Enhancing me - hope or hype? Nature Precedings
Pete Moore.
Many people talk about human enhancement as if it is about to happen. Some suggest that it already has. To join in the debate you need to distinguish between social, personal and species enhancement. Social and personal are certianly with us - but talk of species enhancement leans more to hype than hope.

A unique avenue to control drug resistant pathogens: Experimental evidences Nature Precedings
Sribir Sen.
Drug resistant bacterial pathogens are increasing in the environment due to multiple reasons. If any drug resistant pathogen grows in a host, the host will face death treat because there is no way to resist their growth in the host system. Eventuality is death in almost all cases. Reports on the mechanisms of drug resistance in bacteria are many. However, there is no solution for these cases. The present work is providing an important understanding to fight against drug resistant pathogens and that can be used in gastrointestinal diseases.

Increased regeneration efficiency of _Brassica napus_ L. cultivars Star, Westar and Cyclone from hypocotyle and cotyledonary explants Nature Precedings
Israr Khan; Waqar Ali; Zahir Ali Takar; Arfa Farooqi; Sikandar Khan; Muhammadwaheed Akhtar.
The comparative organogenesis of _Brassica napus_ L cultivars Cyclone, Star and Westar was studied. The cotyledonary explants gave a higher response to all the combinations of 0.5 mg/L 2,4-D and BAP (0.5, 1.0,1.5 and 2.0 mg/L} used for optimizing the conditions for callus induction. The best mean weight and mean length of callus was obtained at 0.5 mg/L 2,4-D and 1.5mg/L BAP for Star cotyledonary explants. For the complete plant regeneration the new method of exposing the explants culture to Growth regulator free medium was performed. The method was applicable to both hypocotyl and cotyledonary explants. The Shoot Induction Frequency for hypocotyl (6-34%) in the three cultivars is higher than the cotyledonary explants (3-23%). The method is speedy and...

Novel NIR Spectroscopy Correlation Approach to Amino Acid Analysis of Soybean Proteins for Composition Improvements. (Version 2.0) Nature Precedings
Valentin Prisecaru; I. C. Baianu.
Amino acid NIR calibrations were developed in our Physical Chemistry of Foods Laboratory of the University of Illinois at Urbana for three selected amino acid groups that include essential amino acids for identified soybean accessions. Conventional “wet chemistry” analytical methods are time-consuming and costly. As a result, soybean breeders and researchers have an imperative need to utilize faster and less expensive methods. NIR Spectroscopy is a rapid and inexpensive method for composition analysis for academia and industry. Recent advancements in instrumentation design, such as the application of the Diode Array (DA) technique and the Fourier Transform (FT) IR and NIR techniques, have significantly improved overall instrument...

Cell-mediated xenograft rejection, including NK cells and CD8+ CTL, is a major obstacle in successful pig-to-human xenotransplantation. Human CD8+ CTL and NK cells display high cytotoxicity for pig cells, mediated at least in part by the Fas/FasL pathway. To prevent cell-mediated xenocytotoxicity, a membrane-bound form of human FasL (mFasL) was generated as an inhibitor for CTL and NK cell cytotoxicity that could not be cleaved by metalloproteinase to produce putative soluble FasL. We produced two healthy transgenic pigs harboring the mFasL gene via somatic cell nuclear transfer (SCNT). In a cytotoxicity assay using transgenic clonal cell lines and transgenic pig ear cells, the rate of CD8+ CTL-mediated cytotoxicity was significantly reduced in transgenic...

Newer Methods of Nanoparticle Synthesis: Nitroimidazole properties with Nanometal oxides in Polymer Cages as Drug-Biomarker Monitors Nature Precedings
Rakesh Sharma.
Nitroimidazoles are radiosensitizers and hypoxia detecting chemosensitizers. Intially nitroimidazoles were single dose antibiotic drugs. Recently, nitroimidazole derivatives have emerged as multifunctional “drug-biomarker monitors” chemical compounds with importance in treatment of tumors, monitoring hypoxia and safer imaging contrast agents to monitor the therapeutic progress. Additionally, the number of nitroimidazole derivatives is growing and identified their importance as multifunctional nanoparticles. The nanoparticle synthesis scheme of nitroimidazole was proposed as nitroimidazole caged with paramagnetic metal oxide inside polymer coating and labeled with marker radiolebels or biomolecules. We report the step by step...

