Beet curly top resistance in USDA-ARS Ft. Collins Germplasm, 2011 NWISRL
Panella, Leonard W.; Strausbaugh, C.A..
Twenty-two sugar beet (Beta vulgaris L.) lines from the USDA-ARS Ft. Collins sugar beet program were screened for resistance to Beet severe curly top virus (BSCTV) and other closely related Curtovirus species in 2011. Commercial cultivars Monohikari and HM PM90 were included as susceptible and resistant checks, respectively. The curly top evaluation was conducted at the USDA-ARS North Farm in Kimberly, ID which has Portneuf silt loam soil and had been in barley in 2010. The field was plowed both in the fall and the spring, fertilized (80 lb N and 120 lb P2O5/A) on 20 Apr 11, sprayed with Ethotron (2 pt/A), and roller harrowed. The germplasm was planted (density of 142,560 seeds/A) on 16 May. The plots consisted of two rows 10 ft long with 22-in row...

USDA-ARS sugar beet germplasm developed in Salinas, CA, evaluated for rhizomania resistance in Idaho NWISRL
Gillen, A.M.; Strausbaugh, C.A.; Gallian, J..
Thirty-one sugar beet germplasm breeding lines and releases produced by the USDA-ARS sugarbeet program at Salinas, CA, and one line from the USDA-ARS sugarbeet program at East Lansing, MI, were evaluated for resistance to rhizomania under south-central Idaho conditions at Twin Falls, ID. The field had been identified as having Beet necrotic yellow vein virus (BNYVV), which overcomes resistance conditioned by the Rzl gene. Single-row plots 10 ft long with 22-in. spacing were planted on 23 May 05. Plants were thinned to 4 to 6-in. spacing in mid-Jun. The materials were predominantly multigerm; they were hand thinned to single plants during the first week of Jul. The trial layout was a randomized complete block with eight replicates, however, the...

Strausbaugh, C.A., Panella, L.W. 2016. Beet curly top resistance in USDA-ARS plant introduction lines, 2015 NWISRL
Strausbaugh, C.A.; Panella, Leonard W..
Curly top caused by Beet curly top virus (BCTV) is a widespread disease problem vectored by the beet leafhopper in semiarid sugar beet production areas. Host resistance is the primary defense against this problem, but resistance in commercial cultivars is only low to intermediate. In order to identify novel sources of curly top resistance, thirty-one Plant Introduction (PI) Lines were screened in a disease nursery in 2015. The lines were arranged in a randomized complete block design with three replications. A curly top epiphytotic was created by releasing approximately 6 viruliferous beet leafhoppers per plant at the four- to six-leaf growth stage on 24 Jun. Foliar symptoms were evaluated on 13 and 20 Jul using a scale of 0-9 (0 = healthy and 9 =...

Vásquez-Castro,Javier A; Baptista,Gilberto C De; Trevizan,Luiz R P; Gadanha Jr,Casimiro D.
Insect-pest mobility can influence insect susceptibility to the insecticides used to control them. The objective of this work was to evaluate the flight activity of Sitophilus oryzae (L) and Sitophilus zeamais Motsch populations reared on corn and wheat grains, and its relationship with insecticide susceptibility. Unsexed adult insects with ages between 10 and 20 days were placed on a plastic tray. A 40-watt incandescent light bulb was positioned at a 50 cm height relative to the tray. Flight activity was evaluated during 30 min. In the LC50 estimation bioassays, the insects were exposed to dry residues of the organophosphorus insecticide fenitrothion and of the pyrethroid insecticide esfenvalerate on the internal surface of glass vials. Mortality was...

Effect of silicon applied to wheat plants on the biology and probing behaviour of the greenbug Schizaphis graminum (Rond.) (Hemiptera: Aphididae) Neotropical Entomology
Goussain,Marcio M.; Prado,Ernesto; Moraes,Jair C..
The effect of silicon-treated wheat plants (Triticum aestivum L.) on the greenbug, Schizaphis graminum (Rond.) (Hemiptera: Aphididae) was evaluated. Plants were treated with silicon incorporated to the soil and by foliar spraying. Aphid development was evaluated by observing the duration of the pre-reproductive, reproductive and post-reproductive periods, as well as fecundity and longevity. Probing behaviour was investigated by using the DC electrical penetration graphs (EPG) technique and a "honeydew clock". Silica treated plants had a clear adverse effect on aphid development. Stylet penetration was not affected by treatments showing no physical barriers by the plant tissue. However, stylet was withdrawn more often on plants treated with silica,...

Background: Acaricide failure has been on the rise in the western and central cattle corridor of Uganda. In this study, we identified the tick species associated with acaricide failure and determined their susceptibility to various acaricide molecules used for tick control in Uganda. Methods: In this cross sectional study, tick samples were collected and identified to species level from 54 purposively selected farms (from 17 districts) that mostly had a history of acaricide failure. Larval packet test was used to screen 31 tick populations from 30 farms for susceptibility at discriminating dose (DD) and 2 x DD of five panels of commercial acaricide molecules belonging to the following classes; amidine, synthetic pyrethroid (SP), organophosphate (OP) and...

