La multiplicación de plantas in vitro presenta varias etapas críticas, desde la selección de los componentes para la germinación, multiplicación y enraizamiento hasta el proceso de aclimatación en invernadero. En la presente investigación se utilizaron los sustratos inertes (vermiculita, agrolita y tezontle) como medio de soporte en sustitución del agar y los azúcares xilosa, fructosa y 10X20F se compararon con la sacarosa, en su efecto sobre la germinación, multiplicación, enraizamiento y sobrevivencia de los portainjertos de cítricos: Citrus volkameriana, Citrumelo swingle y Citrange 35. We evaluated the effects of saline stress on soluble proteins, lipid peroxidation (TBAR), chlorophyll a, chlorophyll b, β-carotene, violaxanthin, and proline in Paulownia imperialis and Paulownia fortunei plants grown in vitro. When the propagated plants reached a determined size, they were transferred aseptically to WPM culture medium containing different sodium chloride concentrations (0, 20, 40, 60, 80, and 160 mM) and were sampled at 15 and 30 days. Proline content was determined at 30 days after transfer only. Protein concentration significantly decreased with the highest salt levels in P. imperialis compared to controls in which no sodium chloride was added. Morphological, physiological and molecular changes were investigated in in vitro salt-stressed barley (Hordeum vulgare L. cv. Tokak). Mature embryos were cultured in Murashige and Skoog medium containing 0 (control), 50 and 100 mM NaCl for 20 days. Both concentrations inhibited shoot growth, decreased fresh weight and protein content, and increased SOD (EC 1.15.1.1) activity in a dose-dependent manner. The lower concentration increased root growth. Salinity caused nucleotide variations in roots, but did not affect shoot DNAs. The higher concentration caused methylation changes, mainly hypermethylation in shoots. This is the first study on genetic and epigenetic effects of salinity in barley. |

Aechmea blanchetiana and Aechmea distichantha are Brazilian native bromeliads with great ornamental value, although none of the species are produced commercially. Therefore, market demands are met through nature predatory exploitation. Garlic cultivars are sexually sterile under standard growth conditions, with direct implications for commercial production costs as well as breeding programs. Garlic is propagated commercially via bulblets, which facilitates disease transmission and virus load accumulation over vegetative generations. One of the most important applications of plant tissue culture is mass propagation of ornamental plants. This experiment evaluated the effect of different concentrations of NH4NO3 and KNO3 and BAP on the in vitro growth of orchid hybrid Brassocattleya 'Pastoral'. Seedlings of this orchid hybrid were used as explants and cultivated in medium with mineral salts and vitamins from the MS medium (Murashige & Skoog, 1962), with the macronutrients P, Ca and Mg reduced by half, and with an addition of 25 g L-1 of sucrose, 0.1 g L-1 of myo-inositol and 1.5 g L-1 of activated charcoal. O objetivo deste trabalho foi estabelecer e multiplicar, sob condições de cultivo in vitro, um acesso de faveleira sem espinhos, visando alternativas para sua conservação e uso futuro. Para isso, segmentos nodais e fragmentos de folhas jovens foram inseridos em meio nutritivo composto pela formulação de sais inorgânicos WPM.
