Cleff,Marlete Brum; Meinerz,Ana Raquel; Xavier,Melissa; Schuch,Luiz Filipe; Meireles,Mário Carlos Araújo; Rodrigues,Maria Regina Alves; Mello,João Roberto Braga de.
The aim of this study was to evaluate the in vitro activity of the essential oil extracted from Origanum vulgare against sixteen Candida species isolates. Standard strains tested comprised C. albicans (ATCC strains 44858, 4053, 18804 and 3691), C. parapsilosis (ATCC 22019), C. krusei (ATCC 34135), C. lusitaniae (ATCC 34449) and C. dubliniensis (ATCC MY646). Six Candida albicans isolates from the vaginal mucous membrane of female dogs, one isolate from the cutaneous tegument of a dog and one isolate of a capuchin monkey were tested in parallel. A broth microdilution technique (CLSI) was used, and the inoculum concentration was adjusted to 5 x 10(6) CFU mL-1. The essential oil was obtained by hydrodistillation in a Clevenger apparatus and analyzed by gas...

The extraction parameters for Pleurotus eryngii SI-02 exopolysaccharide (EPS) produced during submerged culture were optimized using response surface methodology (RSM). The optimum conditions for EPS extraction were predicted to be, precipitation time 20.24 h, ethanol concentration 89.62% and pH 8.17, and EPS production was estimated at 7.27 g/L. The actual yield of EPS under these conditions was 7.21 g/L. The in vitro antioxidant results of the EPS showed that the inhibition effects of EPS at a dosage of 400 mg/L on hydroxyl, superoxide anion and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals were 59.63 ± 3.72%, 38.69 ± 2.59%, and 66.36 ± 4.42%, respectively, which were 12.74 ± 1.03%, 8.01 ± 0.56%, and 12.19 ± 1.05% higher than that of butylated...

Molecular docking, synthesis and in vitro antimalarial evaluation of certain novel curcumin analogues BJPS
Dohutia,Chandrajit; Chetia,Dipak; Gogoi,Kabita; Bhattacharyya,Dibya Ranjan; Sarma,Kishore.
ABSTRACT The receptor protein PfATP6 has been identified as the common target of artemisinin and curcumin. The work was initiated to assess the antimalarial activity of six curcumin derivatives based on their binding affinities and correlating the in silico docking outcome with in vitro antimalarial screening results. A ligand library of thirty two Knoevenagel condensates of curcumin were designed and docked against PfATP6 protein and six compounds with the best binding scores were synthesized and screened for their antimalarial activity against the sensitive 3D7 strain of Plasmodium falciparum. ADME/Tox, pharmacokinetic and pharmacodynamic profiles of the designed compounds were analyzed and reported. 4-FB was found to have similar binding energy to the...

Enhanced in vitro drought tolerance of Solanum tuberosum and Solanum commersonii plants overexpressing the ScCBF1 gene Ciencia e Investigación Agraria
Pino,María Teresa; Ávila,Andrea; Molina,Andrea; Jeknic,Zoran; Chen,Tony H.H.
Cultivated potato crops are sensitive to drought stress, reducing yield and tuber quality when the soil water potential drops to -0.3 MPa. However, drought not only affects plant growth and physiological activity, but this stress also induces biochemical and molecular changes in the cellular gene expression profile. triggering genes that play a direct role in plant protection and gene regulation. The genes involved in regulation include C-repeat Binding Factors/Dehydration responsive element binding (CBF/DREB) transcription factors, which increase cold, drought and salt tolerance in different plant species. The aim of this research was to evaluate whether the overexpression of the ScCBF1 gene from Solanum commersonii enhances drought stress tolerance in...

Plant tissue culture: Current status, opportunities and challenges Ciencia e Investigación Agraria
García-Gonzáles,Rolando; Quiroz,Karla; Carrasco,Basilio; Caligari,Peter.
In the last two decades plant biotechnology applications have been widely developed and incorporated into the agricultural systems of many countries worldwide. Tissue culture tools have been a key factor to support such outcomes. Current results have allowed plant biotechnology and its products -including transgenic plants with several traits- to be the most assimilated technology for farmers and companies, representing several benefits such as: 125 millions ha of transgenic crops in 2008, the reduction of pesticides application by up to 9% in the last ten years, transgenic plants with a better nutritional quality, mass propagation of selected and healthy plants, and the production of proteins for industrial or therapeutic use. The rapid and extensive...

Overlapping of mononuclear cells derived from bone marrow in rats' intervertebral discs: an in vitro study Ciência Rural
Fontes,Erica Batista; Borges,Andréa Pacheco Batista; Pippi,Ney Luis; Rosa,Maurício; Sprada,Arícia; Togni,Monique.
In this study, bone marrow mononuclear cells (BM-MNC) derived from rats were used in order to promote intervertebral disc regeneration. These cells were isolated after centrifugation in a Ficoll-Paque™ PLUS density gradient and then placed in plastic dishes to proliferate during a period of 14 days. The BM-MNCs were previously labeled with the fluorescent membrane marker Chloromethyl-benzamidodialkylcarbocyanine (CM-DIL), and thereafter were implanted in rats' intervertebral discs explants as an in vitro experimental model. Daily analyses of the cells under a fluorescence microscope revealed morphological changes, which assumed a thin and elongated shape similar to cells that originally form the annulus fibroses. Histopathological analysis demonstrated the...
