IgG antibodies against phospholipase A2 from Crotalus durissus terrificus: cross-reaction with venoms from Bothrops species from Argentina J. Venom. Anim. Toxins incl. Trop. Dis.
Rodríguez,JP; De Marzi,MC; Maruñak,S; Teibler,P; Acosta,O; Malchiodi,EL; Leiva,LC.
We examined the ability of IgG anti-crotalic PLA2 to cross-react with Bothrops spp. venoms, from snakes found in the northeast of Argentina. Immunoblotting and ELISA tests showed that IgG anti-crotalic PLA2 recognize antigens of bothropic venoms. Indirect hemolytic activity tests showed that the quantity of antibodies that neutralized 50% of Crotalus durissus terrificus venom (ED50: 2.1 mg IgG anti-crotalic PLA2/100 µg of venom) were also able to neutralize venom from other snakes in the following proportion: 34% of B. alternatus, 18% of B. diporus and 12% of B. jararacussu. Likewise, direct PLA2 activity neutralization tests showed a similar cross-neutralization pattern including 56% of B. alternatus, 29% of B. diporus and 30% of B. jararacussu. In...

Clinical and electrocardiographic evaluation during experimental toad poisoning in dogs J. Venom. Anim. Toxins incl. Trop. Dis.
Camplesi,AC; Sakate,M; Simão,NMB; Marucio,R; Mota,FCD; Moya-Araujo,CF.
Accidents involving toad poisoning are frequent and dogs are the most common victims; they become poisoned by biting or ingesting a toad. When released in the organism, the venom is absorbed by both the oral mucosa and the digestive tract, initiating its toxic action. The aim of this work was to evaluate the clinical and electrocardiographic aspects of dogs subjected to experimental toad poisoning, as well as their response to treatment with propranolol. Twenty dogs were divided into two groups, a control group (n = 5) and a poisoned group (n = 15). After general anesthesia, the control group received a placebo, while the poisoned group received a venom aliquot through an orogastric tube. Results were tested through multivariate analysis (p < 0.05). The...

Deep sequencing analysis of toad Rhinella schneideri skin glands and partial biochemical characterization of its cutaneous secretion J. Venom. Anim. Toxins incl. Trop. Dis.
Shibao,Priscila Yumi Tanaka; Cologna,Camila Takeno; Morandi-Filho,Romualdo; Wiezel,Gisele Adriano; Fujimura,Patricia Tiemi; Ueira-Vieira,Carlos; Arantes,Eliane Candiani.
Abstract Background: Animal poisons and venoms are sources of biomolecules naturally selected. Rhinella schneideri toads are widespread in the whole Brazilian territory and they have poison glands and mucous gland. Recently, protein from toads’ secretion has gaining attention. Frog skin is widely known to present great number of host defense peptides and we hypothesize toads present them as well. In this study, we used a RNA-seq analysis from R. schneideri skin and biochemical tests with the gland secretion to unravel its protein molecules. Methods: Total RNA from the toad skin was extracted using TRizol reagent, sequenced in duplicate using Illumina Hiseq2500 in paired end analysis. The raw reads were trimmed and de novo assembled using Trinity. The...

Comparison of different diagnostic tests in dogs uninfected and naturally infected with visceral leishmaniasis J. Venom. Anim. Toxins incl. Trop. Dis.
Sassaki,CY; Colodel,MM; Ferreira,I; Nogueira,FS; Lucheis,SB; Langoni,H; Rocha,NS.
Uninfected dogs (n = 10) and those naturally infected with leishmaniasis (n = 10) were subjected to several diagnostic tests, namely: hemoculture, polymerase chain reaction (PCR) of hemoculture, indirect immunofluorescence (RIFI), cytological examination of lymph node aspirate, culture of lymph node aspirate and PCR of lymph node aspirate. RIFI - followed by PCR of lymph node aspirate culture - presented more positive results in infected dogs than in uninfected ones. In infected animals, RIFI was more effective than PCR of lymph node aspirate culture. There was no statistical difference in positivity between RIFI and hemoculture; lymph node aspirate culture/cytological examination of lymph node aspirate and PCR of hemoculture; and between PCR of lymph node...