DNA^+Pro^: an Improved Progressive Multiple Sequence Alignment Algorithm for Evolutionary Analysis Using Combined DNA-Protein Sequences Nature Precedings
Xiaolong Wang; Shuang-yong Xu; Deming Gou.
Alignment of DNA and protein sequences is a basic tool in the study of evolutionary, structural and functional relationship among macromolecules. Present sequence alignment methods are somewhat error-prone, often producing systematic bias. Errors in sequence alignments sometimes lead to subsequent misinterpretation of evolutionary, structural and functional information in genes, proteins and genomes. In traditional sequence alignment algorithms, alignments of DNA and protein sequences are conducted separately. It has been long believed that the phylogenetic signal disappears more rapidly from DNA sequences than from encoded proteins. It is therefore generally preferable to align sequences at the amino acid level. Here we present a new...

A Reaction Route Approach to Flux Balance Analysis Nature Precedings
Ilie Fishtik.
Background: Flux balance and network-based pathway analyses are theoretical tools aimed to find optimal steady state flux distributions in a metabolic network subject to additional constraints on the rates of the reaction steps. Although these methods are mathematically accurate, there are several physicochemical and computational aspects that are questionable and misleading. In particular, it is well known that the flux balance analysis may result in multiple flux distributions for the same objective function.

Results: The flux balance and network-based pathway analyses are reformulated in terms of reaction routes (RRs), a theoretical framework that has been developed by Horiuti over 50 years ago. Not only does the...

The Importance of Patents for Scientists Nature Precedings
Sue Scott.
Patents have become a hot topic and the subject of much media attention. If your main source of information about patents is the newspapers, you could be forgiven for having an extremely distorted view of what patents are, what they do, and what benefits and problems are associated with them. All scientists want to be able to improve the world in some way, whether by contributing in a fundamental way to the fund of human knowledge or by delivering new and valuable technology. If you are a scientist making inventions, it is increasingly important that you should have an understanding of the importance of patents in the modern world, and should have sufficient understanding of the patent system to make sure that your aims for your inventions are...

Signatures of spatial organizations – From the universe of proteins to the universe in general Nature Precedings
Aditya Mittal.
Pattern generation and recognition in 3-D space have been of wide interest in fields varying from atomic and molecular structures to organization of celestial objects. Our thoughts towards investigating patterns (and elucidation of geometrical rules) followed by a known number of points with specific or specified coordinates are guided by the assumption/belief that there must be some thermodynamic and/or kinetic basis leading to the organization of coordinates of the known points. In terms of biomolecular structures, the DNA double helix is one of the strongest examples of such structures with coordinates of atoms in the phosphate backbone and the side-chains provide a helical geometrical configuration. It needs to be appreciated (and emphasized) that,...

Optogenetic control of genetically-targeted pyramidal neuron activity in prefrontal cortex Nature Precedings
Michael V. Baratta; Shinya Nakamura; Peter Dobelis; Matthew B. Pomrenze; Samuel D. Dolzani; Donald C. Cooper.
A salient feature of prefrontal cortex organization is the vast diversity of cell types that support the temporal integration of events required for sculpting future responses. A major obstacle in understanding the routing of information among prefrontal neuronal subtypes is the inability to manipulate the electrical activity of genetically defined cell types over behaviorally relevant timescales and activity patterns. To address these constraints, we present here a simple approach for selective activation of prefrontal excitatory neurons in both in vitro and in vivo preparations. Rat prelimbic pyramidal neurons were genetically targeted to express a...

Hierarchical nanomechanics of collagen microfibrils Nature Precedings
Alfonso Gautieri; Simone Vesentini; Alberto Redaelli; Markus J. Buehler.
Collagen constitutes one third of the human proteome, providing mechanical stability, elasticity and strength to organisms and is thus the prime construction materials in biology. Collagen is also the dominating material in the extracellular matrix and its stiffness controls cell differentiation, growth and pathology. However, the origin of the unique mechanical properties of collagenous tissues, and in particular its stiffness, extensibility and nonlinear mechanical response remains unknown. By using x-ray diffraction data of a collagen fibril (Orgel et al., PNAS, 2006) here we present an experimentally validated model of the nanomechanics of a collagen microfibril that incorporates the full biochemical details of the amino acid sequence of constituting...

