Genetic monitoring of a Santa Ines herd using microsatellite markers near or linked to the sheep MHC R. Bras. Zootec.
Petroli,César Daniel; Paiva,Samuel Rezende; Corrêa,Marilma Pachêco Chediak; McManus,Concepta.
This study aimed to analyze genetic diversity in a conservation nucleus of Santa Inês sheep using thirteen microsatellite loci on chromosome 20 (where the Sheep Major Histocompatibility Complex - Ovar-MHC - is found). Seventy three animals from one herd born from 2004 to 2006 were evaluated as a principal nucleus. Seventy one animals from two other herds were used as control comparison. There was a reduction in heterozygosity over the years in relation to the whole population. This may be due to the repeated use of the same sires. The estimates of molecular coancestrality also indicated an increase in genetic similarity between individuals with the herd over the years. A high number of alleles occurred exclusively in the principal nucleus herd, but with a...

Microsatellite markers for identification of Prunus spp. rootstocks Scientia Agricola
Bianchi,Valmor João; Sansavini,Silviero; Fachinello,José Carlos.
Cultivar characterization for fruit trees certification requires fast, efficient and reliable techniques. Microsatellite markers (SSR) were used in the molecular characterization of 29 Prunus spp. rootstocks. The DNA from the rootstocks was analyzed using five pre-selected SSR primers (UDP96-005, UDP96-008, UDP96-013, UDP96-18 and UDP98-414) and revealed 81 alleles, which allowed each genotype to be identified. The UDP96-005 marker generated the most information, i.e., 23 well-distributed, polymorphic alleles among all genotypes. The 21 polymorphisms produced by UDP96-013 occurred mainly as a result of high degree of variability among genotypes of the Prunophora subgenus. In the dendrogram, the five markers allowed the 29 rootstocks to be grouped into...

ABSTRACT Sugarcane orange rust caused by Puccinia kuehnii has recently become an important disease in sugarcane crops and its spread is causing great concern to growers. In this study, we analyzed spores from symptomatic orange rust sugarcane leaves collected in multiple locations in Cuba in a 4-year-period in order to characterize morphological traits of P. kuehnii, establish an adequate molecular technique to characterize it, and determine its infection court in sugarcane. Orange rust caused by P. kuehnii was confirmed by polymerase chain reaction (PCR) and morphological characterization. AFLP markers detected high diversity in P. kuenhnii samples. Sequencing of rDNA regions, as expected, did not reveal differences and SSR markers designed for P....

Genetic structure analysis of Mauritia flexuosa natural population from the Lençóis Maranhenses region using microsatellite markers Scientia Agricola
Castro,Domingos Augusto Mendonça; Costa,Tatiana Santos; Cardoso,Ariane Santos; Ramos,Helaine Christine Cancela; López,Jorge Alberto; Diniz,Leandro Eugenio Cardamone.
ABSTRACT Buriti (Mauritia flexuosa L.f), a palm tree native to South America and widely distributed in Brazil, displays significant ecological, economic and biotechnological importance. However, the disorderly extractivism and environmental degradation of its endemic areas are leading to reductions in, and/or extinction of the buriti palm tree, causing ecological imbalance with significant economic losses for rural communities and genetic diversity. Consequently, populational genetic diversity studies have become relevant as a strategy for conserving the species. Therefore, this study evaluated the genetic structure of 10 populations from the Lençóis Maranhenses region, in the state of Maranhão, Brazil, using microsatellite markers. Results indicated that...

CTAB methods for DNA extraction of sweetpotato for microsatellite analysis Scientia Agricola
Borges,Aline; Rosa,Mariana Silva; Recchia,Gustavo Henrique; Queiroz-Silva,Jurema Rosa de; Bressan,Eduardo de Andrade; Veasey,Elizabeth Ann.
Microsatellite markers have proved to be useful in genetic diversity assessments of sweetpotato (Ipomoea batatas) but practical DNA extraction methods to ensure good quality and quantity DNA for these studies are yet to be established. This study compares the efficiency of three modified methodologies for DNA extraction of six sweetpotato landraces using the CTAB extraction buffer in regard to quantity and purity of DNA quantification and microsatellite band patterns. All methodologies yielded satisfactory results, but the method based in leaf tissue macerated in liquid nitrogen was deemed more adequate because of its simplicity and lower cost. However, the method based in dry leaf tissue was considered more advantageous, first because elicits...

Structure of genetic diversity of Bemisia tabaci (Genn.) (Hemiptera: Aleyrodidae) populations in Brazilian crops and locations Scientia Agricola
Fontes,Fernanda von Hertwig Mascarenhas; Colombo,Carlos Augusto; Lourenção,André Luiz.
Bemisia tabaci (Genn.) represents a complex of cryptic species that causes losses of many valuable crops. Even though there are differences in their ability to colonize hosts, transmit phytovirus and develop resistance to insecticides, the genetic differentiation of the insect populations is important for the adoption of control measures. Therefore, the genetic diversity of B. tabaci populations in economically important crops in Brazilian locations was characterized through a microsatellite analysis. Eight microsatellite markers were used for the analysis of eight populations, three in Solanum tuberosum (States of São Paulo, Minas Gerais and Bahia), two in Glycine max (States of São Paulo and Mato Grosso), one in Phaseolus vulgaris and Brassica oleracea...

Morillo C,Ana Cruz; Morillo C,Yacenia; Ceballos L,Hernán.

ABSTRACT The search for alternatives to increase productivity and sustainability of livestock production in the Amazon region without increasing deforestation is challenging. Mixed pastures of grasses with forage peanut (Arachis pintoi) have shown positive economic impacts. However, gaps in the knowledge of the reproductive biology of A. pintoi have limited the development of new cultivars adapted to the environmental variations in the Brazilian Amazon. Pasture consortiums of Brachiaria humidicola with forage peanuts (cv. Mandobi) resulted in a 42% increase in weight gain productivity. New cultivars better adapted to the Amazon climate should bring even greater gains. We evaluated the mating system in twenty A. pintoi accessions, and approximately 40...

Genetic Characterization of Old Grapevines collected in Oases of the Atacama Desert Chilean J. Agric. Res.
Poblete,Ingrid; Pinto,Manuel; de Andrés,María Teresa; Hinrichsen,Patricio.
Old grapevine (Vitis vinifera L.) accessions are a source of genes that could be rescued for use per se or in modern breeding programs. The first step in this rescuing is collecting and characterizing the germplasm from a particular region. This study presents the genetic characterization of 21 grapevine accessions collected from the Atacama Desert in the far North of Chile. Characterization was based on 12 microsatellites (Simple Sequence Repeats, or SSRs) supplemented with Amplified Fragment Length Polymorphic (AFLP) markers. Most of the collected accessions produced red berries and shared the genetic characteristics of the cv. País, an old genotype found throughout America. However, among those red-berried accessions, one showed a severe abortive...
