Este trabalho teve como objetivo avaliar níveis de AIB no enraizamento in vitro de microestacas de mamoeiro (Carica papaya L. ‘Tainung 01’). Ápices caulinares de plantas com 120 dias de idade foram estabelecidos e avaliados em meio MS, suplementado com complementos orgânicos constituídos de sacarose a 87,64 mM, mio-inositol a 555,06 µM, tiamina-HCl a 0,30 µM, piridoxina-HCl a 2,43 µM, ácido nicotínico a 4,06 µM e 0; 0,74; 1,48; 2,96 e 5,92 µM de AIB, em delineamento inteiramente casualizado, com quatro repetições e cinco tubos de ensaio por parcela. Somatic embryogenesis and in vitro plant regeneration are fundamental processes in the obtainment of transgenic maize plants. Explant, genotype and culture medium are determining factors in these processes. This work aimed to promote in vitro seed germination and disinfestation of the sweet cactus. Seeds were submerged in alcohol at 70% for 1 min. and then treated with sodium hypochlorite solution at different concentrations: 0.0; 0.5; 1.0 and 1.5%. After this treatment, the seeds were washed four times in distillated, deionized and autoclaved water. Seeds were inoculated in MS medium with different concentrations of sucrose (0.0, 2.5, 5.0, 7.5 and 10.0 g L-1). The development of an efficient regeneration system is crucial for the genetic transformation of sugarcane. The aims of this study were to regenerate plants from somatic embryos and analyze the origin and division pattern of cells during the different embryonic developmental stages of the sugarcane cultivars RB855156 and RB72454. For both cultivars, the best results for embryogenic callus induction were obtained with explant cultures on culture medium containing 13.5 µM 2,4-dichlorophenoxyacetic acid (2,4-D) followed by callus culture on solid medium with 4.5 µM 2,4-D. O objetivo deste trabalho foi determinar a fonte de silício que proporcione melhor crescimento em plântulas de Cattleya loddigesii. Plântulas oriundas de sementes germinadas in vitro com aproximadamente 1,0 cm de comprimento foram inoculadas em tubos de ensaio contendo 15 mL de meio de cultura Knudson C modificado, acrescido de silicato de sódio (0, 5, 10 e 20 mg L-1) e silicato de potássio (0, 5, 10 e 20 mL L-1), em todas as combinações possíveis. This work describes an efficient micropropagation protocol for Verbena litoralis and the study of the antinociceptive and antioxidant activities in extracts of this species. For the establishment in vitro, surface-sterilization procedures and PVPP showed high efficiency in fungal-bacterial contamination and phenol oxidation controls. Nodal segments cultivation in MS medium supplemented with 6-benzyladenine (7.5 µM)/α-naphthaleneacetic acid (NAA; 0.005 µM) induced multiple shoots. Elongated shoots were rooted with IAA (0.2 µM). Micropropagation of Glandularia perakii Cov. et Schn. (Verbenaceae): a native species with ornamental potential Biocell

Glandularia perakii is a perennial species with beautiful violet flowers that grows in the stony soil of Mendocine pedemont. A plentiful and prolonged flowering confers it an important ornamental potential. In this paper, a method of propagation of G. perakii from nodal segments is reported. Proliferating microshoot cultures were obtained by placing nodal segment on Murashige and Skoog medium (MS) supplemented with 20 g.L-1 of sucrose without growth regulators. In this medium multiplication rate after 20 days was 7.9. Rooted plants were acclimatized successfully.

Hydrogen peroxide in micropropagation of Lilium: A comparison with a traditional methodology Biocell

The micropropagation of Lilium longiflorum requires adequate equipment which may not be afforded by small laboratories or producers. In this work we compared traditional methodology with a protocol that included easily available elements to sterilize materials and culture media, together with addition of hydrogen peroxide (H2O2) into the nutrient media as chemical sterilizer. A series of H2O2 concentrations (0.005, 0.010, 0.015 and 0.020% p/v) was used to control contamination during in vitro establishment and subsequent cultivation; the explant organogenic response was also examined and compared to the traditional micropropagation technique. Recent gene knock-out studies in mice have suggested that ventricular myosin light chain-2 (vMLC2) has a role in the regulation of cardiogenic development and that perturbation in expression of vMLC2 is linked to the onset of dilated cardiomyopathy. Down syndrome is determined by the presence of an extra copy of autosome 21 and is expressed by multiple abnormalities, with mental retardation being the most striking feature. The condition results in altered electrical membrane properties of fetal dorsal root ganglia (DRG) neurons, as in the trisomy 16 fetal mouse, an animal model of the human condition. Biochemical and morphological responses to abiotc elicitor chitin in suspension-cultured sugarcane cells BABT