Chloroquine resistance status a decade after : Re-emergence of sensitive Plasmodium falciparum strains in malaria endemic and epidemic areas in Kenya OAK
Wangai, L.N; Kimani, F.T; Omar, S.A; Karanja, S.M; Nderu, D.W; Magoma, G; Mutua, D.
Development and spread of chloroquine (CQ) resistance led to its withdrawal in most malaria endemic countries. In Kenya, this occurred in 1998 when clinical efficacy dropped below 50%. Less than a decade after CQ was removed from routine use in Malawi, the drug has reversed to activity and is again effective for first-line treatment of uncomplicated malaria. There is a probability of a similar reversed activity in Kenya for more 10 years of its absence in uncomplicated Plasmodium falciparum malaria treatment. The present study was aimed at establishing the CQ resistance status in the country, 10 years after its withdrawal, by looking at high malaria transmission zone, Mbita, a malaria endemic area and some malaria epidemic areas of the Kenyan highlands....

Mapping a QTL conferring resistance to Fusarium head blight on chromosome 1B in winter wheat (Triticum aestivum L.) OAK
Nishio, Zenta; Orme, Chihiro; Ito, Miwako; Tabiki, Tadashi; Nagasawa, Koichi; Miura, Hideho.
Fusarium head blight (FHB) is one of the most devastating diseases of wheat (Triticum aestivum L.), and the development of cultivars with FHB resistance is the most effective way to control the disease. Yumechikara is a Japanese hard red winter wheat cultivar that shows moderate resistance to FHB with superior bread-making quality. To identify quantitative trait loci (QTLs) for FHB resistance in Yumechikara, we evaluated doubled haploid lines derived from a cross between Yumechikara and a moderate susceptible cultivar, Kitahonami, for FHB resistance in a 5-year field trial, and we analyzed polymorphic molecular markers between the parents. Our analysis of these markers identified two FHB-resistance QTLs, one from Yumechikara and one from Kitahonami. The...

Sequence similarity between the viral cp gene and the transgene in transgenic papayas PAB
Souza Júnior,Manoel Teixeira; Gonsalves,Dennis.
The Papaya ringspot virus (PRSV) coat protein transgene present in 'Rainbow' and 'SunUp' papayas disclose high sequence similarity (>89%) to the cp gene from PRSV BR and TH. Despite this, both isolates are able to break down the resistance in 'Rainbow', while only the latter is able to do so in 'SunUp'. The objective of this work was to evaluate the degree of sequence similarity between the cp gene in the challenge isolate and the cp transgene in transgenic papayas resistant to PRSV. The production of a hybrid virus containing the genome backbone of PRSV HA up to the Apa I site in the NIb gene, and downstream from there, the sequence of PRSV TH was undertaken. This hybrid virus, PRSV HA/TH, was obtained and used to challenge 'Rainbow', 'SunUp', and an...

Inheritance pattern and selection criteria for resistance to soybean cyst nematode races 3 and 9 PAB
Cervigni,Gerardo Domingo Lucio; Schuster,Ivan; Sediyama,Carlos Sigueyuki; Barros,Everaldo Gonçalves de; Moreira,Maurilio Alves.
The objective of this work was to determine soybean resistance inheritance to Heterodera glycines Ichinohe (soybean cyst nematode - SCN) races 3 and 9, as well as to evaluate the efficiency of direct and indirect selection in a soybean population of 112 recombinant inbred lines (RIL) derived from the resistant cultivar Hartwig. The experiment was conducted in a completely randomized design, in Londrina, PR, Brazil. The estimated narrow-sense heritabilities for resistance to races 3 and 9 were 80.67 and 77.97%. The genetic correlation coefficient (r g = 0.17; p<0.01) shows that some genetic components of resistance to these two races are inherited together. The greatest genetic gain by indirect selection was obtained to race 9, selecting to race 3 due to...

Reaction of lettuce genotypes to Lettuce mosaic virus-Most (LMV-Most) and characterization of the translation factor eIF4E PAB
Moura,Mônika Fecury; Silva,Norberto da; Hoffmann,Maria Isabel Motta; Pavan,Marcelo Agenor; Krause-Sakate,Renate.
Abstract: The objective of this work was to evaluate lettuce genotypes for their reaction to Lettuce mosaic virus (LMV; Most-type, isolate AF-199) and variations of the eukaryotic translation initiation factor eIF4E. All inoculated genotypes were susceptible to LMV, which was detected by RT-PCR using specific primer pairs. However, the accessions 169501, 169501C, 172918A, and 162499 showed late development of symptoms that appeared only on the inoculated leaves. Sequencing of the coding region of eIF4E showed that these genotypes have an eIF4E0 (mol 0 ) standard typical for their susceptibility to LMV, indicating that the phenotype found is not correlated to nucleotide variations in this translation factor.

Reaction of arracacha genotypes to the root soft rot caused by Pectobacterium chrysanthemi PAB
Henz,Gilmar Paulo; Reifschneider,Francisco José Becker; Santos,Fausto Francisco dos.
The purpose of this paper was to screen thirty-two arracacha genotypes for their reaction to root soft rot. Twenty roots of each genotype were inoculated with two Pectobacterium chrysanthemi isolates in a randomized experiment (10 roots/isolate). After inoculation, roots were individually wrapped with PVC film and kept at 26ºC in closed plastic bags. Soft rot lesions were recorded after 36 hours and genotypes were grouped in four classes of susceptibility by cluster analysis: 10 were less susceptible, 16 intermediate, 3 susceptible and 3 very susceptible. All the tested arracacha genotypes showed only variation in the degree of susceptibility.
