Carbohydrates on the growth of Cattleya schilleriana Reichb.f. seedlings (Orchidaceae) Ciência Rural
Galdiano Júnior,Renato Fernandes; Mantovani,Cibele; Lemos,Eliana Gertrudes de Macedo.
ABSTRACT: The aim of the present study was to evaluate the effects of carbohydrate supplementation on the propagation of the orchid Cattleya schilleriana. The 120-d-old seedlings were subcultured in fructose-, glucose-, or sucrose-supplemented (0, 15, 30, and 45g L-1) ½ MS culture medium (half-strength macronutrient concentrations), using a completely random design with four repetitions per treatment. After 120d of treatment, root number and length, leaf number and length, and fresh weight were evaluated, and seedling survival was evaluated after 75d of acclimatization in a greenhouse. The in vitro growth data were submitted to regression analysis, whereas the percentage survival data were analyzed using ANOVA and Tukey’s test. Both in vitro growth and ex...

Micropropagation of the new apple rootstock ‘G. 814’ Ciência Rural
Meneguzzi,Aline; Gonçalves,Mayra Juline; Camargo,Samila Silva; Grimaldi,Fernanda; Weber,Gabriela Candido; Rufato,Leo.
ABSTRACT: International breeding programs launched new genetic material of apple rootstocks that in addition to precocity and great yield are resistant to major diseases and soil pests encountered in the largest apple producing regions in Brazil. Given this, there is a necessity for vegetative propagation of these materials for study and possible replacement of existing rootstocks. The objective was to adapt a micropropagation protocol for new apple rootstock ‘G. 814’. In the multiplication phase were evaluated BAP concentrations: 0; 0.5; 1; 2 and 4mg L-1 and in the rooting phase were evaluated IBA concentrations: 0; 0.25; 0.50; 1; 1.5 and 2.5mg L-1. These new results demonstrated that this new rootstock selection can be propagated with this tissue culture...

Anatomical and histochemical aspects of zigotic embryo and leaves in 'Coqueiro Anão' Ciência Rural
Santana,Marlucia Cruz de; Souza,Margarete Magalhães; Pereira,Telma Nair Santana; Teixeira,Sílvio Lopes.
The paper provides information about histochemical staining reactions in leaves and embryos of 'Coqueiro Anão' (Cocos nucifera). It was compared in vitro coconut and autotrophic palm leaves. Reactions for insoluble polysaccharides and acidic compounds, protein, extractable lipids, lignin and other classes of compounds were tested using histochemical tests. None sample gave positive reaction for lignin and phenolic compounds. All the samples gave positive reaction for protein, starch and insoluble polysaccharides while acidic compounds were positive only in in vitro leaves. Both in vitro and autotrophic leaves gave positive reaction for lipids showing presence of cuticle even in in vitro leaves. Only autotrophic palm leaves showed idioblasts containing...

Towards the micropropagation of Euphorbia cyathophora Murray: a wild plant species with medicinal and ornamental potential Ciência Rural
Rodríguez-Elizalde,María de los Ángeles; Ferrera-Cerrato,Ronald; Hernández,Mateo Vargas; León,María Teresa Beryl Colinas y; González,Alejandro Manzo; Alarcón,Alejandro.
ABSTRACT: Euphorbia cyathophora Murray is a plant species with medicinal and ornamental potential but whose in vitro propagation is unknown. Two experiments were performed to know 1) the effect of light (WL) at 25±2 °C and 20 μmol m-2 s-1 luminous intensity, or darkness at 20±2 °C, for the first 20 days and subsequent placement in light (DKL) as previously described, and 2) the best combination of plant growth regulators: benzyl amino purine (BAP), kynetin (KIN), 2-isopentenyl adenine (2iP) and thidiazuron (TDZ) in combination with naphthaleneacetic acid (NAA) and the effect of both light conditions. The DKL treatment did not promote the growth of leaves, cotyledons, or plantlet height; in contrast, WL produced a more developed root system with early...

Optimization of jenipapo in vitro seed germination process Ciência e Agrotecnologia
Souza,Rafaela Ribeiro de; Paiva,Patrícia Duarte de Oliveira; Silva,Raphael Reis da; Reis,Michele Valquíria dos; Nery,Fernanda Carlota; Paiva,Renato.
ABSTRACT The in vitro seed germination is an effective alternative for quickly obtaining explants with sanitary quality. However, jenipapo seeds present slow and uneven germination. Therefore, internal and external factors to seed which directly interfere in the process, they must be identified, in order to adapt better techniques to obtain seedlings. In this sense, this work aimed to optimize the in vitro germination of Genipa americana L. seeds by evaluating different factors (light quality, GA3 treatment, pre-soaking in distilled water, growing media and stratification in the dark). It was found that the seed germination of G. americana was indifferent to light, however, the best results were obtained under conditions of continuous darkness; There was...

In vitro propagation and acclimatization of genipapo accessions Ciência e Agrotecnologia
Sa,Francielen Paola de; Ledo,Ana da Silva; Amorim,Julie Anne Espindola; Silva,Ana Veruska Cruz da; Pasqual,Moacir.
ABSTRACT Genipapo is a species with an intermediate seed storage behavior, and its germination is slow and asynchronous; therefore the tissue culture is an alternative strategy for the propagation in large-scale. This study aimed to evaluate three genipapo accessions at different micropropagation stages (adventitious shoot induction, in vitro rooting and acclimatization). Genipapo nodal segments from the Sabinópolis (AS), Cruz das Almas (CZA) and Núcleo Bandeirante (NB) accessions were used and inoculated in culture media with 0 and 1.0 mg L-1 6-benzylaminopurine (BAP). Different indole-3-butyric acid (IBA) concentrations were tested for in vitro rooting. Subsequently, the effect of two substrates on the seedling development of three genipapo accessions...
