Cytokinin preconditioning enhances multiple shoot regeneration in Pongamia pinnata (L.) Pierre - a potential, non-edible tree seed oil source for biodiesel 69
Belide,Srinivas; Sajjalaguddam,Ramgopal Rao; Paladugu,Anitha.
An efficient, highly reproducible protocol for multiple shoot induction and plant regeneration of Pongamia pinnata has been successfully developed using cotyledonary node explants. This study also demonstrates that preconditioning of explant stimulates production of multiple shoots from cotyledonary nodes of P. pinnata. The highest direct shoot regeneration (90%) with an average of 18.4 3.1 shoots/explant were obtained when cotyledonary node explants were excised from seedlings germinated on Murashige and Skoog (MS) media supplemented with benzyladenine (BA) 1 mg l-1, and subsequently cultured on MS media with 1 mgl-1 thidiazuron (TDZ). Scanning electron microscope observations of cotyledonary node (CN) explants excised from pre-conditioned and normal...

Micropropagation of Homalomena pineodora Sulaiman & Boyce (Araceae): a new species from Malaysia 75
Stanly,Christine; Bhatt,Arvind; Sulaiman,Baharuddin; Keng,Chan Lai.
Homalomena pineodora (family Araceae) is a species found to have impressive foliage characteristics which remain evergreen throughout the year. Therefore, H. pineodora can be grown as an ornamental plant. Generally H. pineodora needs 3-5 years to propagate and multiply. However, the demand for new ornamental plants is increasing worldwide and the quality of planting material is a basic need for boosting productivity. Therefore an efficient micropropagation protocol for large-scale production of H. pineodora was developed. In vitro shoot cultures were initiated from the rhizomatous buds on MS basal medium. The best conditions for propagating H. pineodora was found to be MS medium supplemented with 3% sucrose and 0.5 mg L-1 BA (6-benzyladenine) under 24 h of...

Updating Geographic Distribution of Artemia urmiana GÜNTHER, 1890 (Branchiopoda: Anostraca) in Europe: An Integrated and Interdisciplinary Approach 13
Abatzopoulos, Th. J.; Amat, F.; Baxevanis, A. D.; Belmonte, G.; Hontoria, F.; Maniatsi, S.; Moscatello, S.; Mura, G.; Shadrin, N. V..
Artemia urmiana (a species previously considered endemic of Lake Urmia, NW Iran) has been found in Lake Koyashskoe, a hypersaline lake on the Black Sea coast of the Crimean peninsula (Ukraine). Therefore, this is the first record of A. urmiana in Europe which updates its distribution. The species identification was based on an integrated and interdisciplinary approach using discriminant analysis of the morphometric characters, scanning electron microscopy, and molecular profile analysis. The data derived from the above mentioned approaches converge to significant similarity of the population under investigation with A. urmiana. The updated geographic distribution of the species, deriving from the present report, asks for additional contribution of other...

Etude du biofilm microalgal des vasières intertidales : dynamique spatio-temporelle à micro-échelle et performances photosynthétiques 5
Herlory, Olivier.
The microalgal biofilm, which is present on sediment surface, is the primary production unit on bare intertidal mudlfats. In order to improve our understanding of this primary production system, a study was undertaken at microscale to analyse the structural and functional properties of this biofilm. In the first part of this work, a detailed descriptive analysis lead to the characterisation of the spatiotemporal dynamics of the physical structure of the biofilm. It turns out that the biofilm exhibits its own dynamics at the sediment surface (as a discrete compartment in the top 200 µm), but still has a functional relationship with the total available biomass (PCB) distributed within the top 1 cm (linear relationship between the maximum size of the biofilm...

Polystyrene microbeads modulate the energy metabolism of the marine diatom Chaetoceros neogracile 5
Seoane, Marta; González-fernández, Carmen; Soudant, Philippe; Huvet, Arnaud; Esperanza, Marta; Cid, Ángeles; Paul-pont, Ika.
Due to the growing concern about the presence of microplastics (MP) in the environment, the number of studies evaluating the toxicity of these small persistent particles on different marine species has increased in recent years. Few studies have addressed their impact on marine phytoplankton, a subject of great concern since they are primary producers of the aquatic food web. The aim of this study is to unravel the cytotoxicity of 2.5 μg mL−1 unlabelled amino-modified polystyrene beads of different sizes (0.5 and 2 μm) on the marine diatom Chaetoceros neogracile. In addition to traditional growth and photosynthesis endpoints, several physiological and biochemical parameters were monitored every 24 h in C. neogracile cells by flow cytometry during their...