Documented bites by a yellow sac spider (Cheiracanthium punctorium) in Italy: a case report J. Venom. Anim. Toxins incl. Trop. Dis.
Papini,R.
In Italy reports of human envenomations by yellow sac spiders have been sporadic. Since increasing clinical information would improve understanding of the danger of yellow sac spiders to humans, we report the case of a 7-year-old child and her father bitten by a documented Cheiracanthium punctorium. They developed acute persistent pain with local skin signs and numbness, and required emergency treatment. The father recovered completely within 1 to 2 hours and the child within 3 to 4 days after treatment, probably as a result of spontaneous evolution. Clinicians should be aware of the risks and immediate management of spider bites.

Production of Trichophyton mentagrophytes antigens and their characterization in mice J. Venom. Anim. Toxins incl. Trop. Dis.
Venturini,J; Rossi-Ferreira,R; Arruda,MSP.
The participation of dermatophytic antigens in the host-parasite balance is still poorly understood. One of the difficulties encountered by researchers is the lack of dominant and specific antigens that can be used in such studies. In order to contribute to a better understanding of this aspect of infection, the present study identifies antigen fractions obtained from exoantigen and cytoplasmic extracts of Trichophyton mentagrophytes. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) revealed the presence of 13 proteins in the exoantigen extract, whose molecular weight ranged from 12.5 to 90 kDa. The cytoplasmic extract contained 18 protein fractions ranging from 11 to 110 kDa. Immunoblotting showed the presence of immunodominant...

What is the optimum concentration of m-cresol in antivenoms? J. Venom. Anim. Toxins incl. Trop. Dis.
Abd-Elsalam,MA; Abdoon,N; Al-Ahaidib,MS.
Antivenoms against snake and scorpion envenomations are usually equine in nature and composed mostly of F(ab')2; additionally, phenol and m-cresol are mainly employed for their preservation. Although there is no study on this subject, m-cresol is utilized by most manufacturers in a concentration that ranges from 0.15 to 0.35 g%. Decreasing the concentration of m-cresol to its minimal effective level may protect victims from its toxic effects and keep the antivenom stable during its shelf life without forming any aggregates. In the present work, different concentrations of m-cresol, ranging from 0.1 to 0.35 g%, were used with some selected batches of snake and scorpion antivenoms. A low concentration of 0.15 g% showed an acceptable preserving result that...

Integrins, cancer and snake toxins (mini-review) J. Venom. Anim. Toxins incl. Trop. Dis.
Rádis-Baptista,G..
Integrins encompass a family of transmembrane heterodimeric proteins of adhesion that maintain cells attached to other cells and to the extracellular matrix (ECM). Integrins work as bi-directional mechanotransducers, conveying mechanical signal from outside to inside the cell through a cascade of phosphorylation signals. On the other hand, the signal from inside to outside controls the strength and affinity of integrin adhesion. As proteins of focal contact, integrins are involved in diverse cell functions, such as cell activation, migration, growth, and survival. In the development of neoplastic disease and metastatic tumor, integrins can influence cancer invasiveness and progression, as well as mediate the formation of new blood vessels (angiogenesis)....

Effect of phospholipase A2 inhibitors during infection caused by Leishmania (Leishmania) amazonensis J. Venom. Anim. Toxins incl. Trop. Dis.
Bordon,Maria L. A. C.; Laurenti,Márcia D.; Ribeiro,Susan Pereira; Toyama,Marcos H.; Toyama,Daniela de O.; Passero,Luiz Felipe D..
Abstract Background: Lipid metabolites play an important role in parasite differentiation and virulence. Studies have revealed that Leishmania sp. uses prostaglandins to evade innate barriers, thus enabling the parasites to survive inside immune cells. Despite the role of the enzyme Phospholipase A2 (PLA2) in prostaglandins production, few studies have investigated the role of parasite PLA2 during the interaction between L. (L.) amazonensis and the host (in vitro and in vivo) immune cells. Methods: In the present work, the leishmanicidal effect of PLA2 inhibitors, methyl arachidonyl fluorophosphonate (MAFP), bromoenol lactone (BEL) and aristolochic acid (AA) were investigated in vitro (promastigote and intracellular amastigote forms of L.
