Protein detection in spermatids and spermatozoa of the butterfly Euptoieta hegesia (Lepidoptera) Biocell
Mancini,Karina; Dolder,Heidi.
This study was undertaken to detect protein components in both sperm types of the butterfly Euptoieta hegesia. These spermatozoa possess complex extracellular structures for which the composition and functional significance are still unclear. In the apyrene sperm head, the proteic cap presented an external ring and an internal dense content; basic proteins were detected only in external portions. In the tail, the paracrystalline core of mitochondrial derivatives and the axoneme are rich in proteins. The extratesticular spermatozoa are covered by a proteic coat, which presented two distinct layers. In eupyrene spermatozoa, acrosome and nucleus were negatively stained, probably because of their high compaction. In the tail, there is no paracrystalline core...

Purification of C-phycocyanin from Spirulina platensis in aqueous two-phase systems using an experimental design BABT
Antelo,Francine Silva; Costa,Jorge Alberto Vieira; Kalil,Susana Juliano.
C-phycocyanin from Spirulina platensis was purified in aqueous two-phase systems (ATPS) of polyethylene glycol (PEG)/potassium phosphate, varying the molar mass of the PEG. Results using a full factorial design showed that an increase in the concentration of salt and decrease in the concentration of PEG caused an increment in the purification factor for all the ATPS studied. Optimization of the conditions of the purification was studied using a central composite rotatable design for each molar mass of PEG. The ATPS composed of 7% (w/w) PEG 1500 or 4% (w/w) PEG 8000 (g/gmol) and 23 or 22.5% (w/w) of phosphate resulted a purification factor of 1.6-fold for C-phycocyanin, with total and 57% recovery, respectively. Process conditions were optimized for the...

Study of potential impacts of using sewage sludge in the amendment of desert reclaimed soil on wheat and jews mallow plants BABT
Mazen,Ahmed; Faheed,Fayza A; Ahmed,Atef F.
This investigation was conducted to study the impacts of using sewage sludge at different concentrations (0, 10, 25, 50, 75%) in amendment of desert reclaimed soil properties and some physiological aspects in wheat and jews mallow plants. Generally adding sewage sludge to desert soil improved the soil texture, raised the organic matter contents, water holding capacity and lowered pH value. The contents of NPK gradually increased as the ratio of sewage sludge increased. The fresh and dry weights and biosyntheses of pigment contents of the variously treated test plants were increased by increasing the sewage sludge levels in the soil. Also, total carbohydrate and protein contents of sewage sludge-treated test plants were positively affected. With respect to...

Biochemical and morphological changes during the growth kinetics of Araucaria angustifolia suspension cultures BABT
Santos,André Luis Wendt dos; Silveira,Vanildo; Steiner,Neusa; Maraschin,Marcelo; Guerra,Miguel Pedro.
Embryogenic cultures of Araucaria angustifolia were established in a BM liquid medium supplemented with 2 µM 2,4-dichlorophenoxyacetic acid, 1 µM 6-benzylaminopurine and 1 µM kinetin (BM2) and in a BM medium free of growth regulators (BM0). During 42 days in culture, the cell growth pattern of both cultures was similar. The pH of the culture medium of both BM0 and BM2 underwent progressive reduction during culture time. For both the embryogenic cultures a preferential uptake of glucose in the late stages of cell growth kinetics was observed. The extracellular protein content was similar for both the embryogenic cultures. Acetocarmine and Evan's blue double stain showed major differences for early somatic embryo organisation, in which only the embryogenic...

Alterations in proteins and amino acids of the Nile cyanobacteria Pseudanabaena limnetica and Anabaena wisconsinense in response to industrial wastewater pollution BABT
El-Sheekh,Mostafa Mohamed; El-Otify,Ahmed Mohamed; Saber,Hani.
The effect of industrial wastewater on the Nile cyanobacteria Pseudanabaena limnetica and Anabaena wisconsinense was investigated. The data showed that P. limnetica was more sensitive to pollution than A. wisconsinense. The treatments with different levels of wastewater exerted pronounced reductions in protein and amino acids content. SDS-PAGE analysis revealed that the cyanobacteria grown in the industrial wastewater showed induction in the synthesis of certain polypeptides and repression of others. The treatments of P. limnetica with wastewater stimulated the appearance of six protein bands with molecular masses of 28, 30, 31, 32, 58 and 97 kDa. The same treatments caused the disappearance of 20, 38 and 56 kDa. The structural protein pattern of the...

Santos,Neube Michel dos; Costa,Roberto Germano; Madruga,Marta Suely; Medeiros,Ariosvaldo Nunes de; Albuquerque,Caroline Lima Cavalcante de; Queiroga,Rita de Cássia Ramos do Egypto.
The objective of the present work was to quantify the constitution and composition chemistry of the precooked goat buchada produced in the state of Paraíba, Brazil. The goat buchada presented considerable yield in relation to the live weight. High protein content was observed in the products analyzed. The lipid content presented high variation as result of the lack of standardization of products produced in the state and no control on parameters such as age, weight and breed of the slaughtered animals was observed. Although no difference (P<0.05) in the lipid content was observed, the highest percentages were observed in samples from Remígio and Barra de Santa Rosa, with 14.52 and 15.90%, respectively, as result of the addition of high amounts of...

Compositional changes of proteins and amino acids in germinating coffee seeds BABT
Shimizu,Milton Massao; Mazzafera,Paulo.
Endosperm is the main reserve tissue in coffee seeds. Coffee (Coffea arabica L.) seeds were germinated for six weeks and qualitative and quantitative changes in amino acids and proteins were investigated. The total content of free amino acids were reduced during germination, however, protein content remained constant. SDS-PAGE profiles showed that legumin-like proteins became less stained in the last weeks. Asparagine, glutamic acid, aspartic acid, alanine and lysine were the major free amino acids, although serine and glutamine were also significant. Except for tyrosine, which increased with germination, all other amino acids were reduced. Analysis of the amino acid composition of the total soluble protein showed glutamic acid/glutamine and glycine as the...

Sodium Chloride-Induced leaf senescence in Hydrocotyle bonariensis Lam. and Foeniculum vulgare L. BABT
Haddad,Claudia R. B.; Mazzafera,Paulo.
Leaf senescence induced by sodium chloride was studied in Hydrocotyle bonariensis Lam. and Foeniculum vulgare L. Both species belong to Umbelliferae family, however only H. bonariensis grows spontaneously in sandy soils of coastal regions (saline soils). Leaves of plants receiving nutrient solutions containing different concentrations of NaCl were evaluated for fresh and dry weights and chlorophyll content. The denaturating electrophoretic profiles of leaf proteins were also studied. Sodium chloride changed the protein profile of F. vulgare and hastened the leaf senescence of both the species. However, plants of H. bonariensis receiving 599 mM NaCl lasted longer than F. vulgare. Therefore, the occurrence of H. bonariensis in saline soils might be related...
