Haldane's rule and other aspects of reproductive isolation observed in the Anastrepha fraterculus complex (Diptera: Tephritidae) Genet. Mol. Biol.

Some aspects of reproductive isolation between allopatric populations of two closely related species of the Anastrepha fraterculus complex (A. fraterculus sp. 1 and sp. 2) were evaluated in laboratory conditions. Most of the crosses were fertile in each species as well as between sp. 2 females and sp. 1 males. In the reciprocal cross only 41.7% of the matings yielded viable progeny. Egg hatching occurred at similar rates within the two species, but was significantly lower in the crosses between the species. Adult emergence did not differ significantly among crosses. Heterochromatin patterns and karyotype relationships within and between the genera Brycon and Salminus (Pisces, Characidae) Genet. Mol. Biol.

Chromosomes of two Brycon species (B. lundii and B. microlepis) and Salminus hilarii were analyzed. Based on constitutive heterochromatin distribution patterns, karyotypic relationships within and between Bryconinae and Salmininae were examined. A monophyletic origin for the genus Brycon, comprising at least two chromosome synapomorphies (presence of two large submetacentric bearing paracentromeric and telomeric heterochromatin), is suggested. Moreover, Bryconinae and Salmininae may represent a monophyletic unit among Characidae, as they share several chromosome features.

Expression of a methionine-rich storage albumin from the Brazil nut (Bertholletia excelsa H.B.K., Lecythidaceae) in transgenic bean plants (Phaseolus vulgaris L., Fabaceae) Genet. Mol. Biol.

Bean (Phaseolus vulgaris), an important component in the diet of people in developing countries, has low levels of the essential amino acid, methionine. We have attempted to correct this deficiency by introducing a transgene coding for a methionine-rich storage albumin from the Brazil nut via biolistic methods. The transgene's coding sequence was driven by a doubled 35S CaMV promoter and AMV enhancer sequences. The transgene was stable and correctly expressed in homozygous R2 to R5 seeds. In two of the five transgenic lines the methionine content was significantly increased (14 and 23%) over the values found in untransformed plants.

Conservation genetics of an endangered Catalonian cattle breed ("Alberes") Genet. Mol. Biol.

We biochemically analyzed and characterized the genetic structure of a population in danger of extinction ,"Alberes", a local cattle breed of the Catalonian Pyrenees (Spain and France). Ninety-two individuals were analyzed for five polymorphic genetic loci (Hb, Alb, Tf, Gc and Ptf2). The animals were grouped according to coat color: Fagina Alberes variety (N = 39) and Black Alberes variety (N = 53). The genetic structures and relationships between these subpopulations and one "outgroup" breed ("Bruna dels Pirineus") were analyzed and compared by using F-statistics. A study was made of the genetics of resistance to the soybean cyst nematode, race 3, in a population derived from crosses between the Brazilian soybean genotypes BR 90-4722 and FT-Cristalina. Crosses between the two parents were made, the F1 and F2 generations were obtained and the population was analyzed for the number of cysts found in each plant of each generation as well as the type of reaction to the nematode. The results showed that resistance to the cyst nematode in this soybean population is of a qualitative nature and conditioned by three genes, one dominant and two recessives. Neurocysticercosis, caused by encysted larvae of the tapeworm Taenia solium, is the most common infection of the central nervous system and a major public health problem in many countries. Prevalence in the region of Curitiba, located in the southern Brazilian State of Paraná, is one of the highest in the world. The genetics of host susceptibility to neurocysticercosis (NCC) is still obscure. To investigate if major histocompatibility complex (MHC) genes influence individual susceptibility to NCC, we performed a case-control association analysis. Premature ovarian failure (POF) in Brazilian fragile X carriers Genet. Mol. Biol.

The gynecological and reproductive histories of 193 women from fragile X families were surveyed. Among the 101 carriers of the premutation, 14 experienced premature menopause, contrarily to their 37 fully mutated and 55 noncarrier female relatives. Although premature menopause showed a tendency to cluster in certain fragile X families, as a group, the premutated women experienced menopause earlier than noncarriers. This suggests that premature menopause may be the extreme effect of a spectrum of ovarian anomalies associated with the fragile X premutation.