Rapid antigen detection using the liquid sample as a lens and self-mixer for light scattering detection Nature Precedings
Antonio Garcia; John Schneider.
Superhydrophobic surfaces, both natural and artificial, cause aqueous drops to bead on them. The most water-repellent surfaces produce nearly perfect spheres with very little contact between water, solutes, and surface due to the roughness of the surface ^1^. However, a related phenomenon that has not been well appreciated until recently, is that colloidal particles in the aqueous drop remain highly mobile, and smaller-sized colloids migrate to the drop surface due to both the driving force of evaporation and the expanded liquid/air interface ^2-3^. A newly identified, third useful optical property resulting from the suspension of an aqueous drop by the superhydrophobic surface is that a spheroidal lens is created. Without any additional energy or...

ECoG-based short-range recurrent stimulation techniques to stabilize tissue at risk of progressive damage: Theory based on clinical observations Nature Precedings
Markus A. Dahlem; Felix M. Schneider; Jens P. Dreier; Anthony J. Strong; Eckehard Schoell.
We introduce theoretical concepts based on chaos control to stabilize in acute stroke the tissue at risk of progressive damage by preventing adverse effects of waves of mass neuronal depolarization. Moreover, we present clinical electrocorticography (ECoG) recordings of relevant signals suggested for the feedback control. The recordings are performed in combination with novel subdural opto-electrode technology for simultaneous laser-Doppler flowmetry in patients with aneurysmal subarachnoid haemorrhage (aSAH). In aSAH patients waves of spreading depolarization (SD) have a high incidence and cause hypoxia in tissue at risk, and, importantly, the haemodynamic response is the inverse of that seen in healthy tissue. In previous clinical studies, clusters of...

Use of inadequate data and methodological errors lead to a dramatic overestimation of the water footprint of Jatropha curcas Nature Precedings
Wouter Maes; Wouter M. J. Achten; Bart Muys.
In their recent article, Gerbens-Leenes et al. (1) calculated the water footprint (WF, the amount of water required to produce 1 GJ of energy) of several bioenergy crops. One of the most remarkable findings of this study was the very high water footprint of this species, which has serious management consequences.

However, these results are in apparent contrast with recent findings on this species. We present evidence that several errors were made by the authors when calculating the water footprint of jatropha, which has lead to a dramatic overestimation. These errors include weaknesses concerning the data used for the calculation of the water footprint, as well as flaws in the calculation method, as we demonstrate in the...

Synchronized Mammalian Cells in Culture: A Comparison of Normal and KB Cancer Cell Growth Nature Precedings
I. C. Baianu.
Experimentation and tests with cells in culture and cell synchronization or synchrony are important in order to be able to make both reliable and high-sensitivity measurements. This is also very important for mechanistic studies of cell division control, oncogenesis and tumor growth. A comparison is made in this article between the initially synchronized cell growth of rabbit and KB cells in synchronized cultures.

Post-market monitoring: legal framework in Brazil and first results Nature Precedings
Marcia A. Melo; Ederson A. Kido; Paulo P. Andrade.
Brazil is presently (2010) the second largest producer of Genetically Modified (GM) crops. The legal framework to support both research activities and the commercial release of Genetically Modified Organisms (GMOs) is, however, very recent, as the Biosafety Law dates back from 2005. After the indispensable risk analysis, done by National Technical Committee for Biosafety (CTNBio), a GM plant can be approved for commercial release. Nevertheless, as stated in the Normative Resolution nr. 5 (NR-5), 2008, the post-market monitoring is mandatory. However, the first GM crop in Brazil, the herbicide tolerant soybean, was harvested in 2005. The CTNBio asked, then, for a monitoring plan and the applicant designed a complex study focused on case-specific monitoring....

Co-transformation of Oil Palm Using Agrobacterium-Mediated Transformation Nature Precedings
Vu Le.
Agrobacterium-mediated transformation is being used as an alternative method for improving the conventional breeding. In this study, immature embryos (IEs) of oil palm are chosen as explants for transformation. β-glucuronidase (gus) and green fluorescence protein (GFP) were harbored by the two plasmids pCAMBIA 1304 and pCAMBIA 1302 respectively. The results were obtained at least four days after transformation. The IEs placing in the N6O medium for 7 days were then co-cultivated with A.tumefaciens suspension within 30 mins and then incubated in the dark for 1 day. In single transformation, GUS assay revealed that about 64% transient transformation frequency was obtained for IEs two weeks after co-cultivation, GFP activity was recorded as 41.67%...
