Abnormal meiotic behavior in three species of Crotalaria PAB

The objective of this work was to compare the meiotic behavior and pollen grain viability of three species of Crotalaria. Slides for meiotic analysis were prepared by the air-drying technique. Pollen grain viability was measured by three staining procedures (Alexander's solution, tetrazolium chloride and fluorescein diacetate) and in vitro germination in a sucrose solution. Eight bivalents were observed, confirming previous reports on populations from other regions of Brazil, as well as from other countries. Breeding programs for Brachiaria spp. use both intraspecies and interspecies crosses between sexual and apomictic plants in order to obtain new cultivars with the desired characteristics. As there are different ploidy levels both within and between species of this genus, it becomes necessary to evaluate the genotypes used in breeding programs, as a guide to breeders when adopting crossing strategies. In this work, DNA content and chromosome number were determined in order to characterise ploidy levels in Brachiaria spp. genotypes. Cytogenetic studies in Heteroptera contribute to the analysis of evolutionary trends within the group. Heteroptera are characterized by the possession of holokinetic chromosomes, different sex chromosome mechanisms and a pair of m chromosomes in some species. In the present work, the male karyotype and meiosis in Sigara denseconscripta (Breddin), S. chrostowskii Jaczewski, and S. rubyae (Hungerford) are described. Meiotic behavior of interspecific hybrids between artificially tetraploidized sexual Brachiaria ruziziensis and tetraploid apomictic B. brizantha (Poaceae) Scientia Agricola

The meiotic behavior of four interspecific promising hybrids was evaluated by conventional cytological methods. The female genitors were two artificially tetraploidized sexual accessions of B. ruziziensis (R41 and R44, 2n = 4χ = 36), which were crossed to an agronomically superior natural tetraploid apomictic genotype of B. brizantha (B140 - BRA003395). Three of them (HBGC313, HBGC 315, and HBGC324) were sexual and one (HBGC325) apomictic. Cytogenetic analysis of the Amazon stingless bee Melipona seminigra merrillae reveals different chromosome number for the genus Scientia Agricola

Cytogenetic analysis of the Amazon stingless bee Melipona seminigra merrillae, by conventional Giemsa staining and C-banding, revealed a different chromosome number for Melipona: 2n = 22 for females and diploid drones while the haploid drones present n = 11. There is no evidence of B chromosomes. This result contrasts with previous studies, in which the chromosome number of 19 Melipona species was determined as 2n = 18 for females and n = 9 for haploid males. Based on cytogenetic information available for other Melipona species, we propose that M. s. merrillae has a more derived diploid number. Microsatellite diversity and chromosome number in natural populations of Trifolium riograndense Burkart Scientia Agricola

Twenty eight natural populations of Trifolium riograndense Burkart, an important forage legume from native pastures of the state of Rio Grande do Sul, Brazil, were evaluated for genetic diversity with eight Simple Sequences Repeats (SSR) markers. Chromosome numbers were also determined. The eight markers were polymorphic, with 35 alleles and an average of 4.37 alleles per locus, and Polymorphism Information Content (PIC) between 0.48 and 0.80. Group analysis based on Jaccard´s similarity coefficient separated the 28 accessions in nine groups, with an average genetic similarity of 0.44, indicating a high genetic variability among the populations. ABSTRACT Although karyotype features are useful data for evolutionary studies, cytogenetic data in Bambusoideae are mainly based only on chromosome counts. The first comparative cytogenetic analysis of three Neotropical woody bamboo species - Guadua chacoensis, G. angustifolia and Chusquea tenella - was undertaken based on new and reviewed chromosome counts, CMA/DAPI double staining, fluorescent in situ hybridization (FISH) with 35S and 5S rDNA probes, and genome size estimation. The two species of Guadua were found to have 2n = 46 chromosomes, while the first record for C. tenella was 2n = 44. Karyotypic characterization of Capsicum sp. accessions Agronomy

This study aimed to determine the chromosome number and the karyotype of Capsicum annuum, Capsicum chinense, Capsicum frutencens and Capsicum baccatum accessions in the active Capsicum sp. genebank at the Federal University of Piauí (BGC-UFPI). These species have great economic importance throughout the world, and their cytogenetic characterization can inform taxonomy and lead to improvement in the genus. Avaliou-se o complemento cromossômico de 16 acessos de Pennisetum disponíveis na Coleção de Germoplasma da Embrapa Gado de Leite – Juiz de Fora, Estado de Minas Gerais. Para a avaliação citogenética, utilizou-se a técnica de esmagamento e coloração com Feulgen. Para quatro acessos foram obtidas metáfases com 2n = 28 cromossomos, confirmando sua classificação como P. purpureum. A origem híbrida P. purpureum x P. glaucum foi constatada em oito acessos, os quais apresentaram 2n = 21. The mercury rejected in the water system, from mining operations and lixiviation of soils after deforestation, is considered to be the main contributors to the contamination of the ecosystem in the Amazon Basin. The objectives of the present study were to examine cytogenetic functions in peripheral lymphocytes within a population living on the banks of the Tapajós River with respect to methylmercury (MeHg) contamination, using hair mercury as a biological indicator of exposure. Karyotypic analysis in species of the genus Dasyprocta (Rodentia: Dasyproctidae) found in Brazilian Amazon Anais da ABC (AABC)

A total of 30 animals of the genus Dasyprocta were cytogenetically studied. They belong to the following species: D. prymnolopha (N=20), D. leporina (N=6), D. fuliginosa (N=1) and Dasyprocta sp. (N=3) (Dasyproctidae, Hystricognathi). Cell suspensions were obtained by peripheral blood culture, besides bone marrow and spleen cells, from D. prymnolopha and D. leporina. The diploid number was 64/65 for all samples. The karyotypes showed similarity, and chromosomal polymorphism was not detected by Giemsa conventional staining and G banding. Euglossini are solitary bees considered important pollinators of many orchid species. Information regarding chromosome organization is available for only a small number of species in this group. In the present work, the species Euglossa townsendi and E. carolina were analyzed by cytogenetic techniques to collect information that may aid the understanding of their evolution and chromosomal organization. In Bromeliaceae, cytogenetic and flow cytometry analyses have been performed to clarify systematic and evolutionary aspects. Karyotyping approaches have shown the relatively high chromosome number, similar morphology and small size of the chromosomes. These facts have prevented a correct chromosome counting and characterization. Authors have established a basic chromosome number of x = 25 for Bromeliaceae. Recently, one karyomorphological analysis revealed that x = 25 is no longer the basic chromosome number, whose genome may have a polyploid origin. We investigated the NOR distribution in ten populations of Physalaemus cuvieri from different regions of Brazil and Argentina. A high variability in NOR pattern was observed and provided a useful tool in grouping several populations. The specimens from the state of Tocantins, northern Brazil, could easily be distinguished from all the other analyzed populations, since its karyotype presented NORs in the chromosome pairs 1,3,4 and 10 (and sometimes also in chromosome 5), and several pericentromeric C-bands. Cytogenetics of the freshwater cyclopoid mesocyclops longisetus longisetus (crustacea, copepoda) from São Carlos, São Paulo, Brazil Biota Neotropica

A cytogenetic study was carried out on the early embryonic stages of the cyclopoid species Mesocyclops longisetus longisetus (Crustacea, Copepoda) from freshwater environments of the São Carlos area of south-east Brazil. Chromatin was staining using acetic orcein solution. The species showed 2n = 14 chromosomes and presented chromatin diminution in the 4th embryonic cleavage. The eliminated chromatin was concentrated in the equatorial region of the embryonic cell. The results were compared with data for other cyclopoid species of temperate regions.
