Effect of soil interacting herbicides on soybean nodulation in Balcarce, Argentina PAB

Two trials were performed in Balcarce, Argentina (37° 45' LS; 58° 18' LW) during 1993-94, to assess the effect of eight herbicides applied individually or in tank mixtures, on nodule number, nodule dry weight, seed yield and N percent in seed in soybean Asgrow 3205, inoculated with Bradyrhizobium japonicum CB 1809. Differences in ureide and amino acid content of water stressed soybean inoculated with Bradyrhizobium japonicum and B. elkanii PAB

The objective of this work was to study the response to water stress of a drought sensitive soybean cultivar inoculated with Bradyrhizobium japonicum (strain CB1809, Semia 586) and B. elkanii (strain 29W, Semia 5019). CB1809 nodulated plants produced a significantly higher root fraction (19%) than 29W (14.6%). Plants inoculated with CB1809 produced less nodules and accumulated more nitrogen than those inoculated with 29W. In general, low amounts of ureides in nodules were found in watered plants inoculated with either CB1809 or 29W strains, but those levels were five-fold increased in stressed plants inoculated with CB1809. Residual effects of successive exposure of soybean Bradyrhizobium strains to aluminium on solid defined medium PAB

The aim of these studies was to investigate whether residual toxic effects of exposing soybean root nodule bacteria to Al in a solid defined media (SDM) alter tolerance to Al, survival, sensitivity to antibiotics, N2 fixation effectiveness and genetic diversity of Bradyrhizobium strains. After being exposed four times to Al, strains showed variation in Al tolerance but there was no evidence of change in their original Al tolerance, sensitivity to the antibiotics or genetic diversity. Exposure of Bradyrhizobium strains to SDM plus Al did not alter biological N2 fixation effectiveness of five strains. Lotus corniculatus, L. tenuis, L. pedunculatus, and L. subbiflorus inoculated with Mesorhizobium loti NZP2037 strain were grown in a growth chamber. The plants dry weight (DW), the nodule fresh weight (FW), the nitrogenase activity, the nodule glutamine synthetase (GS) and glutamate synthase (GOGAT) activities, as well as the leghemoglobin content and the amino acid in the stem were measured 28 days after inoculation. The highest DW of plants was measured in L. tenuis and the highest FW of nodules was measured in L. pedunculatus. Rhizobacteria were isolated from root and rhizospheric soil of rice collected from rice paddy fields in Nakhon Pathom and Sukhothai provinces. One hundred and sixteen isolates were screened for plant growth promoting properties. Nitrogen fixation property determined based on PCR amplification using primer Salkowski’s reagent and quantified by spectrophotometer at 530 nm. Siderophore production and phosphate solubilization were tested by plating technique in chrome azurol S and Pikovskaya’s medium. Eighty-one percent of isolated rhizobacteria revealed plant growth promoting properties. Genetic linkage map and mapping of the locus of biological nitrogen fixation inefficiency in cowpea Agronomy

The objectives of the present study were to construct a cowpea genetic map using the F2 population resulting from the cross IC-1 x BRS Marataoã, based on single nucleotide polymorphism (SNP) markers, and to map the cpi gene, with additional reference to introgression with the consensus map of species, aiming to identify markers for assisted selection to develop more efficient cultivars for BNF. The parents and 89 F2 plants were genotyped with 51,128 SNP markers, of which 910 polymorphic markers were used to construct the map. A fixação biológica do nitrogênio é de fundamental importância na sustentabilidade agrícola, e a caracterização fenotípica rizobial contribui para o conhecimento da sua diversidade em coleções e para a utilização posterior em sistemas agrícolas. O objetivo deste trabalho foi avaliar a diversidade fenotípica de rizóbio isolados de solos da Amazônia e suas eficiências simbióticas quando inoculados em feijão caupi (Vigna unguiculata L. Walp.). Os isolados nativos apresentaram diversidade quanto às características morfológicas e fisiológicas. Foi verificado que a inoculação com isolados de rizóbio nativos contribuiu, de forma significativa, para o aumento da biomassa, nodulação de caupi e em eficiência simbiótica.

Azospirillum sp . inoculation in wheat, barley and oats seeds greenhouse experiments BABT

The aim of the present work was to evaluate the inoculation effects of Azospirillum sp. RAM-7 strain, in seeds of wheat, barley and oats cultures on yield and total nitrogen content of the grains in greenhouse experiments. Seeds inoculation with Azospirillum sp. RAM-7 strain enhanced grain yields, even so, the responses vary among the evaluated cultures. For wheat, significant increases were obtained when the inoculation was associated to 100% of the recommended nitrogen, although the treatment that received only inoculation produced 7.4% above the control, however this difference wasn't significant. Growth of Crotalaria juncea L. supplied with mineral nitrogen BABT

Plants of Crotalaria juncea inoculated with Rhizobium were treated with nutrient solution containing 10 or 20mg of either N/NO3 or N/NH4.plant-1.week-1 . The control plants received nutrient solution without N. An investigation was conducted on the effect of these sources of N on growth and nitrogen fixation of plants with 30, 60 and 90 days after sowing (DAS). Those that received mineral N presented higher growth than -N plants, but the presence of nodules occurred in all the treatments. Plants treated with NH4 presented higher N content until 60 days. The highest concentrations of leghemoglobin and protein in nodules were found at 30 DAS and there was no difference in leghemoglobin content between treatments for any age and in protein from 60 DAS....

The A. brasilense transcription regulator NifA, which seems to be constitutively expressed, activates the transcription of nitrogen fixation genes. It has been suggested that the nitrogen status-signaling protein GlnB regulates NifA activity by direct interaction with the NifA N-terminal GAF domain, preventing the inhibitory effect of this domain under conditions of nitrogen fixation. In this organism, the regulation of nitrogen fixation by ammonium ions involves several proteins including the uridylyltransferase/uridylyl-removing enzyme, GlnD, which reversibly uridylylates the two PII proteins, GlnB and GlnZ, in response to the concentration of ammonium ions. In the present study, the uridylylation/deuridylylation cycle of A. brasilense GlnB and GlnZ proteins by GlnD was reconstituted in vitro using the purified proteins. Application of representational difference analysis to identify genomic differences between Bradyrhizobium elkanii and B. Japonicum species BJM

Bradyrhizobium elkanii is successfully used in the formulation of commercial inoculants and, together with B. japonicum, it fully supplies the plant nitrogen demands. Despite the similarity between B. japonicum and B. elkanii species, several works demonstrated genetic and physiological differences between them. In this work Representational Difference Analysis (RDA) was used for genomic comparison between B. elkanii SEMIA 587, a crop inoculant strain, and B. japonicum USDA 110, a reference strain. Two hundred sequences were obtained. From these, 46 sequences belonged exclusively to the genome of B. elkanii strain, and 154 showed similarity to sequences from B. japonicum genome.