Mineral phase in shell repair of Manila clam Venerupis philippinarum affected by brown ring disease 5
Trinkler, Nolwenn; Bardeau, Jean-francois; Marin, Frederic; Labonne, Maylis; Jolivet, Aurelie; Crassous, Philippe; Paillard, Christine.
The mineral phase of shell repair in the Manila clam Venerupis philippinarum affected by brown ring disease (BRD) was characterised at various scales and at various stages of shell repair by confocal Raman microspectrometry and scanning electron microscopy. Spherulitic and quadrangular aragonite microstructures associated with polyene pigments were clearly observed. Von Kossa staining showed that at the beginning of shell repair, hemocytes are filled with insoluble calcium carbonate salts in all fluids and then are transported toward the extrapallial fluids and the repair sites. Our analyses suggest that after a Vibrio tapetis attack and BRD deposit some clams rapidly cover the deposit, resulting in a modification in the microstructure, which could be...

Comparison of the physical and geotechnical properties of gas-hydrate-bearing sediments from offshore India and other gas-hydrate-reservoir systems 5
Winters, W. J.; Wilcox-cline, R. W.; Long, P.; Dewri, S. K.; Kumar, P.; Stern, L.; Kerr, L..
The sediment characteristics of hydrate-bearing reservoirs profoundly affect the formation, distribution, and morphology of gas hydrate. The presence and type of gas, porewater chemistry, fluid migration, and subbottom temperature may govern the hydrate formation process, but it is the host sediment that commonly dictates final hydrate habit, and whether hydrate may be economically developed. In this paper, the physical properties of hydrate-bearing regions offshore eastern India (Krishna-Godavari and Mahanadi Basins) and the Andaman Islands, determined from Expedition NGHP-01 cores, are compared to each other, well logs, and published results of other hydrate reservoirs. Properties from the hydrate-free Kerala-Konkan basin off the west coast of India are...

Recovery in culture of viable but nonculturable Vibrio parahaemolyticus: regrowth or resuscitation? 5
Coutard, Francois; Crassous, Philippe; Droguet, Michael; Gobin, Eric; Colwell, Rita; Pommepuy, Monique; Hervio Heath, Dominique.
The objective of this study was to explore the recovery of culturability of viable but nonculturable ( VBNC) Vibrio parahaemolyticus after temperature upshift and to determine whether regrowth or resuscitation occurred. A clinical strain of V. parahaemolyticus Vp5 was rendered VBNC by exposure to artificial seawater ( ASW) at 4 degrees C. Aliquots of the ASW suspension of cells ( 0.1, 1 and 10 ml) were subjected to increased temperatures of 20 degrees C and 37 degrees C. Culturability of the cells in the aliquots was monitored for colony formation on a rich medium and changes in morphology were measured by scanning ( SEM) and transmission ( TEM) electron microscopy. Samples of VBNC cells were fixed and examined by SEM, revealing a heterogeneous population...

Bioprotective Effect of Lactococcus piscium CNCM I-4031 Against Listeria monocytogenes Growth and Virulence 5
Saraoui, Taous; Leroi, Francoise; Chevalier, Frederique; Cappelier, Jean-michel; Passerini, Delphine; Pilet, Marie-france.
Listeria monocytogenes is a Gram-positive pathogen occurring in many refrigerated ready-to-eat foods. It is responsible for foodborne listeriosis, a rare but severe disease with a high mortality rate (20–30%). Lactococcus piscium CNCM I-4031 has the capacity to prevent the growth of L. monocytogenes in contaminated peeled and cooked shrimp and in a chemically defined medium using a cell-to-cell contact-dependent mechanism. To characterize this inhibition further, the effect of L. piscium was tested on a collection of 42 L. monocytogenes strains. All strains were inhibited but had different sensitivities. The effect of the initial concentration of the protective and the target bacteria revealed that the inhibition always occurred when L. piscium had reached...

Comparative Scanning Electron Microscopy of Platelets and Fibrin Networks of Human and Differents Animals 77
Pretorius,E; Vieira,W. A; Oberholzer,H. M; Auer,R. E. J.
The purpose of the present study was to compare the platelet and fibrin network ultrastructure of humans to eight different animal species in order to determine the differences between human and animal platelet and fibrin morphology, and to determine whether the animals studied differ in their platelet and fibrin morphology, and whether these differences can be observed by scanning electron microscopy. Platelets and fibrin networks play an important role both in the coagulation process as well as physiologically in allergic processes and immunological mechanisms. The thickness of human fibrin networks were compared to mouse (Mus musculus), equine (Equus caballus), vervet monkey (Chlorocebus aethiops previously Cercopithecus aethiops), oryx (Oryx gazella),...

