ABSTRACT The activities of enzymes from a number of metabolic pathways have been used as a tool to evaluate the best use of nutrients on fish performance. In the present study the catfish Rhamdia quelen was fed with diets containing crude protein-lipid-carbohydrate (%) as follows: treatment (T) T1: 19-19-44; T2: 26-15-39; T3: 33-12-33; and T4: 40-10-24. The fish were held in tanks of re-circulated, filtered water with controlled temperature and aeration in 2000L experimental units. The feeding experiment lasted 30 days. Alterations in muscular enzymes of horses competing long-distance endurance rides under tropical climate Arq. Bras. Med. Vet. Zootec.

This study tried to monitor the alterations of muscle enzymes activity - creatinokinase (CK), lactate dehydrogenase (LDH), and aspartate aminotransferase (AST) - in a group of horses that participated of 70 and 100km distance rides in five competitions of an annual endurance championship under tropical climate. Pre ride levels (U/l) were 245.13±9.84 for CK, 496.61±14.76 for LDH, and 328.95±8.65 for AST. When compared to these levels, the results revealed a significant decrease in all enzymes activities in the first moment of the rides. Peak levels, significantly different, were reached, immediately after rides by CK (413.591±50.75); 24 hours post rides by LDH (628.61±33.30); and 48 hours after rides by AST (389.89±16.96). The relationship between production of enzymes and pathogenicity of Malassezia pachydermatis strains was studied. Thirty strains of M. pachydermatis isolated from dogs were used: 15 originating from animals with external otitis and 15 obtained from ears of asymptomatic dogs. The samples, obtained by introduction of a sterile swab into the auditory canal after cleaning the auricle with alcohol-ether solution, were seeded onto Sabouraud dextrose agar with chloramphenicol (100mg/l) and were incubated at 37°C for up to two weeks. The performance and the morphology of intestinal mucosa of broilers fed mannan-oligosaccharides (MOS) and enzymes (E) from one to 21-day-old were evaluated using 750 one-day-old chicks, assigned to a 2 x 2 + 1 factorial design - two levels of MOS (0 and 0.1%), two levels of E (0 and 0.05%) plus an antibiotic positive control diet - performing five treatments of five replications each one. MOS x E interaction was significant for both duodenal (P<0.002 and P<0.002) and ileal (P<0.04 and P<0.05) perimeters and heights of villi, being the values lower in the mucosa of birds fed non-supplemented diets. Enzymatic activity of endophytic bacterial isolates of Jacaranda decurrens Cham. (Carobinha-do-campo) BABT

The objectives of this work was to isolate endophytic bacteria from Jacaranda decurrens Cham. and screening of some enzymes of biotechnological interest. Ten (10) bacterial species were isolated and identified from the leaves and steams. All the isolates presented enzymatic activity, which was ranked as follows: proteolytic (60%) and amilolytic activity (60%), lipolytic (40%), esterasic (40%). However, cellulolytic and pectinolytic activities were not detected. This is the first report on the isolation and identification of endophytic bacteria from Jacaranda decurrens Cham.

Enzymatic programs for broilers BABT

Two trials were conducted on floor pens to evaluate the performance of broilers fed with diets contained different carbohydrases enzyme programs at different ages. Trial 1- The body weight gain (BWG) was affected by the Negative Control (NC) treatment (P<0.05). However, when the birds were fed with the NC diet + Amilase (AMY)+Xilanase (XYL), the BWG improved and reached the same level as the Positive Control (PC). Trial 2- The birds received enzymes supplementation in the total phase and others only in the grower phase.The ME reduction by 120 kcal/kg increased the feed conversion ratio (FCR) (P<0.05) when compared to PC and none of the enzyme programs was able to recover the performance. Liquid-liquid extraction of enzymes by affinity aqueous two-phase systems BABT

From analytical to commercial scale, aqueous two-phase systems have their application in the purification, characterization and study of biomaterials. In order to improve the selectivity of the systems, the biospecific affinity ligands were introduced. In the affinity partitioning aqueous two-phase system, have many enzymes been purified. This review discusses the partitioning of some enzymes in the affinity aqueous two-phase systems in regard to the different ligands, including reactive dyes, metal ions and other ligands. Some integration of aqueous two-phase system with other techniques for more effective purification of enzymes are also presented.

Enzymatic activities for digestion of proteins and carbohydrates were compared among three organs of the digestive system of Pimelodus maculatus in two reservoirs with different trophic conditions during the winter of 2006. The aim was to test the hypothesis that enzymatic activity for the digestion of proteins and carbohydrates differed among organs and that such activities differ between the trophic state of the environment. Influence of stationary and bioreactor cultivation on Lentinula edodes (berk) pegler lignocellulolitic activity BABT

This work aimed to study the stationary and periodically mixed culture of L. edodes to the production of lignocellulolitic enzymes activity. LE 95/17, LE 96/22 and Leax strains were incubated in 25 g of eucalyptus sawdust substrate in Erlenmeyer flasks in stationary culture at 25º C and in a bioreactor with four complete rotations daily at 25º C and 3% CO2. The samples were collected at 8, 11, 14, 17 and 20 days after the incubation. Oxidative and hydrolytic enzymes analyses were performed. Temperature-dependent alterations in metabolic enzymes and proteins of three ecophysiologically different species of earthworms BABT

