Overexpression of Interleukin 2 Receptor, Thymidine Kinase and Immunoglobulin-Associated Alpha-1 Messenger RNA in a Clinical Case of Enzootic Bovine Leukosis OAK
TAWFEEQ, Mohammad Monir; TAGAWA, Michihito; ITOH, Yuuki; SUGIMOTO, Kazuya; KOBAYASHI, Yoshiyasu; INOKUMA, Hisashi; 古林, 与志安; 猪熊, 壽.
A 49-month-old Holstein cow with anorexia, tachypnea, enlarged peripheral lymph nodes, and difficulty standing up was suspected of bovine leukosis. Hematological examination revealed lymphocytosis with the presence of neoplastic cells. Increased total lactate dehydrogenase (LDH) activity, isozymes of LDH-2 and LDH-3 activities and thymidine kinase activity were observed. Cytological findings of fine needle aspiration of subiliac lymph nodes indicated lymphosarcoma. Histopathology and antibody analysis confirmed the diagnosis of enzootic bovine leukosis, a B-cell bovine lymphoma caused by bovine leukemia virus. Gene expressions known as biomarkers of hematopoietic neoplasia in human were also examined in the present case. Increased messenger RNA expression...

Calf Form Bovine Leukosis with Lameness in a Holstein Heifer OAK
TAWFEEQ, Mohammad Monir; MIURA, Saori; NAKANISHI, Yuuki; SUGIMOTO, Kazuya; KOBAYASHI, Yoshiyasu; FURUOKA, Hidefumi; INOKUMA, Hisashi; 古林, 与志安; 猪熊, 壽.
A 12-month-old Holstein heifer with anorexia, lameness, and enlargement of peripheral lymph nodes was suspected of having bovine leukosis. Although lymphocytosis was not observed, cytology of fine needle aspirate from a superficial cervical node, and increased serum lactate dehydrogenase and thymidine kinase activities, strongly suggested lymphosarcoma. Increased numbers of mononuclear cells as well as mitotic cells were observed in synovial fluid collected from swollen joints. Pathological examination confirmed B-cell calf form bovine leukosis and joint swelling related to neoplastic cell infiltration. Both interleukin-2 receptor and thymidine kinase 1 genes were highly expressed in cells from superficial cervical lymph node aspirate.

Isolation of a cotton NADP(H) oxidase homologue induced by drought stress PAB
NEPOMUCENO,ALEXANDRE LIMA; STEWART,JAMES MCD; OOSTERHUIS,DERRICK; TURLEY,RICKIE; NEUMAIER,MORMAN; FARIAS,JOSÉ RENATO BOUÇAS.
The aim of this study was to identify and isolate genes that are differentially expressed in four selected cotton (Gossypium hirsutum L.) genotypes contrasting according to their tolerance to water deficit. The genotypes studied were Siokra L-23, Stoneville 506, CS 50 and T-1521. Physiological, morphological and developmental changes that confer drought tolerance in plants must have a molecular genetic basis. To identify and isolate the genes, the mRNA Differential Display (DD) technique was used. Messenger RNAs differentially expressed during water deficit were identified, isolated, cloned and sequenced. The cloned transcript A12B15-5, a NADP(H) oxidase homologue, was up regulated only during the water deficit stress and only in Siokra L-23, a drought...

Transgenic tomato plants expressing the antigen gene PfCP-2.9 of Plasmodium falciparum PAB
Kantor,Mihail; Sestras,Radu; Chowdhury,Kamal.
The objective of this work was to obtain transgenic tomato plants expressing the PfCP-2.9 protein (a chimera of the antigens MSP1 and AMA1 of Plasmodium falciparum). Cotyledons of seven-day-old tomatoes, cultivar Summers, were transformed via Agrobacterium tumefaciens. Transgenic expression in the T0 plants was verified in the DNA extracted from fruits. PCR analysis was used to test the presence of the gene of interest in the T1 generation. Reverse transcriptase PCR provided evidence of gene expression at the RNA level, and Western blot analysis confirmed the presence of the protein of interest in the T1 plants. This is the first report of successful transformation with the expression of a malaria antigen (PfCP-2.9) in transgenic tomato plants from the T0...