Preservation of Haemal Nodes Fixed by Means of Two Different Aldehyde Fixatives: a Scanning Electron Microscopic Study 77
Guerrero,F; Cerutti,P; Marcaccini,A.
Glutaraldehyde is the fixative most commonly used in electron microscope studies of biological tissues, however it is often necessary to use samples which were not fixed in this fixative, even with the usual uncertainty of the results that may be obtained. The fixation is the more delicate step of the sample processing. Therefore in this work, the quality of preservation of haemal nodes fixed with two classic aldehyde fixatives: formaldehyde and glutaraldehyde we have compared under the scanning electron microscope. Our results showed that both fixatives were successful in preserving the morphology of haemal nodes components; however glutaraldehyde conferred satisfactory results mainly in the preservation of parenchymal cells, whereas formaldehyde was...

Scanning Electron Microscopy of the Gastrointestinal Tract of Nile Perch (Lates niloticus, Linneaus, 1758) 77
Namulawa,Victoria Tibenda; Kato,Charcles Drago; Nyatia,Edward; Rutaisire,Justus; Britz,Peter.
The morphology of the gastrointestinal tract of adult Nile perch was described using standard SEM procedures. Investigations revealed the presence of cardiform teeth in the oral cavity, goblet cells and finger print-like microridges on the hard palate and oesophagus lumenal surface. Elaborate patterns and bacterial cells were observed on the stomach lumenal surface and intense foldings in the intestinal region. These observations provide a better understanding of the morphology of the gut in Nile perch and how it is suited for its digestive function.

Gill Morphology in Two Bottom Feeder Mediterranean Sea Fishes: Grey Gurnard Fish (Eutrigla gurnardus, Linnaeus, 1758) and Striped Red Mullet Fish (Mullus barbatus surmuletus, Linnaeus, 1758) by Scanning Electron Microscopy 77
Abumandour,Mohamed M. A; El-Bakary,Neveen E. R.
The purpose of the current investigation was to describe the gill morphology of two bottom feeders Mediterranean Sea fishes: Striped red mullet fish (M. surmuletus) and grey gurnard fish (E. gurnardus) that showed the same feeding habits. The gill system of the grey gurnard consisted of four pairs of gill arches while consisted of three pairs of gill arches in the striped red mullet. Our study focused on the scanning electron microscopical (SEM) features of the gills, where some differences in the gill arches and gill rakers surface in both species were observed. Our results marked that there was an interbranchial septum carrying a four transverse elevated crest in grey gurnard while in striped red mullet carrying a median longitudinal elevated crest....

Vestured pits: a diagnostic character in the secondary xylem of Myrtales 16
Jansen, S.; Pletsers, A.; Rabaey, D.; Lens, F..
Vestures are small projections from the secondary cell wall associated with tracheary elements of the secondary xylem. They are usually associated with bordered pits and characterize various angiosperm families, including important timber species such as Dipterocarpaceae and Eucalyptus trees. The micromorphology and distribution of vestures were studied in 22 species representing all families within the order Myrtales based on light and scanning electron microscopy. Vestures are consistently present near the outer pit aperture of bordered vessel pits, suggesting the synapomorphic character of this feature for the entire order. It is unclear in which geological period this feature originated in the evolution of the pre-Myrtalean lineages. In some species...

The phylogenetic significance of vestured pits in Boraginaceae 16
Rabaey, D.; Lens, F.; Smets, E.; Jansen, S..
The bordered pit structure in tracheary elements of 105 Boraginaceae species is studied using scanning electron microscopy to examine the systematic distribution of vestured pits. Forty-three species out of 16 genera show a uniform presence of this feature throughout their secondary xylem. Most vestures are small, unbranched and associated with the outer pit aperture of bordered intervessel pits. The feature is likely to have originated independently in the distantly related subfamilies Boraginoideae (tribe Lithospermeae) and Ehretioideae. The distribution of vestures in Ehretia agrees with recent molecular phylogenies: (1) species with vestured pits characterise the Ehretia I group (incl. Rotula), and (2) species with non-vestured pits belong to the...

Sodiomyces alkalinus, a new holomorphic alkaliphilic ascomycete within the Plectosphaerellaceae 16
Grum-Grzhimaylo, A.A.; Debets, A.J.M.; Diepeningen, A.D. van; Georgieva, M.L.; Bilanenko, E.N..
In this study we reassess the taxonomic reference of the previously described holomorphic alkaliphilic fungus Heleococcum alkalinum isolated from soda soils in Russia, Mongolia and Tanzania. We show that it is not an actual member of the genus Heleococcum (order Hypocreales) as stated before and should, therefore, be excluded from it and renamed. Multi-locus gene phylogeny analyses (based on nuclear ITS, 5.8S rDNA, 28S rDNA, 18S rDNA, RPB2 and TEF1-alpha) have displayed this fungus as a new taxon at the genus level within the family Plectosphaerellaceae, Hypocreomycetidae, Ascomycota. The reference species of actual Heleococcum members showed clear divergence from the strongly supported Heleococcum alkalinum position within the Plectosphaerellaceae, sister...
