Activity of cellulase from Thermoactinomycetes and Bacillus spp. isolated from Brassica waste compost Scientia Agricola
Chang,Chen-Chin; Ng,Chang-Chai; Wang,Chung-Yi; Shyu,Yuan-Tay.
Plant wastes present a high cellulose content, which is an ideal organic material for composting. Five strains of thermophiles from processed Brassica waste were isolated, and the hydrolytic activity on various cellulosic biomass substrata and their temperature profiles were determined. 16S rRNA sequencing identified these strains as Thermoactinomyces and Bacillus spp. Maximal cellulase activity corresponded to 2.3 U mL-1 of enzyme. The application of these strains on Brassica rapa residues demonstrates increased total nitrogen content). TA-3, a Thermoactinomycetes sp. strain, performs best among all inoculants, increasing the nitrogen content from 0.74 to 0.91%, and decreasing the carbon content from 15.4 to 12.2%, showing its high efficiency and...

Invertase and sucrose synthase activities in coffee plants sprayed with sucrose solution Scientia Agricola
Silva,José Carlos da; Alves,José Donizeti; Alvarenga,Amauri Alves de; Magalhães,Marcelo Murad; Livramento,Dárlan Einstein do; Fries,Daniela Deitos.
One management practice of which the efficiency has not yet been scientifically tested is spraying coffee plants with diluted sucrose solutions as a source of carbon for the plant. This paper evaluates the effect of foliar spraying with sugar on the endogenous level of carbohydrates and on the activities of invertase and sucrose synthase in coffee (Coffea arabica L.) seedlings with reduced (low) and high (normal) levels of carbon reserve. The concentrations used were 0.5 and 1.0% sucrose, and water as a control. The use of sucrose at 1.0% caused an increase in the concentration of total soluble sugars in depauperate plants, as well as increased the activity of the following enzymes: cell wall and vacuole acid invertase, neutral cytosol invertase and...

Enzymes immobilized in Langmuir-Blodgett films: Why determining the surface properties in Langmuir monolayer is important? Anais da ABC (AABC)
CASELI,LUCIANO.
ABSTRACT In this review we discuss about the immobilization of enzymes in Langmuir-Blodgett films in order to determine the catalytic properties of these biomacromolecules when adsorbed on solid supports. Usually, the conformation of enzymes depends on the environmental conditions imposed to them, including the chemical composition of the matrix, and the morphology and thickness of the film. In this review, we show an outline of manuscripts that report the immobilization of enzymes as LB films since the 1980’s, and also some examples of how the surface properties of the floating monolayer prepared previously to the transfer to the solid support are important to determine the efficiency of the resulting device.

Metabolic responses of channel catfish (Ictalurus punctatus) exposed to phenol and post-exposure recovery Anais da ABC (AABC)
Moraes,Fernanda D.; Rossi,Priscila A.; Figueiredo,Juliana S.L.; Venturini,Francine P.; Cortella,Lucas R.X.; Moraes,Gilberto.
Metabolic adjustments were studied in channel catfish Ictalurus punctatus exposed to 1.5 mg L-1 of phe nol (10% LC50) for four days and recovered for seven days. Lower triacylglycerol (TGA) stores and increased muscle fat free acids (FFA) suggest fat catabolism in muscle. Remarkable liver FFA decrease (-31%) suggests liver fat catabolism as well. Increased muscular ammonia levels and ASAT (aspartate aminotransferase) and decreased plasma aminoacids suggest higher muscular amino acid uptake. Constant levels of glucose and increased liver glycogen stores, associated with lower amino acids in plasma, indicate gluconeogenesis from amino acids. This is supported by higher hepatic ALAT and ASAT. Higher hepatic LDH followed by lower plasma lactate may indicate...
