Chromosomal characterization of three native and one cultivated species of Lathyrus L. in Southern Brazil Genet. Mol. Biol.
Battistin,Alice; Biondo,Elaine; Coelho,Liliana Gressler May.
Mitotic metaphase chromosomes and interphase nuclei of nine populations of three South American species of Lathyrus (L. pubescens, L. nervosus and L. crassipes) and six populations of the cultivated species L. odoratus were analyzed. All populations had 2n = 2x = 14 chromosomes. There were significant differences among populations within each species and among species in the number of metacentric, submetacentric and subtelocentric chromosomes, the number and location of secondary constrictions, chromosome length (longest and shortest), total haploid complement, arm ratio, and centromeric index. L. odoratus showed the highest tendency towards karyotype symmetry whereas the three South American species showed a moderate tendency towards asymmetry, with L....

Phenotypic plasticity in colonizing populations of Drosophila subobscura Genet. Mol. Biol.
Pegueroles,Glòria; Mestres,Francesc; Argemí,Mercè; Serra,Lluís.
The phenotypic plasticity of some quantitative traits of two colonizing populations of Drosophila subobscura (Davis and Eureka, California) was studied. Temperature effects and the effect of rearing in the laboratory were studied. Laboratory rearing during four generations at 18ºC significantly increased the wing and tibial length. This increase was similar to that obtained when the flies were reared at 13ºC during two generations. The low temperature environment can be considered more stressful for females than for males, as shown by the increase of phenotypic variance. The two populations analyzed had great phenotypic plasticity in spite of the genetic bottleneck during the colonization event. Our study shows that keeping flies for a relatively short time...

Comparison of similarity coefficients based on RAPD markers in the common bean Genet. Mol. Biol.
Duarte,Jair Moura; Santos,João Bosco dos; Melo,Leonardo Cunha.
The alterations caused by eight different similarity coefficients were evaluated in the clustering and ordination of 27 common bean (Phaseolus vulgaris L.) cultivars analyzed by RAPD markers. The Anderberg, simple matching, Rogers and Tanimoto, Russel and Rao, Ochiai, Jaccard, Sorensen-Dice, and Ochiai II's coefficients were tested. Comparisons among the coefficients were made through correlation analysis of genetic distances obtained by the complement of these coefficients, dendrogram evaluation (visual inspection and consensus fork index - CIC), projection efficiency in a two-dimensional space, and groups formed by Tocher's optimization procedure. The employment of different similarity coefficients caused few alterations in cultivar classification, since...

Genetic control of plant architecture in the common bean (Phaseolus vulgaris L.) Genet. Mol. Biol.
Teixeira,Flavia França; Ramalho,Magno Antonio Patto; Abreu,Ângela de Fátima Barbosa.
More erect plant architecture has been a goal in the development of bean cultivars. Aiming to obtain more information about genetic control of traits related to plant architecture, this work was carried out between August 1995 and July 1997 in the southern and Alto São Francisco regions, in the State of Minas Gerais, Brazil. Initially, analyses were performed with individual plants of parents and different segregant generations from the crosses Carioca-MG x H-4 and Carioca x FT-Tarumã. In these experiments, besides degree of erectness, other traits were evaluated: ramification degree, internode length, internode diameter and height of insertion of the first pod. Mean and variance components and heritability at an individual level were estimated. Later,...

Transplacental inhibitory effect of carrot juice on the clastogenicity of cyclophosphamide in mice Genet. Mol. Biol.
Gimmler-Luz,Maria Clara; Cardoso,Valesca Veiga; Sardiglia,Cassius Ugarte; Widholzer,Débora da Silva.
Genetic damage during the prenatal period can provoke important neoplastic alterations and other diseases in postnatal life. Beta-carotene (ßC) is considered to be one of the most important anticarcinogens in the diet and can protect mammalian cells against genotoxic events. As carrots are important dietary source of ßC, we decided to test the effect of fresh carrot juice (CaJ) on cyclophosphamide (CP)-induced genotoxicity in maternal and fetal erythropoietic tissues. The treatment with CaJ started on the 7th day of the pregnancy of BALB/c female mice. We observed, on the 16th gestational day, that this treatment did not modify the spontaneous frequency of micronucleated polychromatic erythrocytes (mPCE) in the bone marrow of the females nor in the livers...