Molecular biology of baculovirus and its use in biological control in Brazil PAB
Castro,Maria Elita Batista de; Souza,Marlinda Lobo de; Sihler,William; Rodrigues,Júlio Carlyle Macedo; Ribeiro,Bergmann Morais.
Baculoviruses are insect viruses found mainly in Lepidoptera. The family Baculoviridae is taxonomically divided in two genera, Nucleopolyhedrovirus and Granulovirus, which differ by occlusion body morphology. NPVs (Nucleopolyhedroviruses) have polyhedrical inclusion bodies (PIBs) containing multiple viral particles, while GVs (Granuloviruses) appear to be generally single particles occluded in oval shaped occlusion bodies. During the life cycle, two different viral progenies are produced: BV (Budded Virus) and PDV (Polyhedra Derived Virus), which are essential for the infectious process and virus propagation in host cells. Baculoviruses are being used for pest control and they are especially safe due to their specificity and invertebrate-restricted host...

Differential gene expression, induced by salicylic acid and Fusarium oxysporum f. sp. lycopersici infection, in tomato PAB
Amaral,Daniel Oliveira Jordão do; Lima,Marleide Magalhães de Andrade; Resende,Luciane Vilela; Silva,Márcia Vanusa da.
The objective of this work was to determine the transcript profile of tomato plants (Lycopersicon esculentum Mill.), during Fusarium oxysporum f. sp. lycopersici infection and after foliar application of salicylic acid. The suppression subtractive hybridization (SSH) technique was used to generate a cDNA library enriched for transcripts differentially expressed. A total of 307 clones was identified in two subtractive libraries, which allowed the isolation of several defense-related genes that play roles in different mechanisms of plant resistance to phytopathogens. Genes with unknown roles were also isolated from the two libraries, which indicates the possibility of identifying new genes not yet reported in studies of stress/defense response. The SSH...

Wu,Y; Liang,H; Zhang,H; Pi,JS; Pan,AL; Shen,J; Pu,YJ; Du,JP.
ABSTRACT Ovoinhibitor (OIH) is the main proteinase inhibitor in the egg white. In the present study, real-time quantitative PCR and Western-Blot were used to analyze different expression pattern of OIH in ovaries as a candidate gene of reproductive traits in Jingjiang ducks (JJ ducks) and Shaoxing ducks (SX ducks) during three laying stages. To study the polymorphism of the OIH gene in those two duck populations, we designed five pairs of primers to detect SNPs of exon 3-5, 5-6, 14-16 and intron 7, 9 of the OIH gene by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and DNA pool sequencing methods. The results showed that OIH expression increased during the laying stage in the ovaries of both duck breeds. The relative...

Changes in gene expression during drying and imbibition of desiccation sensitive Magnolia ovata (A. St.-Hil.) spreng. seeds Rev. bras. sementes
José,Anderson C.; Ligterink,Wilco; Davide,Antonio Claudio; Silva,Edvaldo A. Amaral da; Hilhorst,Henk W.M..
Seeds of Magnolia ovata were dried to different water contents to assess the viability and transcript abundance of genes related to seed development, cell cycle, cytoskeleton and desiccation tolerance. The expression of development, cell cycle and cytoskeleton relative genes (ABI3, CDC2-like and ACT2) alone could not explain the germination behaviour of M. ovata seeds in relation to drying damage. Irrespective of their initial water content, the seeds performed in the same way during the initial period of germination and the deleterious effects of desiccation only occurred in later stages. Expression of PKABA1, sHSP17.5 and 2-Cys-PRX did not show a relationship with desiccation. However, the expression patterns of PKABA1 and sHSP17.5 suggested the...
