To establish an efficient regeneration system for Anthurium andreanum cv Rubrun, seeds from plant spadixes were germinated on a medium supplemented with 2.2 muM BA. After 2 weeks, 74% of the seeds germinated and four weeks later, micro-cuttings from these plantlets were subcultured on a medium containing 4.4 muM BA and 0.05 muM NAA. On average, 3.6 shoots per explant were obtained. Four weeks old in vitro plants from germinated seeds and the plantlets obtained from micro-cuttings, showed callus proliferation at the stem base. These tissues were subcultured on a medium supplemented with 8.9 muM BA and 2.7 muM NAA. After 6 weeks of culture, about 43.8 plantlets per square cm of callus were obtained. Anatomical studies showed the organogenic nature of these... |

Thirty rice cultivars were evaluated for salinity tolerance during the seedling stage and were divided into five tolerance groups including tolerant (T), moderately tolerant (MT), moderately susceptible (MS), susceptible (S) and highly susceptible (HS) which comprised 5, 10, 9, 4 and 2 cultivars respectively. Genetic diversity of all rice cultivars was evaluated using random amplified polymorphic DNA (RAPD) and simple sequence repeats (SSR) markers. The cultivars were evaluated for polymorphisms after amplification with 20 random decamer primers and 20 SSR primer pairs. A total of 161 RAPD markers and 190 SSR alleles were produced which revealed 68.94% and 89.47% polymorphism respectively. Mean genetic similarity coefficient was 0.82 for RAPD and 0.70 for... |

In the present study we examined the possibility of propagating different Cyclamen species (C. africanum Boiss. and Reut., C. cilicium Boiss. and Heldr., C. coum Mill., C. hederifolium Ait., C. persicum Mill., C. purpurascens Mill.) including some of their subspecies and cultivars in vitro using explants of adult plants. For this purpose two protocols have been applied to eleven genotypes combined with mostly four explant types (placentas with ovules, leaves, petioles and peduncles). The use of these protocols has given rise to either somatic embryo-like structures and/or adventitious shoots in all genotypes. This way it was possible to propagate each of the examined genotypes in vitro using explants of adult plants in a time less than one year. These... |

Morpho-physiological attributes exhibited in response to drought hardening at the end of the growing season of Eucalyptus globulus Labill under nursery conditions were studied to evaluate the effect of three drought hardening treatments in morpho-physiological traits used as suitable indicators of drought hardiness, such as, plant growth, root growth potential, plant water relationships and survival. Freezing resistance of drought hardened plants was also studied in order to evaluate cross hardening effects in cuttings of Eucalyptus globulus Labill. Drought hardening consisted in induced water stress by watering restriction, until plant stem xylem water potentials (Ψpd) reached to-0.2, -1.3 and -2.4 MPa. Two water stress-rewatering cycles were... |

The removal of contaminants in slow sand filters occurs mainly in the colmation layer or schmutzdecke - a biologically active layer consisting of algae, bacteria, diatoms and zooplankton. A ripening period of 6 - 8 weeks is required for this layer to form, during which time filter performance is sub-optimal. In the current study, an environmental scanning electron microscope was used to visualise the ripening process of a pilot-scale slow sand filter over a period of eight weeks. To achieve this, sand particles were removed at weekly intervals and observed for biofilm development. Biological mechanisms of removal in slow sand filtration are not fully understood. A visualisation of the colonisation process would enhance the knowledge and understanding of... |

The aim of this study, was to design and validate 16S rRNA targeted oligonucleotide genus specific primers for amplifying the predominant members of gut flora using polymerase chain reaction. Primers were validated against human faecal samples. Gut flora of a normal individual was compared with that of two diseased individuals. Our observations showed that the genera Lactobacillus, Bacteroides, Peptococcus, Bifidobacterium, and E. coli were invariably present in all studied subjects however, the absence of butyrate producing bacteria Ruminococcus and Peptostreptococcus were significant. Presence of the members of the genus, Campylobacter in both the diseased samples were also unusual. |

