Cultivar characterization for fruit trees certification requires fast, efficient and reliable techniques. Microsatellite markers (SSR) were used in the molecular characterization of 29 Prunus spp. rootstocks. The DNA from the rootstocks was analyzed using five pre-selected SSR primers (UDP96-005, UDP96-008, UDP96-013, UDP96-18 and UDP98-414) and revealed 81 alleles, which allowed each genotype to be identified. The UDP96-005 marker generated the most information, i.e., 23 well-distributed, polymorphic alleles among all genotypes. The 21 polymorphisms produced by UDP96-013 occurred mainly as a result of high degree of variability among genotypes of the Prunophora subgenus. In the dendrogram, the five markers allowed the 29 rootstocks to be grouped into... |

ABSTRACT Sugarcane orange rust caused by Puccinia kuehnii has recently become an important disease in sugarcane crops and its spread is causing great concern to growers. In this study, we analyzed spores from symptomatic orange rust sugarcane leaves collected in multiple locations in Cuba in a 4-year-period in order to characterize morphological traits of P. kuehnii, establish an adequate molecular technique to characterize it, and determine its infection court in sugarcane. Orange rust caused by P. kuehnii was confirmed by polymerase chain reaction (PCR) and morphological characterization. AFLP markers detected high diversity in P. kuenhnii samples. Sequencing of rDNA regions, as expected, did not reveal differences and SSR markers designed for P.... |

ABSTRACT Buriti (Mauritia flexuosa L.f), a palm tree native to South America and widely distributed in Brazil, displays significant ecological, economic and biotechnological importance. However, the disorderly extractivism and environmental degradation of its endemic areas are leading to reductions in, and/or extinction of the buriti palm tree, causing ecological imbalance with significant economic losses for rural communities and genetic diversity. Consequently, populational genetic diversity studies have become relevant as a strategy for conserving the species. Therefore, this study evaluated the genetic structure of 10 populations from the Lençóis Maranhenses region, in the state of Maranhão, Brazil, using microsatellite markers. Results indicated that... |

Bemisia tabaci (Genn.) represents a complex of cryptic species that causes losses of many valuable crops. Even though there are differences in their ability to colonize hosts, transmit phytovirus and develop resistance to insecticides, the genetic differentiation of the insect populations is important for the adoption of control measures. Therefore, the genetic diversity of B. tabaci populations in economically important crops in Brazilian locations was characterized through a microsatellite analysis. Eight microsatellite markers were used for the analysis of eight populations, three in Solanum tuberosum (States of São Paulo, Minas Gerais and Bahia), two in Glycine max (States of São Paulo and Mato Grosso), one in Phaseolus vulgaris and Brassica oleracea... |

ABSTRACT The search for alternatives to increase productivity and sustainability of livestock production in the Amazon region without increasing deforestation is challenging. Mixed pastures of grasses with forage peanut (Arachis pintoi) have shown positive economic impacts. However, gaps in the knowledge of the reproductive biology of A. pintoi have limited the development of new cultivars adapted to the environmental variations in the Brazilian Amazon. Pasture consortiums of Brachiaria humidicola with forage peanuts (cv. Mandobi) resulted in a 42% increase in weight gain productivity. New cultivars better adapted to the Amazon climate should bring even greater gains. We evaluated the mating system in twenty A. pintoi accessions, and approximately 40... |

ABSTRACT The genus Manihot Mill. contains about 120 species of which about 104 occur in Brazil. We tested the cross-amplification of ten microsatellite markers developed for Manihot esculenta in 15 species of Manihot endemic to the Brazilian Cerrado. We also evaluated the genetic diversity of Manihot irwinii, M. orbicularis, and M. purpureocostata. Ten pairs of primers were amplified among 14 species of Manihot. The percentage of polymorphic loci per species varied from 70 to 100 %. Nine markers showed amplification and polymorphism when evaluated on polyacrylamide gel. The markers were combined to form three sets for multiplex genotyping for genetic diversity analysis, and showed 51, 75, and 75 alleles in M. irwinii, M. orbicularis, and M.... |

Abstract: Eugenia uniflora L. (Myrtaceae) is a tree species widely distributed in South America suffering the effects of the exploitation of natural populations. In this study, we employed low coverage sequencing of the E. uniflora genome for mining of SSR markers. The de novo assembly generated 2,601 contigs with an average length of 1139 bp and spans 3.15 Mb. A total of 76 dimer, 33 trimer and two compound SSR loci were identified. Twelve selected SSR loci were employed to genotype 30 individuals from two natural populations. A total of 73 alleles were detected (mean A= 6.1) were observed, the mean effective number of alleles was Ae = 3.91, mean Ho was 0.23 and mean HE was 0.70). The mean Wright’s within population fixation index was FIS = 0.66 and... |

Microsatellite markers previously developed for Pitcairnia albiflos Herb. and Pitcairnia geysksii L.B.Sm. were used in cross-amplification tests of five other Bromeliaceae species. Ten (76.9%) out of the 13 evaluated pair of primers had positive results for some of the species tested. The number of polymorphic alleles ranged between two and four in most species. Ho values ranged between zero, in Pitcairnia flammea Lindl. (PaA05), Aechmea ramosa Mart ex Schult & Schult and Billbergia horrida Regel (PaC05), and one in Billbergia euphemiae E. Morren (PaA05, PaA10, PaC05 and PaD07). This study showed that microsatellite markers developed for P. albiflos and P. geysksii effectively amplified the DNA samples of Pitcairnia flammea, Aechmea nudicaulis (L.)... |

ABSTRACT Thousands of years of tradition of olive cultivation in Montenegro is witnessed with the ‘Old olive’ estimated to be more than 2000 years old. ‘Old olive’ draws attention to its genetic potential and the relation to the closest grown olive cultivars. Therefore, the aim of this paper was to give the basic description of ‘Old olive’ (through 20 morphological and 37 chemical properties) as well as to determine its DNA profile and to examine the closeness with major olive cultivar grown, cv. ‘Žutica’. The obtained data showed the differences between examined samples. ‘Old olive’ started flowering (27. May) more than 10 days later than cv. ‘Žutica’ (14. May), and began ripening 15 days later (20th October and 5th October, respectively). From 21... |

The aim of this study was to evaluate the selected 41 SSR markers developed for yellow passion fruit (Passiflora edulis f. flavicarpa Sims.) for their transferability to 11 different Passiflora species. Twenty-one SSR were successfully amplified in 10 wild species of passion fruit producing 101 bands. All the markers were amplifiable for at least one species. The mean transferability was 68,8%, ranging from 15,4% (primer PE11) to 100 % (PE13, PE18, PE37, PE41 and PE88). Transferability was higher for the species from the Passiflora subgenus than for those from the Decaloba and Dysosmia subgenus. The results indicated a high level of nucleotide sequence conservation of the primer regions in the species evaluated, and consequently, they could potentially be... |
