Dunaliella tertiolecta (DT) was chemically investigated to isolate molecules inhibiting cancer cell proliferation and inducing apoptosis in vitro. The potency to inhibit cell growth was used for the bio-guided fractionation and isolation of active compounds using chromatographic techniques. The DT dichloromethane extract exhibited a strong anti-proliferative activity on MCF-7 and LNCaP cells, and was further fractionated and sub-fractionated by RP-HPLC. High resolution mass spectrometry and spectrophotometric analysis unequivocally identified violaxanthin as the most antiproliferative molecule present in DT DCM extract. Violaxanthin purified from DT induced MCF-7 dose- dependent growth inhibition in continuous and discontinuous treatments, at... |

Industrial potentialities related to biotechnology of microalgae are examined, highlighting a wide spectrum of applications which concern principally nutrition, health, environment and energy. The french situation of research on microalgae, especially at Ifremer, is assessed, showing that the main axis are fine chemistry, aquaculture, environment, bio-process engineering. It covers the majority of the applications, but leaves in shade certain important aspects such as human food and CO2 fixation. Concerning the general approach, some significant improvements can be expected from a more accurate control of bacteriological and genetic quality of cultures. About the industrial aspects, a national initiative seems in progress. The variety of potential... |

Acantharia (Radiolaria) are widespread and abundant heterotrophic marine protists, some of which can host endosymbiotic eukaryotic microalgae. Although this photosymbiotic association was first described at the end of the 19th century, the diversity of the symbiotic microalgae remains poorly characterized. Here, we examined the identity of the microalgae associated with the acantharian species Acanthochiasma sp. by sequencing partial 18S and internal transcribed spacer (ITS) ribosomal DNA genes from cultured symbionts and directly from isolated holobiont specimens. Single Acanthochiasma cells contained multiple symbiotic partners, including distantly related dinoflagellates (Heterocapsa sp., Pelagodinium sp., Azadinium sp. and Scrippsiella sp.) as well as... |

Melanoma is an aggressive tumor with invasive and metastatic potential, frequently exhibiting multidrug resistance mechanisms. In our continuous search for antimelanoma molecules, we have identified some effective marine compounds capable of not only inducing cell death, but also of sensitizing chemoresistant tumor cells to clinically used anticancer drugs. In this report, the cryptophyte Rhodomonas salina (Wislouch) D.R.A.Hill & R.Wetherbee, Pyrenomonadaceae, was chemically investigated in order to identify pigments efficiently inhibiting melanoma cells proliferation. All pharmacological tests were performed on A2058 cells expressing the oncogenic BRAF V600E mutation and resistant to dacarbazine treatment. Flash chromatography of R. salina ethanol... |

Due to the growing concern about the presence of microplastics (MP) in the environment, the number of studies evaluating the toxicity of these small persistent particles on different marine species has increased in recent years. Few studies have addressed their impact on marine phytoplankton, a subject of great concern since they are primary producers of the aquatic food web. The aim of this study is to unravel the cytotoxicity of 2.5 μg mL−1 unlabelled amino-modified polystyrene beads of different sizes (0.5 and 2 μm) on the marine diatom Chaetoceros neogracile. In addition to traditional growth and photosynthesis endpoints, several physiological and biochemical parameters were monitored every 24 h in C. neogracile cells by flow cytometry during their... |

The number of microalgae of high nutritional value actually available to mollusc hatcheries is limited to a few species and this represents a constraint for the industry. Fifteen microalgal species belonging to seven different taxonomic classes were selected from the Algobank-Caen microalgal culture collection of the University of Caen. An initial screening of these microalgae, based on their cytomorphological characteristics and growth performances, led to the selection of four species (Imantonia rotunda, Emiliania huxleyi, Pseudoisochrysis paradoxa and Diacronema vlkianum), which were then tested as food for Crassostrea gigas larvae. Two species (L rotunda and E huxleyi) were of poor food value for oyster larvae, while P paradoxa and D. vlkianum resulted... |

