The present study investigated the effect of electrolysis on the biomineralization capacities of juveniles of the mollusk Pinctada margaritifera for the first time. Size-selected individuals from two groups, “Medium” and “Large”, from a multi-parental family produced in a hatchery system were subjected to electrolysis under a low voltage current over a nine-week experimental period. The growth of the juveniles was individually monitored and assessed weekly by wet weight and shell height measurements. At the end of the experiment, mantle tissue was sampled for biomineralization-related gene expression analysis. Phosphoglucomutase is a key enzyme in glycolysis and has been widely studied in vertebrates and some invertebrates but no molecular information is available in marine invertebrates despite the importance of this marker in ecological and genetical studies. In this work, we isolated a cDNA and the corresponding genomic sequence that encode PGM-2 locus in the Pacific oyster Crassostrea gigas. We used sequences drawn from the database to construct an evolutionary framework for examining the position of mollusc PGM sequences among prokaryotic and eukaryotic homologues and showed that oyster PGM gene organization was closer to vertebrates PGM genes than other invertebrates as previously found in other Lophotrochozoa species. Pacific oyster hatchery production remains highly variable, partially due to inconsistent gamete quality. However, little attention has been paid to oocyte quality in Pacific oyster compared to important vertebrate aquaculture species such as fishes. The aims of the present study were to identify criteria of oocyte and trochophore quality used as predictive tools for embryo developmental success at the D-larval stage. To examine further a previously reported association between amylase gene polymorphism and growth in the Pacific oyster Crassostrea gigas, ecophysiological parameters and biochemical and molecular expression levels of alpha-amylase were studied in Pacific oysters of different amylase genotypes. Genotypes that previously displayed significantly different growth were found to be significantly different for ingestion and absorption efficiency. These estimated parameters, used in a dynamic energy budget model, showed that observed ingestion rates (unlike absorption efficiencies) allowed an accurate prediction of growth potential in these genotypes. This study was designed to identify in the pearl oyster Pinctada margaritifera, used as a bio-accumulator, molecular biomarkers for the presence of heavy metals in the lagoon environment. Pearl oysters were exposed to 2 concentrations (1 and 10 μg L− 1) of cadmium (Cd) and chromium (Cr) compared to a control. Twelve target genes encoding proteins potentially involved in the response to heavy metal contamination with antioxidant, detoxification or apoptosis activities were selected. A semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) assay was developed to estimate tissue-specific expression of two amylase genes in five different tissues of the Pacific oyster Crassostrea gigas and the experimental effects of food level on the cellular, enzymatic and gene expression changes of the digestive gland. We have cloned and characterized for the first time an allograft inflammatory factor 1 (Sn-AIF-1) from the Antarctic sea urchin. We report the cloning of Sn-AIF-1 cDNA and the characterization of its expression in coelomocytes after a bacterial challenge. The cDNA Sn-AIF-1 has a size of 608 bp and encodes a polypeptide of 151 aa. The deduced amino acid sequence has a putative size of 17.430 Da, an isoelectric point of 4.92, and shows 2 elongation factor handlike motifs that normally bind calcium ions. Expression of six selective genes in peripheral blood cells was evaluated as diagnostic biomarkers for enzootic bovine leukosis (EBL) by using 10 EBL and 15 clinically healthy cattle. The clinically healthy cattle generally showed lower gene expression levels. Although wide variations of gene expression were found in some clinical cases of EBL, 4 and 5 among 10 EBL cattle showed higher expression of interleukin 2 receptor gene (IL2R) and Wilms’ tumor gene (WT1), respectively. Expression of IL2R in peripheral blood cells in EBL cattle was statistically increased; however, the lower sensitivity and higher variation in the gene expressions among clinical cases of EBL would be problems as diagnostic biomarkers. |