Effects of sorghum (sorghum bicolor L.) root exudates on the cell cycle of the bean plant (phaseolus vulgaris L.) root Genet. Mol. Biol.
Hallak,Angela Maria Gattás; Davide,Lisete Chamma; Souza,Itamar Ferreira.
Two experiments were conducted to test the allelopathic effect of sorghum (Sorghum bicolor L.) root exudates on bean (Phaseolus vulgaris L.) cell division. Research was conducted in the greenhouse of the Wistock Agricultural Research Institute of Minas Gerais State (EPAMIG) and in a laboratory of the Federal University of Lavras (UFLA). Sorghum variety BR-601 and bean variety Carioca MG were used. The exudate, called sorgoleone (SGL), was obtained by methylene chloride and acetic acid extraction from sorghum roots seven days after sowing on Petri dishes, and refrigerated until use. Solutions of 0.01, 0.05, 0.10, and 0.15 mM were prepared using Johanson solution as the SGL solvent. Seven-day-old bean seedlings grown in vermiculite in a greenhouse were...

Is the American Zebu really Bos indicus? Genet. Mol. Biol.
Meirelles,Flávio V.; Rosa,Artur J.M.; Lôbo,Raysildo B.; Garcia,Joaquim M.; Smith,Lawrence C.; Duarte,Francisco A.M..
The American continent was colonized in the 16th century by Europeans who first introduced cattle of Bos taurus origin. Accounts register introduction of Bos indicus cattle into South America in the 19th and continuing through the 20th century, and most reported imports were males derived from the Indian subcontinent. In the present study we show, by using mitochondrial DNA (mtDNA) polymorphism, major participation of matrilineages of taurus origin in the American Zebu purebred origin, i.e., 79, 73 and 100% for the Nellore, Gyr and Brahman breeds, respectively. Moreover, we have created a restriction map identifying polymorphism among B. taurus and B. indicus mtDNA using three restriction enzymes. Results are discussed concerning American Zebu origins and...

Evidence for biological inheritance of the eosinophil response to internal parasites in southeastern Brazil Genet. Mol. Biol.
Conti,Fátima; Dal' Colletto,Glória M. Duccini; Feitosa,Mary Furlan; Krieger,Henrique.
One hundred and seventy-seven individuals belonging to 120 complete or incomplete nuclear families from Bambui, in the State of Minas Gerais, southeastern Brazil, were studied in order to examine causes of variation in the eosinophil rate among subjects infested by intestinal worms with an extra-digestive cycle. Segregation analysis without correction for skewness showed that the hypothesis of the presence of an additive major gene was consistent with the data, although a dominant, recessive, or a multifactorial hypothesis could not account properly for the observed significant familial aggregation. The most parsimonious correction for skewness showed similar results, but could not distinguish between dominant and recessive models, although co-dominance...

Estimates of direct, maternal and grandmaternal genetic effects for growth traits in Gobra cattle Genet. Mol. Biol.
Diop,M.; Dodenhoff,J.; Van Vleck,L.D..
Estimates of genetic parameters for birth (N = 3909), weaning (N = 3425), yearling (N = 2764) and final (N = 2144) weights were obtained from the records of Gobra cattle collected at the Centre de Recherches Zootechniques de Dahra, Senegal. Three animal models were fitted to obtain estimates by REML using an average information (AI) approach. Model 1 considered random direct, maternal genetic and maternal permanent environmental effects. In model 2, a general grandmaternal effect was added to the random effects considered in model 1, and in model 3, the general grandmaternal effect was divided into grandmaternal genetic and grandmaternal permanent environmental effects. All models allowed covariances among genetic effects. The inclusion of grandmaternal...

Anticlastogenic effect of vitamin C on cisplatin in vivo Genet. Mol. Biol.
Antunes,Lusânia M. Greggi; Darin,Joana D.C.; Bianchi,Maria de Lourdes P..
The ability of vitamin C (VC) to protect against the clastogenic action of the chemotherapeutic agent cisplatin (DDP, cis-diamminedichloroplatinun II) in rat bone marrow cells was evaluated. DDP was administered to Wistar rats either alone or after treatment with VC. The rats were treated with VC (50, 100 or 200 mg/kg body weight) by gavage 10 min before the administration of DDP (5 mg/kg body weight, ip) and then sacrificed 24 h after treatment. VC significantly reduced (by about 70%) the clastogenicity of DDP in rat bone marrow cells. The antioxidant action of VC presumably modulates the clastogenic action of DDP.

