In order to understand the temporal dynamics of actinomycete communities of the rhizosphere of the desert plant Artemisia tridentata (sagebrush), two complementary methods were used. They were: (1) 16S rDNA-based PCR coupled with denaturing gradient gel electrophoresis (DGGE), and (2) an agar plate enumeration methodology in which three different media were used to quantify total bacteria, actinomycetes, and fungi. The objective of this research were: (1) to obtain a comprehensive picture of the structure of actinomycete populations, and (2) their dynamics in the rhizosphere of young and old sagebrush plants during two distinct seasons. PCR-DGGE analysis showed that actinomycete groups were less diverse in rhizosphere soils collected in winter than in bulk... |

It is well-known that Amazon tropical forest soils contain high microbial biodiversity. However, anthropogenic actions of slash and burn, mainly for pasture establishment, induce profound changes in the well-balanced biogeochemical cycles. After a few years the grass yield usually declines, the pasture is abandoned and is transformed into a secondary vegetation called "capoeira" or fallow. The aim of this study was to examine how the clearing of Amazon rainforest for pasture affects: (1) the diversity of the Bacteria domain evaluated by Polymerase Chain Reaction and Denaturing Gradient Gel Electrophoresis (PCR-DGGE), (2) microbial biomass and some soil chemical properties (pH, moisture, P, K, Ca, Mg, Al, H + Al, and BS), and (3) the influence of... |

We tested if the microbial functional redundancy concept (theory relating changes in ecosystem functioning by species loss) apply for changes in the ammonium-oxidizing community (nitrifier bacteria) due to: a) grazing disturbance, b) seasonal variations (dry season vs. wet season), c) habitat type (soil vs. organic residues), and d) ecological conditions (two eco-regions), in an arid region of central-western of Argentina. We determined: a) abundance of culturable nitrifier bacteria, b) nitrification activity and efficiency, and c) nitrifier community genetic structure by PCR-DGGE (richness and similarity index). Grazing did not change the nitrifier abundance, activity and richness, while seasonally and eco-region effects were scarce. Contrarily, the... |

Polycyclic aromatic hydrocarbons (PAHs) constitute a group of priority pollutants which are present at high concentrations in the soils of many industrial contaminated sites. Pollution by these compounds may stimulate growth of organisms able to live in these environments causing changes in the structure of the microbial community due to some cooperative process of metabolization of toxic compounds. A long-term PAH-contaminated soil was stored for several years and used to analyze the native microbiota regarding their ability to grow on pyrene, benzo[a]pyrene, as well as in mixtures of LMW- and HMW-PAHs. Molecular profiles of the microbial community was assessed by PCR-DGGE of 16S rRNA gene, and the number of bands observed in DGGE analyses was interpreted... |

Abstract Shenqu is a fermented product that is widely used in traditional Chinese medicine (TCM) to treat indigestion; however, the microbial strains in the fermentation process are still unknown. The aim of this study was to investigate microbial diversity in Shenqu using different fermentation time periods. DGGE (polymerase chain reaction-denaturing gradient gel electrophoresis) profiles indicated that a strain of Pediococcus acidilactici (band 9) is the predominant bacteria during fermentation and that the predominant fungi were uncultured Rhizopus, Aspergillus oryzae, and Rhizopus oryzae. In addition, pathogenic bacteria, such as Enterobacter cloacae, Klebsiella oxytoca, Erwinia billingiae, and Pantoea vagan were detected in Shenqu. DGGE analysis... |