Genetic causes involved in Leishmania Chagasi infection innortheastern: Brazil Genet. Mol. Biol.

A sample of 502 individuals from 94 families from Jacobina, State of Bahia, Brazil, was investigated to determine the causal mechanisms involved in Leishmania chagasi (the causal agent of visceral leishmaniasis in the American hemisphere) infection, as measured by the intradermic reaction to antigens derived from this parasite, using complex segregation analyses. The results showed evidence of a major genetic mechanism acting on infection, with a frequency of a recessive (or additive) susceptibility gene (q) of approximately 0.45. A small multifactorial component (H = 0.29) acting in conjunction with a major recessive gene (q = 0.37) is not ruled out as a concomitant causative factor.

Protein and mitochondrial DNA have been used as molecular markers to assess variability in stock identification studies of fishes. Protein and mtDNA used for electrophoretic analysis are extracted from tissues, which often leads to death of the individuals. In this study, we present a skeletal muscle biopsy procedure to extract mitochondrial DNA that does not require specimen sacrifice. Eighty pirapitinga-do-sul (Brycon opalinus) were biopsied by the present technique, with no mortalities recorded. Gryllus assimilis L. has a karyotype of 2n = 29 (X0, male) and 30 (XX, female). The above karyotype was encountered along with another in which 2n = 28 (X0, male) and 2n = 29 (XX, female) in a population from the outskirts of Rio Claro city (São Paulo State, Brazil). Of eight specimens studied, five had the heterozygous karyotype involving a translocation and three had the basic karyotype. There were no individuals homozygous for the rearrangement. A leukocyte cryopreservation technique for cytogenetic studies Genet. Mol. Biol.

Several culture, cryopreservation, freezing and thawing methods were tested to develop an efficient technique for storing cervid and ovine leukocytes. The best results were obtained with McCoy or Vega y Martinez (VYM) solution with dimethyl sulfoxide (DMSO), horse serum and polyvinylpyrrolidone (PVP) as cryopreservatives. The best protocol for freezing was 4°C for 30 min followed by 15 min in liquid nitrogen vapor. To thaw, the still frozen material was placed onto cultivation medium for propagation.

The critical electrolyte concentrations (CEC) of sperm chromatin from animal species known or suspected to contain histone H1 variants were compared by examining the affinity of their DNA-protein complexes for toluidine blue in the presence of Mg2+. Bullfrog, sea urchin, bee and bumblebee spermatozoa were studied. The CEC for Rana catesbeiana and two sea urchin species were similar to that of histone H5-containing chromatin from chicken erythrocytes, thus confirming the biochemical and structural similarities of these DNA-protein complexes. Varroa jacobsoni infestation of adult Africanized and Italian honey bees (Apis mellifera) in mixed colonies in Brazil Genet. Mol. Biol.

Different levels of infestation with the mite Varroa jacobsoni have been observed in the various Apis mellifera races. In general, bees of European races are more susceptible to the mite than African honey bees and their hybrids. In Brazil honey bee colonies are not treated against the mite, though apparently both climate and bee race influence the mite infestation. Six mixed colonies were made with Italian and Africanized honey bees. The percentage infestation by this parasite was found to be significantly lower in adult Africanized (1.69 ± 0.44) than Italian bees (2.79 ± 0.65). Records of Gyr cows selected for milk production were obtained from the National Gyr Dairy Cattle Breeding Program (Embrapa/CNPGL) and analyzed, in order to estimate genetic parameters for the first three lactations and to verify the effects of some environmental factors on milk production from 1979 to 1994. Genetic parameters were estimated by REML with an animal model and a group of fixed effects that included classes of herd, year, season and age at calving. Milk production means and standard deviations were 2,183 kg, 707 kg; 2,682 kg, 762 kg and 2,638 kg, 851 kg, for first, second, and third lactations, respectively. Heritability estimates were 0.20, 0.12, and 0.19 for first, second, and third lactations, respectively, and repeatability was 0.44....
