Background In sheep breeding, there are situations where relationships recorded at the farm among pedigrees such as parent-offspring, full-sibs or half-sibs need to be tested. A panel of 28 microsatellite (MST) markers was tested to provide accurate pedigree information and resolve the common problem of significant error in pedigree records in Merino sheep. Three different flocks of Australian Merino sheep were investigated. A private farm flock represents a flock with no record availability. Many gene therapy strategies require transfer of high-molecular weight DNA into human cells. To enable clinical trials, these vectors need to be produced on a large scale and at low cost. The production of effective high-capacity vectors like HSV-amplicons and helper-dependent adenoviral vectors is difficult to up-scale, so new inexpensive vectors are needed for the efficient delivery of high-molecular weight DNA to human cells. Enhancement of wanlongmycin production by nitrogen ion beam implantation Electron. J. Biotechnol.

In an attempt to obtain an industrial strain with higher yield of wanlongmycin, the wild strain Streptomyces griseovariabilis GAAS2507 was mutated by a novel mutagen, nitrogen ion beam with energy of 20 kilo electron volts (KeV) and dose ranging from 7.80 x 10(14) to 2.86 x 10(15) ions/cm². One mutant strain WN939 was obtained. Its yield of wanlongmycin reached 271.24 µg/mL, which was 82.10% higher than that of the wild strain. The mutant strain WN939 was relatively stable for the production of wanlongmycin through six successive transfers of cultures and a repeat fermentation in a 30 L fermentor. Improved isolation of good-quality total RNA from the optic stalk of Mud crab, Scylla paramamosain Electron. J. Biotechnol.

An improved and efficient protocol was developed based on the TaKaRa RNAiso Plus Kit (Code: D9108A) for isolating good-quality total RNA from the optic stalk of mud crab, Scylla paramamosain. The protocol was based on the Trizol method with modifications. The carapace overlapping the optic stalk was retained with RNA in regular protocol. In order to remove the abundant deposition correlative with the carapace which makes the isolation of RNA particularly difficult, 5M potassium acetate solution (pH = 6.0) was added before the precipitation of RNA, and the temperature of RNA deposition was also decreased to -70ºC to ensure the stabilization of RNA. Efficient transformation of Penicillium chrysogenum mediated by Agrobacterium tumefaciens LBA4404 for cloning of Vitreoscilla hemoglobin gene Electron. J. Biotechnol.

The vgb and bleomycin resistance genes could be efficiently transferred into the filamentous fungus Penicillium chrysogenum under help of T-DNA of Agrobacterium tumefaciens LBA4404 and the transferred genes were integrated at a chromosomal locus of P. chrysogenum. Transformation using A. tumefaciens LBA4404 could be enhanced in the presence of acetosyringone (AS) when being carried out in the conidiaand mycelium. The efficiencies of transformation could be improved up to 10 folds compared with conventional techniques for transformation of P. chrysogenum protoplasts.

Generic skills are skills that are common to all graduates rather than specific for a particular field of study. They include communication and problem-solving skills and the ability to work successfully in teams. To produce graduates with these skills, university courses must include exercises that encourage development of these skills and assessment procedures which reward achievement in these areas. The influence of centrifugation on Zymomonas mobilis aggregation Electron. J. Biotechnol.

Flocculent Zymomonas mobilis breaks down in smaller flocs and individual cells when centrifuged. The main consequence of it is an increase in the dispersion of the sample, suggesting that the influence of the centrifugal force on the aggregation of cells is worth to study. The experiments showed that the degree of dispersion varied between 30% and 100% when the centrifugal acceleration changed from 25 g to 2500 g. Observation under the electronic microscope showed that a slimy material covered the cells recovered by gentle gravitational settling and, that the centrifuged cells presented a bare cell wall.

Optimization of the citric acid production by Aspergillus niger through a metabolic flux balance model Electron. J. Biotechnol.

Idiophase, the citric acid producing stage of Aspergillus niger was mathematically modeled to identify required genetic manipulations to optimize citric acid production rate. For this reason, a consistent picture of cell functioning had to be achieved. The transient idiophase nature was established by stoichiometric analysis. The main intracellular fluxes were computed by application of material and physiological constraints (ATP, reduction equivalents, proton motive force) at culture time 120 hours.
