A preliminary survey of M. hyopneumoniae virulence factors based on comparative genomic analysis Genet. Mol. Biol.
Ferreira,Henrique Bunselmeyer; Castro,Luiza Amaral de.
Mycoplasma hyopneumoniae is the etiological agent of porcine enzootic pneumonia (PEP), a major problem for the pig industry. The mechanisms of M. hyopneumoniae pathogenicity allow to predict the existence of several classes of virulence factors, whose study has been essentially restricted to the characterization of adhesion-related and major antigenic proteins. The now available complete sequences of the genomes of two pathogenic and one non-pathogenic strain of M. hyopneumoniae allowed to use a comparative genomics approach to putatively identify virulence genes. In this preliminary survey, we were able to identify 118 CDSs encoding putative virulence factors, based on specific criteria ranging from predicted cell surface location or variation between...

Genetic parameters for five traits in Africanized honeybees using Bayesian inference Genet. Mol. Biol.
Padilha,Alessandro Haiduck; Sattler,Aroni; Cobuci,Jaime Araújo; McManus,Concepta Margaret.
Heritability and genetic correlations for honey (HP) and propolis production (PP), hygienic behavior (HB), syrupcollection rate (SCR) and percentage of mites on adult bees (PMAB) of a population of Africanized honeybees were estimated. Data from 110 queen bees over three generations were evaluated. Single and multi-trait models were analyzed by Bayesian Inference using MTGSAM. The localization of the hive was significant for SCR and HB and highly significant for PP. Season-year was highly significant only for SCR. The number of frames with bees was significant for HP and PP, including SCR. The heritability estimates were 0.16 for HP, 0.23 for SCR, 0.52 for HB, 0.66 for PP, and 0.13 for PMAB. The genetic correlations were positive among productive traits...

Molecular characterization of Chaetomium species using URP-PCR Genet. Mol. Biol.
Aggarwal,Rashmi; Sharma,Vandana; Kharbikar,Lalit L.; Renu,.
Chaetomium spp. are common colonizers of soil and cellulose-containing substrates. Seventeen isolates of Chaetomium spp., which included 15 isolates of C. globosum and one each of C. reflexum and C. perlucidum , were genetically characterized with universal rice primers (URP - primers derived from DNA repeat sequences in the rice genome) using polymerase chain reaction (URP-PCR). Out of the 12 URP's used in the study, nine primers were effective in producing polymorphic fingerprint patterns from DNA of Chaetomium spp . Analysis of the entire fingerprint profile using the unweighted pair-group method with arithmetic averages (UPGMA) clearly differentiated C. globosum isolates from C. perlucidum and C. reflexum . One of the primers, URP-2R,...

Expression, purification and DNA-binding activities of two putative ModE proteins of Herbaspirillum seropedicae (Burkholderiales, Oxalobacteraceae) Genet. Mol. Biol.
Souza,André L.F.; Chubatsu,Leda S.; Souza,Emanuel M.; Pedrosa,Fábio O.; Monteiro,Rose A.; Rego,Fabiane G.M.; Rigo,Liu U..
In prokaryotes molybdenum is taken up by a high-affinity ABC-type transporter system encoded by the modABC genes. The endophyte β-Proteobacterium Herbaspirillum seropedicae has two modABC gene clusters and two genes encoding putative Mo-dependent regulator proteins (ModE1 and ModE2). Analysis of the amino acid sequence of the ModE1 protein of H. seropedicae revealed the presence of an N-terminal domain containing a DNA-binding helix-turn-helix motif (HTH) and a C-terminal domain with a molybdate-binding motif. The second putative regulator protein, ModE2, contains only the helix-turn-helix motif, similar to that observed in some sequenced genomes. We cloned the modE1 (810 bp) and modE2 (372 bp) genes and expressed them in Escherichia coli as His-tagged...

