Effect of temperature and initial pH on biohydrogen production from palm oil mill effluent: long-term evaluation and microbial community analysis Electron. J. Biotechnol.

Anaerobic sludge from palm oil mill effluent (POME) treatment plant was used as a source of inocula for the conversion of POME into hydrogen. Optimization of temperature and initial pH for biohydrogen production from POME was investigated by response surface methodology. Temperature of 60ºC and initial pHof 5.5 was optimized for anaerobic microflora which gave a maximum hydrogen production of 4820 ml H2/l-POME corresponding to hydrogen yield of 243 ml H2/g-sugar. Total sugar consumption and chemical oxygen demand (COD) removal efficiency were 98.7% and 46%, respectively. Immobilization and stabilization of papain on chelating sepharose: a metal chelate regenerable carrier Electron. J. Biotechnol.

A method for immobilization of papain has been selected based on the interaction between its histidine, cysteine and tryptophan residues with the immobilized metal ion (IMI) carrier for maximum binding on a small volume of the carrier. The immobilized papain retained high activity has improved thermal stability and the carrier could be recovered from the spent bound enzyme, to be reused. Reimmobilization of papain on the regenerated matrix was equally effective with the retention of maximum enzyme activity.

Here, we describe two alternative protocols that use inexpensive reagents and are not hazardous to the environment. Phenolic antioxidants extraction from raspberry wastes assisted by-enzymes Electron. J. Biotechnol.

The effect of enzymatic hydrolysis on phenolic antioxidant extraction was studied in raspberry solid wastes. This by-product possesses high content of crude fiber (60%) and low values of protein, oil and ash. Raspberry fiber composition suggests that biocatalysts with cellulase, hemicellulase and pectinase activities would be useful for carrying out an enzymatically assisted antioxidant extraction. Use of Aspergillus niger in the bioleaching of colemanite for the production of boric acid Electron. J. Biotechnol.

Colemanite is one of the most important underground riches of Turkey, having approximately 60% of the world boron deposits, and it has a large portion in the deposits. In this study, chemical leaching and biological leaching methods were used for production of boric acid from colemanite (2CaO · 3B3O3 · 5H2O) (Emet-Kütahya, Turkey). Oxalic acid concentration, temperature, stirring time and solid-to-liquid ratio were taken as parameters in the chemical leaching process. Optimization of b-galactosidase production using Kluyveromyces lactis NRRL Y-8279 by response surface methodology Electron. J. Biotechnol.

This paper investigates the production and optimization of b-galactosidase enzyme using synthetic medium by Kluyveromyces lactis NRRL Y-8279 in shake flask cultures. Among the different cell disintegration methods used, the highest specific activity was obtained when the cells were permeabilized using isoamyl alcohol. Response surface methodology was used to investigate the effects of four fermentation parameters (agitation speed, pH, initial substrate concentration and incubation time) on b-galactosidase enzyme production. A response surface technique was used to analyze the effect of Glomus fasciculatum, Pseudomonas fluorescens and worm-bed leachate (WBL) on growth, yield and characteristics of tomato (Lycopersicon esculentum Mill). The treatments combined inoculation with or without P. fluorescensor G. fasciculatum and the application of WBL at 20% (v/v) each day or every three days. Plant height, number of leaves and yield of tomato fruits was not affected by the factors studied. The lysozyme enzyme was immobilized on vitreous surface (fragments with diameters of 0.3 and 1.0 mm) for remediation of the microorganism Escherichia coli JM 109 into fresh water and saline solutions with 0.9% NaCl (w/v). Characterization of enzymatic film was carried out by infrared spectroscopy and atomic force microscopy techniques. Bactericide activity of the enzyme was evaluated by spectrophotometric analysis. It was verified that the enzymatic film was strongly coupled with the vitreous surface. The topographic analysis demonstrated that the deposited film was uniform and homogeneous. Gene expression and characterization of 2-keto-3-deoxy-gluconate kinase, a key enzyme in the modified Entner-Doudoroff pathway of Serratia marcescens KCTC 2172 Electron. J. Biotechnol.

We cloned 2-keto-3-deoxy-gluconate kinase (KDGK), which catalyzes the phosphorylation of 2-keto-3-deoxygluconate (KDG) to 2-keto-3-deoxy-6-phophogluconate (KDPG) from Serratia marcescens KCTC 2172. The nucleotide sequence revealed a single open reading frame containing 1,208 bp and encoding for 309 amino acids, with a molecular weight of 33,993 Da. The enzyme was purified via GST affinity chromatography. The putative KdgT binding site was detected upstream of the initial codon. The KDG kinase utilized 2-ketogluconate (KG) and KDG as substrates. The optimal temperature and pH for KDGK activity were 50ºC and 8.0, respectively.

Production of polyclonal antisera to manganese superoxide dismutase expressed in downy mildew resistant pearl millet and its application for immunodiagnosis Electron. J. Biotechnol.

Level of superoxide dismutase (SOD) in the seedlings of downy mildew resistant pearl millet genotype (IP 18293) increased 3-fold upon inoculation with Sclerospora graminicola. Mn-SOD was purified from pearl millet by ion-exchange and gel filtration chromatography. Polyclonal antibody was raised in rabbit against Mn-SOD of pearl millet. Anti-Mn SOD had an antibody titer of 1:20,000. ELISA results revealed a 1.5-fold increase in pure Mn-SOD preparation compared to the crude preparation. The antibody reactivity as detected by ELISA revealed that Mn-SOD is higher at 4 hrs post-inoculation in resistant pearl millet seedlings. The influence of centrifugation on Zymomonas mobilis aggregation Electron. J. Biotechnol.

Flocculent Zymomonas mobilis breaks down in smaller flocs and individual cells when centrifuged. The main consequence of it is an increase in the dispersion of the sample, suggesting that the influence of the centrifugal force on the aggregation of cells is worth to study. The experiments showed that the degree of dispersion varied between 30% and 100% when the centrifugal acceleration changed from 25 g to 2500 g. Observation under the electronic microscope showed that a slimy material covered the cells recovered by gentle gravitational settling and, that the centrifuged cells presented a bare cell wall.

Performance of biofilm carriers in anaerobic digestion of sisal leaf waste leachate Electron. J. Biotechnol.

Three methanogenic biofilm bioreactors were studied to evaluate the performance of three types of carriers. The carrier material were consisted of sisal fibre waste, pumice stone and porous glass beads, and the bioprocess evaluated was the methanogenesis anaerobic digestion of sisal leaf waste leachate. Process performance was investigated by increasing the organic loading rate (OLR) step-wise. The best results were obtained from the bioreactor packed with sisal fibre waste. It had the highest chemical oxygen demand (COD) removal efficiencies in the range of 80-93% at OLRs in the range of 2.4-25 g COD L-1d-1.
