Background In sheep breeding, there are situations where relationships recorded at the farm among pedigrees such as parent-offspring, full-sibs or half-sibs need to be tested. A panel of 28 microsatellite (MST) markers was tested to provide accurate pedigree information and resolve the common problem of significant error in pedigree records in Merino sheep. Three different flocks of Australian Merino sheep were investigated. A private farm flock represents a flock with no record availability. Two other flocks were maintained under good managements of full keeping records and being selected for high and low parasite resistances. Many gene therapy strategies require transfer of high-molecular weight DNA into human cells. To enable clinical trials, these vectors need to be produced on a large scale and at low cost. The production of effective high-capacity vectors like HSV-amplicons and helper-dependent adenoviral vectors is difficult to up-scale, so new inexpensive vectors are needed for the efficient delivery of high-molecular weight DNA to human cells. The vgb and bleomycin resistance genes could be efficiently transferred into the filamentous fungus Penicillium chrysogenum under help of T-DNA of Agrobacterium tumefaciens LBA4404 and the transferred genes were integrated at a chromosomal locus of P. chrysogenum. Transformation using A. tumefaciens LBA4404 could be enhanced in the presence of acetosyringone (AS) when being carried out in the conidiaand mycelium. Idiophase, the citric acid producing stage of Aspergillus niger was mathematically modeled to identify required genetic manipulations to optimize citric acid production rate. For this reason, a consistent picture of cell functioning had to be achieved. The transient idiophase nature was established by stoichiometric analysis. The main intracellular fluxes were computed by application of material and physiological constraints (ATP, reduction equivalents, proton motive force) at culture time 120 hours.
