Summer mortality outbreaks of French Pacific oysters Crassostrea gigas in 2008 and 2009 ArchiMer

Abnormal mortality outbreaks occurred in the majority of Pacific oyster (Crassostrea gigas) producing areas in France in 2008 during the summer period. These outbreaks were usually sudden and severe (up to 100%) and affected essentially spat (oysters less than 1 year-old) and juveniles (12 to 18 month-old oysters). Crassostrea gigas was the single affected species Mortality outbreaks were first observed from mid May to mid June in Brittany, Vendée, Charente Maritime and Mediterranean coast A second episode of massive mortality events was reported at the end of June and the beginning of July. The life histories of oysters in the genus Crassostrea, like those of most marine bivalves, are typified by high fecundity and low survival in nature. Rearing conditions in hatcheries however ensure optimized density, diet, and temperature. Hatcheries are becoming increasingly important for the production of juveniles in aquaculture, and their culture practices often include culling of slow growing larvae to reduce and synchronize the time taken to reach settlement. Single Nucleotide polymorphisms and their relationship to codon usage bias in the Pacific oyster Crassostrea gigas ArchiMer

DNA sequence polymorphism and codon usage bias were investigated in a set of 41 nuclear loci in the Pacific oyster Crassostrea gigas. Our results revealed a very high level of DNA polymorphism in oysters, in the order of magnitude of the highest levels reported in animals to date. A total of 290 single nucleotide polymorphisms (SNPs) were detected, 76 of which being localised in exons and 214 in non-coding regions. Average density of SNPs was estimated to be one SNP every 60 bp in coding regions and one every 40 bp in non-coding regions. Non-synonymous substitutions contributed substantially to the polymorphism observed in coding regions. La cellule de veille zoosanitaire de La Tremblade s'occupe de trois zones situées au sud de la Loire: la Baie de Bourgneuf, la région de Marennes-Oléron-Charentes, et le bassin d'Arcachon. Ces régions produisent principalement des huîtres creuses. Le naissain d'huîtres creuses produit à Arcachon et en Charente représente près de 80% de la production d'huître creuse, qui sera élevée dans toutes les régions ostréicoles françaises. First evidence of the activation of Cg-timp, an immune response component of pacific oysters, through a damage-associated molecular pattern pathway ArchiMer

In a previous work, we characterized a Crassostrea gigas cPNA (Cg-timp) encoding a protein which presents all the features of vertebrate tissue inhibitor of metalloproteinase (TIMP). The expression pattern of this gene led us to propose that Cg-timp is an important factor in oyster wound heating and defense mechanisms. Here we describe the analysis of Cg-timp expression in oysters challenged by live or dead bacteria as well as by bacterial secretory/excretory products and metalloproteinase. The summer mortality patterns was studied in 1996 on the Ronce-les-Bains oyster bank (175 ha), located on the southern part of the Bay Marennes-Oleron (Atlantic coastline). Studies on growth, sexual maturation, survival rates and environmental variables were compared at fifteen experimental sites (on- and off-bottom culture). After 7 months, cumulative mortality reached 8-19 and 23-33% for off-bottom and on-bottom culture, respectively which is significantly higher for the latter case. "Il est impossible d'imaginer un lieu plus favorable à l'ostréiculture que le bassin d'Arcachon" (Yonge, 1950). En effet, l'huître indigène, Ostrea edulis, et l'espèce plus récemment introduite, Crassostrea gigas, peuvent s'y reproduire en abondant. Si l'exploitation de la première a pris fin à la suite de deux épizooties consécutives, la seconde est l'objet d'une culture intensive dont la production annuelle est d'environ 12 000 tonnes de mollusques commercialisés. Optimisation of the traditional Pacific cupped oyster (Crassostrea gigas Thunberg) culture on the French Atlantic coastline: autumnal fattening in semi-closed ponds ArchiMer

Oyster farming in France is a traditional activity. Each year, 60,000 tons of C. gigas are fattened before being sold. Present-day fattening techniques for improving oyster taste and colour and increasing the meat weight are not particularly reliable. To optimize the fattening process, large phytoplanktonic blooms were induced in ponds, then distributed in oyster ponds. Despite the variability of the autumn weather conditions, diatoms (Skeletonema costatum) have been successfully cultured in outdoor ponds. Characterization of Vibrio isolated from Crassostrea gigas spat suffering summer mortality outbreaks (slides) ArchiMer

The pathogens related to summer mortality outbreaks are a herpes virus and two bacterial strains one belonging to Vibrio splendidus biovar II and the other to Vibrio splendidus spp. However, the feature pathogen/opportunist of these strains is still unknown. Several strains belonging to the genus Vibrio have been identified as pathogen for different mollusk species. The authors are comparing the recruitment, the commercialised production, the cultivated biomasses of oysters Crassostrea gigas and Crassostrea angulata with the statistics of the meteorology (1885-1984). The temperature plays a role up on the date of spawning but partialy explains the lack of recruitment, which has less impact on the commercialised production than on the total biomass. The annual and seasonal characteristics of the precipitations and water temperatures do not show tight correlations with the variations of the production and of the biomass, for these cultivated populations in opposition with the sensitivity of the natural molluscan populations towards the climatic factors.
