Dynamics and diversity of the bacterial community during the spontaneous decay of açai (Euterpe oleracea) fruits BJM
Moura,Fábio Gomes; Graças,Diego Assis das; Santos,Agenor Valadares; Silva,Artur Luiz da Costa da; Rogez,Hervé.
Abstract The biodiversity and evolution of the microbial community in açai fruits (AF) between three geographical origins and two spontaneous decay conditions were examined by applying culture-independent methods. Culture-independent methods based on 16S rRNA from fifteen samples revealed that Proteobacteria, Firmicutes, Actinobacteria, Bacteroidetes and Acidobacteria were the most abundant phyla. At the genus level, Massilia (taxon with more than 50% of the sequences remaining constant during the 30 h of decay), Pantoea, Naxibacter, Enterobacter, Raoultella and Klebsiella were identified, forming the carposphere bacterial microbiota of AF. AF is fibre-rich and Massilia bacteria could find a large quantity of substrate for its growth through cellulase...

Detection of shigella in lettuce by the use of a rapid molecular assay with increased sensitivity BJM
Jiménez,Kenia Barrantes; McCoy,Clyde B.; Achí,Rosario.
A Multiplex Polymerase Chain Reaction (PCR) assay to be used as an alternative to the conventional culture method in detecting Shigella and enteroinvasive Escherichia coli (EIEC) virulence genes ipaH and ial in lettuce was developed. Efficacy and rapidity of the molecular method were determined as compared to the conventional culture. Lettuce samples were inoculated with different Shigella flexneri concentrations (from 10 CFU/ml to 10(7) CFU/ml). DNA was extracted directly from lettuce after inoculation (direct-PCR) and after an enrichment step (enrichment PCR). Multiplex PCR detection limit was 10(4) CFU/ml, diagnostic sensitivity and specificity were 100% accurate. An internal amplification control (IAC) of 100 bp was used in order to avoid false...

Ultrasound pretreatment as an alternative to improve essential oils extraction Ciência Rural
Nora,Flávia Michelon Dalla; Borges,Caroline Dellinghausen.
ABSTRACT: Essential oils are substances originated from plants in general. These compounds are well known to have a high biological activity, specially the antioxidant and antimicrobial. Several extraction techniques are employed to obtain these substances. However, the majority of these techniques require a long extraction time. In this sense, innovative and alternative extraction techniques, such as ultrasound, have recently been the target of studies. In view of the small amount of publications using ultrasonic pretreatment, this review aimed to congregate current relevant information on ultrasound-assisted extraction of essential oils. In this sense, theoretical aspects, such as the main factors that influence the performance of this technique as well...

Occurrence and molecular characterization of different virulence-associated genes of Cronobacter sakazakii isolates from some foods and dust samples Ciência Rural
Demirci,Ülkü; Tekiner,İsmail Hakkı; Çakmak,Burcu; Özpınar,Haydar.
ABSTRACT: Among the Cronobacter genus, Cronobacter sakazakii is the most common species posing a severe health risk for newborns, infants and children. Some infant formulas, cereal-based foods, and food production environments may be the potential reservoirs of C. sakazakii. This pathogen possesses different virulence factors encoded by different virulence genes. Therefore, characterizing these genes is important for distinguishing pathogenic strains from nonpathogenic ones. The objective of this study was to characterize some virulence genes [OmpA, OmpX, zpx, and Cpa] by real-time polymerase chain reaction (PCR) in C. sakazakii isolates from a total of 120 samples (20 each of milk powder, starch, rice flour, semolina, infant formula and dust samples from...

Mixing models and stable isotopes as tools for research on feeding aquatic organisms Ciência Rural
Moraes,Cyntia Rafaela Ferreira de; Henry-Silva,Gustavo Gonzaga.
ABSTRACT: Implementation of mixture models in isotopic analysis has been increasing due to the need to analyze sources and understand their variations in relation to consumers, which requires an accurate determination of their discriminant rate and fractioning. In this context, we presented the main concepts related to the application of stable isotopes and mixing models, with the aim to analyze the benefits and problems of these tools in surveys on nutrition of aquatic organisms. The predominant methods identified included application of the models IsoSource, MixSIR, SIBER and SIAR, with the use of C and N isotopes in research with aquatic organisms. Studies in both freshwater and marine environments were evaluated to determine the relative contribution...