Evaluation of the in vivo mutagenic potential of hydroalcoholic extracts of the northern highbush blueberry (Vaccinium corymbosum L. Ericales, Ericaceae) on peripheral blood cells of Swiss mice (Mus musculus Rodentia, Muridae) Genet. Mol. Biol.
Freitas,Patrícia Scotini; Andrade,Sérgio Faloni de; Maistro,Edson Luis.
The northern highbush blueberry (Vaccinium corymbosum L. Ericales, Ericaceae) is very rich in anthocyanins, natural pigments which have strong antioxidant properties and potential health benefits, resulting in the worldwide use the blueberry as a medicinal plant. We investigated the mutagenic potential of simple hydroalcoholic extracts of V. corymbosum acutely administrated by gavage to Swiss mice at doses of 1 g kg-1, 1.5 g kg-1 and 2 g kg-1. Peripheral blood cells were collected 4 h and 24 h post-gavage and assessed by the alkaline comet assay, with further blood samples being collected at 48 h and 72 h for assessment using the micronucleus (MN) assay. Our results show that the V. corymbosum extracts did not induce any statistically significant increase...

Conservation genetics of an endangered Catalonian cattle breed ("Alberes") Genet. Mol. Biol.
Jordana,J.; Piedrafita,J.; Carre,X.; Martell,A..
We biochemically analyzed and characterized the genetic structure of a population in danger of extinction ,"Alberes", a local cattle breed of the Catalonian Pyrenees (Spain and France). Ninety-two individuals were analyzed for five polymorphic genetic loci (Hb, Alb, Tf, Gc and Ptf2). The animals were grouped according to coat color: Fagina Alberes variety (N = 39) and Black Alberes variety (N = 53). The genetic structures and relationships between these subpopulations and one "outgroup" breed ("Bruna dels Pirineus") were analyzed and compared by using F-statistics. We determined that inbreeding in the Alberes breed is not significant, since negative and nonsignificant FIT and FIS values were obtained. The average genetic differentiation between...

Assessment of two medicinal plants, Psidium guajava L. and Achillea millefolium L., in in vitro and in vivo assays Genet. Mol. Biol.
Teixeira,Rosangela de Oliveira; Camparoto,Marjori Leiva; Mantovani,Mário Sérgio; Vicentini,Veronica Elisa Pimenta.
The use of medicinal plants by the general population is an old and still widespread practice, which makes studies of their genotoxicity essential. Psidium guajava L. and Achillea millefolium L. are examples of plants commonly used in popular medicine. P. guajava L. is indicated for diarrhea and also as an antiseptic, while A. millefolium L. is indicated as an analgesic, antispasmodic, digestive, diuretic, antiseptic, astringent, emollient, wound healer and hemorrhoid medication. The aim of this study was to determine the effects of the infusions of these two plant species on chromosomes and the cell cycle. Leaves from the plants were used to prepare infusions, in the same manner as teas, but at two different concentrations. Allium cepa L. root-tip cells...

Molecular identification, phylogeny and geographic distribution of Brazilian mangrove oysters (Crassostrea) Genet. Mol. Biol.
Melo,Aline Grasielle Costa de; Varela,Eduardo Sousa; Beasley,Colin Robert; Schneider,Horacio; Sampaio,Iracilda; Gaffney,Patrick Michael; Reece,Kimberly S.; Tagliaro,Claudia Helena.
Oysters (Ostreidae) manifest a high degree of phenotypic plasticity, whereby morphology is of limited value for species identification and taxonomy. By using molecular data, the aim was to genetically characterize the species of Crassostrea occurring along the Brazilian coast, and phylogenetically relate these to other Crassostrea from different parts of the world. Sequencing of the partial cytochrome oxidase c subunit I gene (COI), revealed a total of three species of Crassostrea at 16 locations along the Brazilian coast. C. gasar was found from Curuçá (Pará state) to Santos (São Paulo state), and C. rhizophorae from Fortim (Ceará state) to Florianópolis (Santa Catarina state), although small individuals of the latter species were also found at Ajuruteua...

Allozyme variation and sociogenetic structure of Polistes satan Bequaert 1940 colonies (Hymenoptera, Vespidae) Genet. Mol. Biol.
Gaspar,Jane Cristina Wehren; López-Uribe,Margarita María; Del Lama,Marco Antônio.
We collected 40 colonies of the wasp Polistes satan in November 1993 (Population A = 23 colonies) and October 1995 (Population B = 17 colonies) from the town of Delfinopólis in the Brazilian state of Minas Gerais. Pupae from these nests were used to assess allozyme variation and genetic relatedness among nestmates. Of the 24 gene loci sampled six showed allozyme polymorphism (Est-2, Pgm-1, 6Pgd, Hk-1, Idh and Pep-A) and all loci except Idh were at Hardy-Weinberg equilibrium. Populations A and B showed distinct intra-loci heterozygosities, but similar average heterozygosity over all loci. Significant inter-population differentiation (Fst = 0.051; p < 0.01) was detected, probably due to genetic drift driven by bottlenecking that occurred due to the first...

