The conventional technology for in vitro plant conservation for cassava (Manihot esculenta Crantz) germplasm collections is laborious due to the need for several sub-culturing procedures per year. This practice implies high costs for medium preparation, tissue culture tubes, time-consuming labor, risks of contamination, mislabeling of accession, and the need for large growth chambers. We have developed a new procedure using in vitro cultivated nodal axillary buds treated with different abscisic acid (ABA) concentrations to reduce the time for recycling transplants cultivated in a SP basic nutritive medium. The experiment was carried out to determine the appropriate dose of coconut water as supplement for in vitro cultivation of zygotic embryos from 19 olive genotypes. The isolated embryos of the olive seeds were immersed on culture medium containing 0 (control), 25, 50, and 100mL L-1 of fresh and sterile coconut water and kept for 45 days under controlled environment. The percentage of germination, shoot length, number of roots, number of leaves and number of internodes were measured for all 19 olive genotypes. ABSTRACT: Hyptis marrubioides (Lamiaceae) is a medicinal plant that is native from Brazilian Cerrado. In vitro propagation techniques make use of elicitors to alter metabolic pathways, affecting how molecules are produced both qualitatively and quantitatively. This research aimed to evaluate how abiotic elicitors salicylic acid (SA) and silver nitrate (SN) at concentrations of 30µM or 60µM influence Hyptis marrubioides seedling growth by two different in vitro culture methods. That is why it is important to study ways to promote a production of lavender raw material. For this, four experiments were carried out in order to study the germination and initial in vitro development of lavender. ABSTRACT: The availability of an efficient protocol for in vitro regeneration is imperative for genetic transformation. Using genotypes adapted to the target region as a transgenic platform accelerates the development of cultivars. Therefore, this study aimed to adapt an in vitro regeneration protocol for Brazilian wheat genotypes. For this purpose, the in vitro regeneration capacity of immature embryos from six Brazilian wheat genotypes using two protocols of regeneration of somatic embryos was analysed. With the objective to promote in vitro callus induction, cotyledon and hypocotyl segments of "perennial soybean" (Glycine wightii (Wight & Arn.) Verdc.) were inoculated in basal medium MS supplemented with sucrose (1.5 e 3%) and 0.8% agar and different concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-furfurylaminopurine (kinetin). The study was carried out in order to evaluate the effect of temperature and in vitor stigma receptivity on regeneration of orange cultivar (Valência, Pêra and Natal) pollen. Two experiments were carried out, in the first the ideal temperature of germination was assessed.
