single nucleotide polymorphisms (SNP) base variations in Musa accessions. Results In the 810-base...

Genetic diversity of breeding popcorn lines determined by SSR markers Electron. J. Biotechnol.
Ribeiro Trindade,Ana Paula; Barth Pinto,Ronald José; Amaral Júnior,Antonio Teixeira do; Mangolin,Claudete Aparecida; Silva Machado,Maria de Fátima Pires da; Scapim,Carlos Alberto.
Information about genetic dissimilarity is very important to corroborate genealogical relationships and to predict the most heterozygotic hybrid combinations. Eight popcorn S6 lines of diverse germplasm types were evaluated using simple sequence repeats (SSR) markers. Of a total of 51 evaluated polymorphic primers, 15 were used for polymerase chain reaction (PCR) amplification. The genetic distance was estimated by Rogers’ modified distance. The different popcorn breeding programs in Brazil are possibly using highly similar base-populations. The genetic similarity of lines P1-3 and P8-1 was lowest, while P3-3 and P8-2 were genetically more similar. The cophenetic correlation showed that the Unweighted Pair-Group Method Using Arithmetic Averages...

Cinnamic acid, ethanol and temperature interaction on coumarate decarboxylase activity and the relative expression of the putative cd gene in D. bruxellensis Electron. J. Biotechnol.
Ganga,María Angélica; Salinas,Francisco; Ravanal,Cristina; García,Verónica; Carrasco,Carolina; Martínez,Claudio; Saavedra,Jorge.
Dekkera bruxellensis is one of the main contaminating yeasts in wine due to its ability to metabolize cinnamic acids into volatile phenols. This yeast metabolizes p-coumaric acid into 4-vinylphenol through a coumarate decarboxylase (CD) and then transforms it into to 4-ethylphenol (EF) through a vinylphenol reductase. In this work we investigated the influence of the interaction between the concentration of p-coumaric acid, ferulic acid and ethanol as well as growth temperature on the production of CD activity and the expression of a putative gene that codes for this enzymatic activity. For this, a Box Behnken experimental design was used. The concentration of p-coumaric acid (5-26 ppm) and ferulic acid (3-9 ppm) alone did not show any significant effect...

Using a neural network to backtranslate amino acid sequences Electron. J. Biotechnol.
White,Gilbert; Seffens,William.
A neural network (NN) was trained on amino and nucleic acid sequences to test the NN’s ability to predict a nucleic acid sequence given only an amino acid sequence. A multi-layer backpropagation network of one hidden layer with 5 to 9 neurons was used. Different network configurations were used with varying numbers of input neurons to represent amino acids, while a constant representation was used for the output layer representing nucleic acids. In the best-trained network, 93% of the overall bases, 85% of the degenerate bases, and 100% of the fixed bases were correctly predicted from randomly selected test sequences. The training set was composed of 60 human sequences in a window of 10 to 25 codons at the coding sequence start site. Different NN...

Mechanisms and roles of the RNA-based gene silencing Electron. J. Biotechnol.
Jana,Snehasis; Chakraborty,Chiranjib; Nandi,Shyamsundar.
RNA silencing is a remarkable type of gene regulation. This process has been found to occur in many different organisms such as plants (co-suppression), fungi (quelling), and animals (RNA interference; RNAi). Double-stranded RNA (dsRNA) is a potent trigger in RNA silencing mechanisms operating in a wide range of organisms. This mechanism recognizes dsRNA and processes them into small 21-25nt RNAs (smRNAs). Small RNAs can guide post-transcriptional degradation of complementary messenger RNAs and in plants, transcriptional gene silencing is occurred by methylation of homologous DNA sequences. In plants, it serves as an antiviral defense, and many plant viruses encode suppressors of silencing such as helper component-proteinase of potyviruses (HC-Pro) and the...

Biofilm formation for organic matter and sulphate removal in gas-lift reactors Electron. J. Biotechnol.
Pizarro,Carolina; Donoso-Bravo,Andrés; Jeison,David; Ruiz-Filippi,Gonzalo; Chamy,Rolando.
A start-up strategy was presented and evaluated to obtain a well-established biofilm in a gas lift-reactor capable both for the removal of organic matter and sulphate. Pumice stone was used as material support. The influence of shear forces, given by the biogas recirculation, the effect of the COD/SO4-2 ratio and the OLRs increase were evaluated on the reactor performance. From the first stages, cell colonization was observed along with the presence of extracellular polymeric substances. The COD and sulphate removal was over 70%, for all conditions. The increase of gas flow did not have an adverse effect on biofilm development even though there was some detachment. Specific methanogenic activity of the biofilm increased along the experiments. Operational...

Intein-mediated expression of cecropin in Escherichia coli Electron. J. Biotechnol.
Díaz,Mauricio; Venturini,Elena; Marchetti,Stefano; Arenas,Gloria; Marshall,Sergio H.
Different strategies have been used to overcome the difficulties to produce antimicrobial peptides. Here we used Intein Mediated Purification with an Affinity Chitin-binding Tag (IMPACT-System, New England Biolabs) for the expression of the antimicrobial peptide cecropin to reduce its sensitivity to intracellular proteases and use its inducible self-cleaving capability to remove the carrier. Cecropin was cloned into suitable expression vector pTYB11, and expression induced by IPTG in Escherichia coli ER2566. The use of 22ºC induction allowed the expression of cecropin with its intein carrier in soluble form. Cell extracts were purified by chitin affinity chromatography and intein-mediated splicing of the target protein was achieved by thiol addition,...