Microalgae, which are the foundation of aquatic food webs, may be the indirect target of herbicides used for agricultural and urban applications. Microalgae also interact with other compounds from their environment, such as natural dissolved organic matter (DOM), which can itself interact with herbicides. This study aimed to evaluate the influence of natural DOM on the toxicity of three herbicides (diuron, irgarol and S-metolachlor), singly and in ternary mixtures, to two marine microalgae, Chaetoceros calcitrans and Tetraselmis suecica, in monospecific, non-axenic cultures. Effects on growth, photosynthetic efficiency (Ф’M) and relative lipid content were evaluated. The chemical environment (herbicide and nutrient concentrations, dissolved organic carbon... |

In this paper, a simple detection of a toxic algae, Alexandrium minutum, was developed using highly sensitive quartz crystal microbalance. In terms of performance, compared with other conventional analytical tools, the main interest of our immunosensor is based on a fast and direct detection of these living cells. This system requires the use of one monoclonal antibody directed against the surface antigen of A. minutum. We demonstrate that the whole living and motile algae are caught and detected. The high specificity of the biosensor is also demonstrated by testing several other dinoflagellate species. The frequency shift is correlated to the A. minutum cells concentration. This simple system is potentially promising for environmental monitoring purposes. |

The periodically forced light-limited Droop model represents microalgae growth under co-limitation by light and a single substrate, accounting for periodic fluctuations of factors such as light and temperature. In this paper, we describe the global dynamics of this model, considering general monotone growth and uptake rate functions. Our main result gives necessary and sufficient conditions for the existence of a positive periodic solution (i.e. a periodic solution characterized by the presence of microalgae) which is globally attractive. In our approach, we reduce the model to a cooperative planar periodic system. Using results on periodic Kolmogorov equations and on monotone sub-homogeneous dynamical systems, we describe the global dynamics of the... |

Given their key roles in almost all ecosystems and in several industries, understanding and predicting microorganism growth is of utmost importance. In compliance with evolutionary principles, coarse-grained or genome-scale models of microbial growth can be used to determine optimal resource allocation scheme under dynamic environmental conditions. Resource allocation approaches have given important qualitative results, but it still remains a gap towards quantitiative predictions. The first step in this direction is parameter calibration with experimental data. But fitting these models results in a bi-level optimization problem, whose numerical resolution involves complex optimization issues. As a case study, we present here a coarse-grained model... |

The search for novel compounds of marine origin has increased in the last decades for their application in various areas such as pharmaceutical, human or animal nutrition, cosmetics or bioenergy. In this context of blue technology development, microalgae are of particular interest due to their immense biodiversity and their relatively simple growth needs. In this review, we discuss about the promising use of microalgae and microalgal compounds as sources of natural antibiotics against human pathogens but also about their potential to limit microbial infections in aquaculture. An alternative to conventional antibiotics is needed as the microbial resistance to these drugs is increasing in humans and animals. Furthermore, using natural antibiotics for... |

Integrated multi-trophic aquaculture (IMTA) systems are a promising solution for sustainable aquaculture combining nutrient recycling with increased biomass production. An innovative land-based recirculating aquaculture system (RAS) was studied in France for a 60-day experiment. It combined a European sea bass (Dicentrarchus labrax) RAS with two other production systems: high rate algal ponds (HRAP) with natural marine polyspecific algal assemblages, and oysters in separate open tanks. The objective was the assessment of: 1) the efficiency and the stability of the microalgae bioremediation of the effluent from a fish RAS in spring and summer, 2) the abundance and the diversity patterns of the microalgae biomass for consumption in the oyster compartment of... |

To investigate the ability of microalgae to develop stable, long-term resistance to herbicides, the marine microalga Tetraselmis suecica was exposed to the herbicide diuron (5 µg/L) for a 43-generation exposure period followed by a 12-generation depuration phase. During the first 25 generations, diuron-exposed cultures showed doubling times ranging from 1.95 to 2.6 days, which was 2 to 2.5-fold longer than control cultures. Between generations 25 and 38, during diuron exposure, two out of the three exposed cultures exhibited a spontaneous drop in doubling time. These results provided evidence of culture adaptation to diuron. To assess persistence of the diuron adaptation observed on growth performance, one of the adapted cultures (D3) was maintained for 12... |