The effects of varying temperatures (12 - 44° C) on the specific activity of cytoplasmic malate dehydrogenase ((cMDH), mitochondrial malate dehydrogenase (mMDH) and lactate dehydrogenase (LDH) of some earthworms (Metaphire posthuma, Perionyx sansibaricus and Lampito mauritii) were studied. The effects of different temperatures on supernatant and mitochondrial protein contents were also investigated. The specific activities of cMDH, mMDH and LDH of the earthworms decreased gradually as a function of increasing temperature from 12 to 44°C. Higher metabolic energy was needed to maintain the activity at low temperatures. Hence, the earthworms showed increased enzyme specific activity at low temperatures. ABSTRACT This study aimed to isolate a potential probiotic amylolytic strain from the gut of jundiá catfish to improve carbohydrate digestibility in fish. Two of 31 strains isolated from the foregut of Rhamdia quelen were able to grow on starch-agar medium and were considered amylolytic. The strain that presented higher amylolytic potential, based on a qualitative amylase assay, was chosen. The strain was phenotypically characterized and analysed to determine bile and pH tolerance and extracellular quantitative amylase activity. Isolation of lipase producing fungi from palm oil Mill effluent (POME) dump sites at Nsukka BABT

In this study, twelve fungal lipase producing strains belonging to Aspergillus, Penicillium, Trichoderma and Mucor genera were isolated from palm oil mill effluent composts. The Aspergillus spp. were more frequent (42%) and was present in all the samples assayed. Mucor sp. was the least encountered (8.3%).The lipase producing profile showed that Trichoderma (8.07-8.24 u/mL) and Aspergillus (6.25 -7.54 u/mL) spp. were the highest lipase producers while Mucor (5.72 u/mL) was the least.

Two related thrombin-like enzymes present in Bothrops atrox venom BJMBR

This article describes the presence of two new forms of a thrombin-like enzyme, both with apparent molecular masses of 38 kDa, in Bothrops atrox venom. Both share the ability to cleave fibrinogen into fibrin and to digest casein. Both present identical Km on the substrate BApNA. Their N-terminal amino acid sequences are identical for 26 residues, sharing 80% homology with batroxobin and flavoxobin. Two groups of monoclonal antibodies (mAbs) raised against the purified enzyme forms recognized different epitopes of the putative corresponding enzymes present in B. atrox crude venom. Screening of actinomycetes from earthworm castings for their antimicrobial activity and industrial enzymes BJM

Actinomycetes from earthworm castings were isolated and screened for their antimicrobial activity and industrial enzymes. A total of 48 isolates were obtained from 12 samples of earthworm castings. Highest numbers of isolates were recovered from forest site (58.33 %) as compared to grassland (25%) and agricultural land (16.66%). The growth patterns, mycelial coloration of abundance actinomycetes were documented. ABSTRACT The multi-enzyme complex (crude extract) of white rot fungi Pleurotus ostreatus, Pleurotus eryngii, Trametes versicolor, Pycnosporus sanguineus and Phanerochaete chrysosporium were characterized, evaluated in the hydrolysis of pretreated pulps of sorghum straw and compared efficiency with commercial enzyme. Electrochemical biosensors in pharmaceutical analysis BJPS

Given the increasing demand for practical and low-cost analytical techniques, biosensors have attracted attention for use in the quality analysis of drugs, medicines, and other analytes of interest in the pharmaceutical area. Biosensors allow quantification not only of the active component in pharmaceutical formulations, but also the analysis of degradation products and metabolites in biological fluids. Thus, this article presents a brief review of biosensor use in pharmaceutical analysis, focusing on enzymatic electrochemical sensors.

Antioxidant system response induced by aluminum in two rice cultivars Braz. J. Plant Physiol.

The antioxidant defense system response was evaluated in two rice cultivars (Oryza sativa L.), Fernandes (CNA-1158) and Maravilha (CNA-6843-1), treated with toxic levels of aluminum. After exposure to aluminum we determined plant growth, H2O2 and O2•- contents, lipid peroxidation, antioxidant enzymes activities and ascorbate and dehydroascorbate contents. Al predominantly accumulated in roots of both cultivars but it reduced root and shoot growth only in the Maravilha cultivar. Treatment with aluminum resulted in a reduction of 84 and 60% in the levels of H2O2 in the roots of the cultivars Maravilha and Fernandes, respectively, and of 26% in the levels of O2•- only in the roots of Fernandes cultivar. The impact of supplemental UV-B (sUV-B) radiation and heavy metals: Cd (68 µmol.kg-1 soil) and Ni (68 µmol.kg-1 soil) singly and in combination was studied on photosynthetic and non-photosynthetic pigments, metabolites, enzyme activities, biomass accumulation and the net primary productivity of spinach (Spinacia oleracea L. var. All Green), a common leafy vegetable plant.