AFLP marker linked to water-stress-tolerant bulks in barley (Hordeum vulgare L.) Genet. Mol. Biol.
Altinkut,A.; Kazan,K.; Gozukirmizi,N..
The amplified fragment length polymorphism (AFLP) assay is an efficient method for the identification of molecular markers, useful in the improvement of numerous crop species. Bulked Segregant Analysis (BSA) was used to identify AFLP markers associated with water-stress tolerance in barley, as this would permit rapid selection of water-stress tolerant genotypes in breeding programs. AFLP markers linked to water-stress tolerance was identified in two DNA pools (tolerant and sensitive), which were established using selected F2 individuals resulting from a cross between water-stress-tolerant and sensitive barley parental genotypes, based on their paraquat (PQ) tolerance, leaf size, and relative water content (RWC). All these three traits were previously shown...

Integrated analysis of the critical region 5p15.3–p15.2 associated with cri-du-chat syndrome Genet. Mol. Biol.
Corrêa,Thiago; Feltes,Bruno César; Riegel,Mariluce.
Abstract Cri-du-chat syndrome (CdCs) is one of the most common contiguous gene syndromes, with an incidence of 1:15,000 to 1:50,000 live births. To better understand the etiology of CdCs at the molecular level, we investigated theprotein–protein interaction (PPI) network within the critical chromosomal region 5p15.3–p15.2 associated with CdCs using systemsbiology. Data were extracted from cytogenomic findings from patients with CdCs. Based on clinical findings, molecular characterization of chromosomal rearrangements, and systems biology data, we explored possible genotype–phenotype correlations involving biological processes connected with CdCs candidate genes. We identified biological processes involving genes previously found to be associated with CdCs,...

Genetic variability in Amerindian populations of Northern Argentina Genet. Mol. Biol.
Dejean,Cristina B.; Crouau-Roy,Brigitte; Goicoechea,Alicia S.; Avena,Sergio A.; Carnese,Francisco R..
The allelic variability of four dinucleotide microsatellites located in the HLA region (MOGc, D6S265, MIB, and TNFa) was analyzed in 67 individuals representing three Amerindian populations of the Argentine Gran Chaco: Toba, Wichi and Chorote. Genomic DNA was prepared from peripheral blood and DNA was extracted using the standard phenol-chloroform procedure. Alleles were identified by PCR, using an end-labelled reverse oligonucleotide primer (fluorescent 6 - Fam labeling). Despite the low number of samples studied, a high level of gene diversity was observed in each population and for each locus. Moreover, the mean number of alleles was 7.7, 5.3, 10.0, and 7.0 at loci MOGc, D6S265, MIB and TNFa, respectively. Differentiation tests between pairs of...

Restriction-modification systems in Mycoplasma spp Genet. Mol. Biol.
Brocchi,Marcelo; Vasconcelos,Ana Tereza Ribeiro de; Zaha,Arnaldo.
Restriction and Modification (R-M) systems are present in all Mycoplasma species sequenced so far. The presence of these genes poses barriers to gene transfer and could protect the cell against phage infections. The number and types of R-M genes between different Mycoplasma species are variable, which is characteristic of a polymorphism. The majority of the CDSs code for Type III R-M systems and particularly for methyltransferase enzymes, which suggests that functions other than the protection against the invasion of heterologous DNA may exist. A possible function of these enzymes could be the protection against the invasion of other but similar R-M systems. In Mycoplasma hyopneumoniae strain J, three of the putative methyltransferase genes were clustered...

Tandem repeat distribution of gene transcripts in three plant families Genet. Mol. Biol.
Maia,Luciano Carlos da; Souza,Velci Queiróz de; Kopp,Mauricio Marini; Carvalho,Fernando Irajá Félix de; Oliveira,Antonio Costa de.
Tandem repeats (microsatellites or SSRs) are molecular markers with great potential for plant genetic studies. Modern strategies include the transfer of these markers among widely studied and orphan species. In silico analyses allow for studying distribution patterns of microsatellites and predicting which motifs would be more amenable to interspecies transfer. Transcribed sequences (Unigene) from ten species of three plant families were surveyed for the occurrence of micro and minisatellites. Transcripts from different species displayed different rates of tandem repeat occurrence, ranging from 1.47% to 11.28%. Both similar and different patterns were found within and among plant families. The results also indicate a lack of association between genome size...