Cells of Saccharum officinarum submitted to hydrolyzated chitin for 1 to 8h produced phenolic compounds. These alterations were observed through cytochemical methods using Toluidine Blue and Phloroglucinol/HCl. After 4 h, besides cell wall change, there was a change in nuclear pattern of chitin treated cells. There was a 96% increase in nuclear area in 6 h chitin treated material, as observed by Feulgen reaction. The treated cells showed chromatin compacted regions and a degeneration process of nucleoli. Plant regeneration through somatic embryogenesis of yacón [smallanthus sonchifolius (poepp. and endl.) H. Robinson] BABT

This work studied a new protocol for organogenic calli induction and characterization of the morphology and ultrastructure of callogenesis in leaf explants of Passiflora gibertii N. E. Brown, a native passion fruit species from Brazil. Calli induction was performed in different growth conditions (light and dark), different MS medium salt concentrations (MS and MS half strength) and the presence or absence of coconut water. Bauhinia forficata link shoot regeneration: histological analysis of organogenesis pathway BABT

Plant regeneration was achieved from cells of callus induced from hypocotyl segments of Bauhinia forficata on half strength Murashige and Skoog culture medium supplemented with several concentrations of BAP. Within 40 days of culture shoot buds formation was observed on callus surface. Calli were then transferred to a same composition culture medium without plant growth regulator in order to induce shoot elongation. In order to achieve adventitious shoot regeneration from young tissues of Racosperma mangium, several combinations of growth regulators were tested. Kinetin (KIN) and a -naphthalenacetic acid (NAA), thidiazuron (TDZ) and NAA or indole-3-butyric acid (IBA), 4-CPPU and 2,4-dichlorophenoxyacetic acid (2,4-D) were added to basic medium composed of MS salts, MS vitamins and 3% sucrose. Influence of Silver Nitrate on Somatic Embryogenesis Induction in Arabica Coffee (Coffea arabica L.) BABT

Abstract The influence of silver nitrate (AgNO3), benzyladenine (BAP), and indole-3-acetic acid (IAA) on low frequency somatic embryogenesis (LFSE) induction in Caturra and Catuaí arabica coffee was evaluated. For the Caturra cultivar, the production of somatic embryos was significantly increased by adding AgNO3 to the semisolid culture medium. The highest average number of somatic embryos for this cultivar was obtained using 6.6 μM BAP, 2.85 μM IAA, and 40 μM AgNO3. Plant regeneration from cotyledonary explants of Eucalyptus camaldulensis dehn and histological study of organogenesis in vitro BABT

The present work aimed at regenerating plants of Eucalyptus camaldulensis from the cotyledonary explants and describing the anatomy of the tissues during callogenesis and organogenesis processes, in order to determine the origin of the buds. The cotyledonary leaves of E. camaldulensis were cultured in Murashige and Skoog (MS), WPM and JADS media supplemented with 2.7 µM NAA and 4.44 µM BAP. The best results for bud regeneration were obtained on MS and WPM media (57.5 and 55% of calluses formed buds, respectively). Shoot elongation and rooting (80%) were obtained on MS/2 medium (with half-strength salt concentration) with 0.2% activated charcoal. Indução in vitro de raízes adventícias em explantes de salix (Salix humboldtiana Willdenow)(Salix humboldtiana Willdenow) BABT

With the objective to investigate the in vitro behavior of salix (Salix humboldtiana Willdenow), leaf and nodal segments were inoculated in WPM medium supplemented with NAA and IBA. In leaf explants, the presence of these growth regulators, isolated or in combination, induced the formation of adventicious roots. Root hairs were only observed when NAA was present. In nodal explants, while the combination of 2,68 µM ANA + 2,46 µM IBA induced bud growth, the other treatments (4,92 µM IBA; 2,68µM ANA + 4,92 µM IBA; 5,37 µM ANA + 4,92 µM IBA; 5,37µM ANA) induced the formation of adventicious roots.
