The thesis work is part of the very current issue on mass mortality outbreaks affecting Pacific oyster, C. gigas, spat and juveniles and questions about the involvement of pesticides in this phenomenon. The first part of this thesis was devoted to study the effects of pesticides on hemocyte parameters in the Pacific oyster (in vitro and in vivo). The immunomodulator effect of selected pesticides (alone or in mixture) has been explored principally through flow cytometry. Shellfish industry is mostly realized in estuary zones, which are subjected to pollutions due to anthropic activities. The harmful effects of pollutants on animals inhabiting these estuarine zones are poorly known. We investigated the effects of genotoxicant exposure in gametes and embryos to find a possible link between genotoxicity and reproduction/developmental impairment, and explore the impact of chemical genotoxicity on population dynamics. Our study focused on the genotoxic effects of two herbicides on oyster gametes and embryos: glyphosate (both as an active substance and in the Roundup formulation) and diuron. Bay of Marennes-Oleron is an important French area of reproduction for sea bass, Dicentrarchus labrax. This area is often subjected to many pollutions due to anthropic activities, including pesticides and polycyclic aromatic hydrocarbons (PAH) which cause adverse effects in aquatic organisms. The harmful effects of such contaminants on fish immune system leading to a decreased resistance against pathogens begin to be described, but remain poorly known. The release of pollutants into the environment, either during normal operations or due to accidental discharges, has focussed attention on the fate and effects of xenobiotic compounds in the marine environment. This interest has been heightened by the development of the off-shore industry with the subsequent discharge of biocides from platforms. The environmental effect and fate of several agricultural and industrial source toxicants were evaluated in controlled laboratory microcosm systems in the presence of the aminopolysaccharide polymer, chitin. Simulated conservative tracer as a proxy for S-metolachlor concentration predictions compared to POCIS measurements in Arcachon Bay ArchiMer

The work presented here aims at comparing monitoring of S-metolachlor, the major pesticide in use in the Arcachon Bay (South West of France, transitional coastal area), by chemical analysis (monthly passive sampling) and contaminant dissipation modeling from sources (Mars-2D model). The global strategy consisted in i) identifying the major sources of S-metolachlor to the Bay, ii) monitoring these sources for 12 months, and iii) comparing modeled data in the Bay based on measured inputs, to chemical measurements made inside the Bay along with the 12-month source monitoring. Acetylcholinesterase activity in copepods (Tigriopus brevicornis) from the Vilaine River estuary, France, as a biomarker of neurotoxic contaminants ArchiMer

From April 1997 to June 1998, 14 measurements of acetylcholinesterase (AChE) enzymatic activity were performed with the copepod, Tigriopus brevicornis, collected at five stations in the Vilaine River estuary (South Brittany, France). Simultaneously, four chemical analyses of triazines and one analysis of total pesticides in water were undertaken. AChE activity levels in T brevicornis were compared to the levels measured at a reference site not exposed to effluents from Vilaine River. Breast cancer mortality has been increasing in Arica Chile where it has surpassed the national rates 11 times between 1990 and 2010. The city of Arica was sprayed with the organophosphrous pesticide malathion in order to control the Mediterranean fly 33 years ago. Moreover we have demonstrated that a malathion treatment induces the formation of breast carcinomas in Sprague Dowley female rats. The objective of this work was to find a relationship between malathion aerial spraying and the increased mortality rate due to breast cancer that has been observed in Arica in recent years. Oxidative stress and comet assay in tissues of mice administered glyphosate and ampa in drinking water for 14 days JBAG

Excessive amounts of the herbicide glyphosate are incorporated daily to the soil and the ecosystems. AMPA is its major environmental breakdown product. In this study we determined the levels of thiobarbituric acid reactive substances (TBARs); quantified superoxide dismutase (SOD) and catalase (CAT) activity in liver, kidney, lung and heart, and performed the comet assay in blood and liver of mice administered glyphosate (40 or 400 mg/kg/day) or AMPA (100 mg/kg/day) in drinking water for 14 days. Exposure to glyphosate 400 mg/kg induced a statistically significant (p <0.05) decrease of SOD activity in heart and an increase in CAT activity in kidney. Simple and rapid detection of pesticides and chemicals using surface enhanced Raman spectroscopy OAK

独自の銀ナノ粒子・基板を試作し,小型ラマン分光装置を用いジチオカーバメート系農薬(Thiram: tetramethylthiuram disulfide)・パルノックスおよびメラミンのラマンおよび表面増強ラマン(Surface Enhanced Raman Spectroscopy:SERS)スペクトルの計測を行った。濃度が4ppm から0.04ppmの範囲では,濃度とピーク強度との間に良い直線関係が示された。同様に小麦粉に混合したメラミンのラマンスペクトルおよびSERS スペクトルを顕微ラマン分光装置により計測した。1ppm濃度程度まで特有のSERS 吸収ピークが観測され,定性的検出が可能であった。この銀ナノ粒子・基板を用いた小型で安価な装置による表面増強ラマン(SERS)スペクトル計測は,農薬や化学物質の分析法として感度が高く実用性が十分あると考えられた。