Evidence of high inbreeding in a population of the endangered giant anteater, Myrmecophaga tridactyla (Myrmecophagidae), from Emas National Park, Brazil Genet. Mol. Biol.
Collevatti,Rosane G.; Leite,Kelly C.E.; Miranda,Guilherme H.B. de; Rodrigues,Flavio H.G..
We report the genetic structure, relatedness and mating structure of a population of the endangered giant anteater Myrmecophaga tridactyla Linnaeus, 1758 in the Emas National Park, Brazil, based on variability at five microsatellite loci. Additionally, we addressed the hypothesis that the M. tridactyla population studied has low levels of polymorphism and high levels of inbreeding and relatedness and that animals with overlapping home range are highly related. All five microsatellite loci displayed low levels of polymorphism and of expected and observed heterozygosity. The low level of polymorphism and high inbreeding showed by the population studied may be the outcome of high mortality and reduction in population size due to recurrent fire events in the...

Association of homocysteine and methylene tetrahydrofolate reductase (MTHFR C677T) gene polymorphism with coronary artery disease (CAD) in the population of North India Genet. Mol. Biol.
Tripathi,Rajneesh; Tewari,Satyendra; Singh,Prabhat Kumar; Agarwal,Sarita.
The implications of the methylene tetrahydrofolate reductase (MTHFR) gene and the level of homocysteine in the pathogenesis of coronary artery disease (CAD) have been extensively studied in various ethnic groups. Our aim was to discover the association of MTHFR (C677T) polymorphism and homocysteine level with CAD in north Indian subjects. The study group consisted of 329 angiographically proven CAD patients, and 331 age and sex matched healthy individuals as controls. MTHFR (C677T) gene polymorphism was detected based on the polymerase chain reaction and restriction digestion with HinfI. Total homocysteine plasma concentration was measured using immunoassay. T allele frequency was found to be significantly higher in patients than in the control group. We...

Analysis of the interaction between human kidney anion exchanger 1 and kanadaptin using yeast two-hybrid systems Genet. Mol. Biol.
Wongthida,Phonphimon; Akkarapatumwong,Varaporn; Limjindaporn,Thawornchai; Kittanakom,Saranya; Keskanokwong,Thitima; Eurwilaichitr,Lily; Yenchitsomanus,Pa-thai.
Kidney anion exchanger adaptor protein (Kanadaptin) is a protein which interacts with the cytoplasmic N-terminal domain of kidney anion exchanger 1 (kAE1) and was first detected in mice using the yeast two-hybrid system and was also found to co-localize with kAE1 in rabbit a-intercalated cells. Impaired trafficking of human kAE1 can result in the kidney disease-distal renal tubular acidosis (dRTA), and defective interaction between human kAE1 and kanadaptin may cause this trafficking impairment and be the basis for dRTA pathogenesis. However, it is unknown whether kAE1 can really interact with kanadaptin in humans. We have thus investigated the interaction between human kAE1 and human kanadaptin by using both Gal4 and LexA yeast two-hybrid systems. It was...

Survey of transposable elements in sugarcane expressed sequence tags (ESTs) Genet. Mol. Biol.
Rossi,Magdalena; Araujo,Paula Gonçalves; Van Sluys,Marie-Anne.
The sugarcane expressed sequence tag (SUCEST) project has produced a large number of cDNA sequences from several plant tissues submitted or not to different conditions of stress. In this paper we report the result of a search for transposable elements (TEs) revealing a surprising amount of expressed TEs homologues. Of the 260,781 sequences grouped in 81,223 fragment assembly program (Phrap) clusters, a total of 276 clones showed homology to previously reported TEs using a stringent cut-off value of e-50 or better. Homologous clones to Copia/Ty1 and Gypsy/Ty3 groups of long terminal repeat (LTR) retrotransposons were found but no non-LTR retroelements were identified. All major transposon families were represented in sugarcane including Activator (Ac),...