The forest industry in Uruguay has grown considerably during the last decade. Eucalyptus plantations account for 74% of the forested land, with Eucalyptus globulus being the most widely distributed species. This industry is dedicated exclusively to the production of wood without exploiting the by-products (leaves and small branches). Eucalyptus leaves are known to contain important amounts of essential oils composed primarily of 1,8-cineole (1,3,3-trymethyl-2-oxabicyclo[2.2.2]octane). In this work, the biotransformation of 1,8-cineole, is achieved using a native bacterium (Rhodococcus sp.) which was isolated from the soil of Eucalyptus forest. A 98% of bioconversion was achieved. Three different optically pure compounds were obtained, and they were... |

The batch removal of copper (II) ions from aqueous solution under different experimental conditions using Ulva reticulata was investigated in this study. The copper (II) uptake was dependent on initial pH and initial copper concentration, with pH 5.5 being the optimum value. The equilibrium data were fitted using Langmuir and Freundlich isotherm model, with the maximum copper (II) uptake of 74.63 mg/g determined at a pH of 5.5. The Freundlich model regression resulted in high correlation coefficients and the model parameters were largely dependent on initial solution pH. At various initial copper (II) concentrations (250 to 1000 mg/L), sorption equilibrium was attained between 30 and 120 min. The copper (II) uptake by U. reticulata was best described by... |

Background Industrial food processing induces protein glycation modifications and toxic advanced glycation end products (AGEs) which affect human health. Therefore, it is of interest to monitor AGEs in food processing. The present study was carried out to investigate the influence of lotus seedpod oligomeric procyanidin (LSOPC) concentrations, solution pH value and metal ions on AGE formation by heat treatment of lactose-lysine model solutions. Ne-(carboxymethyl) lysine (CML), as one of the common AGEs was also determined by HPLC-MS/MS in this experiment. Results The results showed that LSOPC can inhibit the formation of AGEs effectively at higher concentrations, lower temperature, and it can reverse the promotion function of metal ions because of its high... |

Arylalkylamine N-acetyltransferase (AA-NAT) is critical enzyme in Melatonin (MLT) biosynthesis for MLT regulating the animal seasonal breeding. In this study, DNA sequencing methods were applied to detect the polymorphisms of the AA-NAT gene in 179 Chinese sheep belonging to two non-seasonal reproduction breeds and two seasonal reproduction breeds. One mutation at exon 3 (NM_001009461:c.486A > G) was firstly described at the sheep AA-NAT locus. Hence, we described the SmaI PCR-RFLP method for detecting EX3 486A > G mutation, frequencies of the AA-NAT-G allele varied from 0.871 to 0.908 in two non-seasonal reproduction breeds and 0.517 to 0.578 in two seasonal reproduction breeds. The associations of SmaI polymorphism with estrus... |

A large number of micropropagated plantlets of banana, Musa acuminata var. Nanjanagudu Rasabale (NR), that were developed from axillary shoot bud explants over 10 years ago were screened for genetic variation, if any, using RAPD (Random Amplified Polymorphic DNA) and ISSR (Inter-Simple Sequence Repeats) markers. Of the 4000 in vitro plantlets, 11 were used for screening that involved shoot cultures with distinct variation in morphological characteristics (morphotypes). Similarly, the mother maintained in the field was also subjected for genetic analysis. Out of the 50 RAPD and 25 ISSR primers screened, 30 RAPD and 5 ISSR primers produced totally 424 clear, distinct and reproducible band classes resulting in a total of 5088 bands where the banding patterns... |

Sesame (Sesamum indicum L.) is one of the oldest oil crops and is widely cultivated in Asia and Africa. To determine the level of genetic diversity in relation to geographical origins and morphological characteristics, a total of 96 accessions have been collected from different parts of the world and were analyzed using AFLP techniques. Twenty-one primer pairs generated a total of 445 bands and among them 157 (35%) were polymorphic. Using UPGMA clustering analysis method based on the similarity coefficient, accessions were separated into two major groups. The first group mostly consists of Eastern Asian origin and another group consists of South Asian origin. Sub-clusters separated the accessions and form distinct diversity among groups. Considering the... |