Effects of turmeric and its active principle, curcumin, on bleomycin-induced chromosome aberrations in Chinese hamster ovary cells Genet. Mol. Biol.
Araújo,Maria Cristina P.; Dias,Francisca da Luz; Kronka,Sergio N.; Takahashi,Catarina S..
Naturally occurring antioxidants have been extensively studied for their capacity to protect organisms and cells from oxidative damage. Many plant constituents including turmeric and curcumin appear to be potent antimutagens and antioxidants. The effects of turmeric and curcumin on chromosomal aberration frequencies induced by the radiomimetic agent bleomycin (BLM) were investigated in Chinese hamster ovary (CHO) cells. Three concentrations of each drug, turmeric (100, 250 and 500 mg/ml) and curcumin (2.5, 5 and 10 mg/ml), were combined with BLM (10 mg/ml) in CHO cells treated during the G1/S, S or G2/S phases of the cell cycle. Genetic structure of different cat populations in Europe and South America at a microgeographic level: importance of the choice of an adequate sampling level in the accuracy of population genetics interpretations Genet. Mol. Biol.
Ruiz-Garcia,Manuel.
The phenotypic markers, coat color, pattern and hair length, of natural domestic cat populations observed in four cities (Barcelona, Catalonia; Palma Majorca, Balearic Islands; Rimini, Italy and Buenos Aires, Argentina) were studied at a microgeographical level. Various population genetics techniques revealed that the degree of genetic differentiation between populations of Felis catus within these cities is relatively low, when compared with that found between populations of other mammals. Two different levels of sampling were used. One was that of "natural" colonies of cat families living together in specific points within the cities, and the other referred to "artificial" subpopulations, or groups of colonies, inhabiting the same district within a city....

Chromosomal characterization of cultured populations of Chilean coho salmon (Oncorhynchus kistuch) Genet. Mol. Biol.
Colihueque V.,Nelson.
Chromosomal characterization of coho salmon samples from three fish farms in southern Chile (Polcura, Castro and Coyhaique) was carried out in order to compare their chromosome constitutions. All populations had a 2n = 60; however, Polcura and Coyhaique had a different chromosome arm number (NF = 110; 40m + 10sm + 10st/t) than Castro (NF = 108; 40m + 8sm + 12st/t). Variation in NF was due to chromosome pair 25, which was submetacentric in Coyhaique and Polcura, but subtelocentric in Castro. In all karyotypes, a large submetacentric chromosome pair exhibited an interstitial secondary constriction in the short arm. The observed variability in chromosome arm number agrees with previous reports for O. kisutch, and in this particular case it seemed to be caused...

The nucleation of microtubules in Aspergillus nidulans germlings Genet. Mol. Biol.
Andrade-Monteiro,Cristina de; Martinez-Rossi,Nilce M..
Microtubules are filaments composed of dimers of alpha- and beta-tubulins, which have a variety of functions in living cells. In fungi, the spindle pole bodies usually have been considered to be microtubule-organizing centers. We used the antimicrotubule drug Benomyl in block/release experiments to depolymerize and repolymerize microtubules in Aspergillus nidulans germlings to learn more about the microtubule nucleation process in this filamentous fungus. Twenty seconds after release from Benomyl short microtubules were formed from several bright (immunofluorescent) dots distributed along the germlings, suggesting that microtubule nucleation is randomly distributed in A. nidulans germlings. Since nuclear movement is dependent on microtubules in A. nidulans...

Genetic effect on apgar score Genet. Mol. Biol.
Franchi-Pinto,Carla; Colletto,Glória Maria Duccini Dal; Krieger,Henrique; Beiguelman,Bernardo.
Intraclass correlation coefficients for one- and five-min Apgar scores of 604 twin pairs born at a southeastern Brazilian hospital were calculated, after adjusting these scores for gestational age and sex. The data support a genetic hypothesis only for 1-min Apgar score, probably because it is less affected by the environment than 4 min later, after the newborns have been under the care of a neonatology team. First-born twins exhibited, on average, better clinical conditions than second-born twins. The former showed a significantly lower proportion of Apgar scores under seven than second-born twins, both at 1 min (17.5% vs. 29.8%) and at 5 min (7.2% vs. 11.9%). The proportion of children born with "good" Apgar scores was significantly smaller among twins...

Bleomycin sensitivity in patients with familial and sporadic polyposis: a pilot study Genet. Mol. Biol.
Sales,Magaly M.; Lucca,Edmundo J. de; Yamashita,Seizo; Saad,Luis Henrique Cury.
Human peripheral blood lymphocytes from 10 patients with familial adenomatous polyposis (FAP) showed a significantly higher incidence of chromatid breaks when compared to cells from 10 normal individuals, after exposure to bleomycin (BLM) during the G2 phase. However, no significant increase in bleomycin sensitivity was observed in lymphocytes from 10 patients with sporadic adenomatous polyps (AP) vs. 10 normal individuals (P = 0.67). Individuals that exhibited an average number of chromatid breaks per cell higher than 0.80 were considered sensitive to the drug. No control showed susceptibility to BLM, as compared to 3 out of 20 patients.
