Molecular cytogenetic characterization of parental genomes in the partial amphidiploid Triticum aestivum x Thinopyrum ponticum 74
Brasileiro-Vidal,Ana Christina; Cuadrado,Angeles; Brammer,Sandra P.; Benko-Iseppon,Ana Maria; Guerra,Marcelo.
The wheat line PF 839197 and six hybrid derivatives from a cross between PF 839197 and Thinopyrum ponticum were cytologically characterized by fluorescent in situ hybridization (FISH). Probes for the 5S and 45S rDNA genes (pTa794 and pTa71, respectively), a highly repetitive rye sequence (pSc119.2), the synthetic oligonucleotide (AAG)5, and total genomic DNA from Th. ponticum and rye were used. In the wheat line, a 1RS.1BL translocation was revealed by the labeling patterns produced with pSc119.2 and (AAG)5, and confirmed by genomic in situ hybridization (GISH) using rye genomic DNA as a probe. Analyses of partial amphiploids confirmed previous results indicating mitotic instability, with a tendency to stabilize at 2n = 42 or 56. GISH with Th. ponticum...

Genome composition and pollen viability of Jatropha (Euphorbiaceae) interspecific hybrids by Genomic In Situ Hybridization (GISH) 74
Souza,Rosilda Cintra de; Marques,Daniela de Argollo; de Carvalho Filho,Marcel Mamede; Oliveira,Ana Rafaela da Silva; Siqueira,Walter José; Benko-Iseppon,Ana Maria; Brasileiro-Vidal,Ana Christina.
Abstract Interspecific hybridization is required for the development of Jatropha curcas L. improved cultivars, due to its narrow genetic basis. The present study aimed to analyze the parental genomic composition of F1 and BC1F1 generations derived from interspecific crosses (J. curcas/J. integerrima and J. curcas/J. multifida) by GISH (Genomic In Situ Hybridization), and the meiotic index and pollen viability of F1 hybrids. In F1 cells from both hybrids, 11 chromosomes of each parental was observed, as expected, but chromosome rearrangement events could be detected using rDNA chromosome markers, suggesting unbalanced cells. In the BC1F1, both hybrids had 22 chromosomes, suggesting that only n = 11 gametes were viable in the next generation. However, GISH...

Genotoxic potential of the latex from cotton-leaf physicnut (Jatropha gossypiifolia L.) 74
de Almeida,Pedro Marcos; de Sousa Araújo,Silvany; Marin-Morales,Maria Aparecida; Benko-Iseppon,Ana Maria; Brasileiro-Vidal,Ana Christina.
Jatropha gossypiifolia L. (Euphorbiaceae), popularly known as cotton-leaf physicnut, is a milky shrub notable for its medicinal properties. The present study aimed to evaluate the toxic, cytotoxic and genotoxic effects of the latex of J. gossypiifolia, using Allium cepa L. as test system. Seeds of A. cepa were exposed to five concentrations of the latex (1.25; 2.5; 5; 10 and 20 mL/L) in order to evaluate parameters of toxicity (evaluation of root growth), cytotoxicity (mitotic index frequency) and genotoxicity (frequency of chromosome alterations). The latex showed a significant decrease in root mean growth value as well as mitotic index for the tested concentrations, except for 1.25 mL/L, when compared to results from the negative control. The 1.25, 2.5...

Karyotype and genome size comparative analyses among six species of the oilseed-bearing genus Jatropha (Euphorbiaceae) 74
Marinho,Anne C.T.A.; Vasconcelos,Santelmo; Vasconcelos,Emanuelle V.; Marques,Daniela A.; Benko-Iseppon,Ana Maria; Brasileiro-Vidal,Ana Christina.
Abstract Jatropha is an important genus of Euphorbiaceae, with species largely used for various purposes, including the manufacturing of soaps and pharmaceutical products and applications in the bioenergetic industry. Although there have been several studies focusing J. curcas in various aspects, the karyotype features of Jatropha species are poorly known. Therefore, we analyzed six Jatropha species through fluorochrome staining (CMA/DAPI), fluorescent in situ hybridization (FISH) with 5S and 45S rDNA probes and genome size estimation by flow cytometry. Our results revealed several chromosome markers by both CMA/DAPI and FISH for the analyzed species. Five Jatropha species (J. curcas, J. gossypiifolia, J. integerrima, J. multifida and J. podagrica) showed...

First molecular analysis of the genus Bryopsis (Bryopsidales, Chlorophyta) from Brazil, with an emphasis on the Pernambuco coast 39
Oliveira,Marcella Guennes Tavares de; Pereira,Sonia Maria Barreto; Benko-Iseppon,Ana Maria; Balbino,Valdir Queiroz; Silva Junior,Wilson José da; Ximenes,Caroline Feijão; Carvalho,Maria de Fátima de Oliveira; Cassano,Valéria.
ABSTRACT The green algal genus Bryopsis has simple morphology and high phenotypic plasticity, making it difficult to identify its species based on morphological characteristics alone. This study evaluated the diversity of Bryopsis in northeastern Brazil (the State of Pernambuco), based on morphological and molecular data using the markers tufA and rbcL. Molecular analyses were incongruent with morphology, demonstrating the existence of cryptic and polymorphic species in the genus. Of the four taxa cited for the area based only on morphological data (Bryopsis corymbosa, B. pennata, B. plumosa, Bryopsis sp.), only B. pennata was recorded. Typical specimens of B. pennata and B. plumosa were grouped with low genetic divergence, 0-0.21 % for tufA and no...

Evidence of genetic differentiation and karyotype evolution of the sedges Cyperus ligularis L. and C. odoratus L. (Cyperaceae) 39
Cruz,Geyner Alves dos Santos; Vasconcelos,Santelmo; Pinangé,Diego Sotero de Barros; Gonçalves-Oliveira,Rodrigo Cesar; Alves,Marccus Vinícius; Torres,Rodrigo Augusto; Benko-Iseppon,Ana Maria.
ABSTRACT The taxonomy of Cyperaceae is complex, with genera like Cyperus harboring species complexes. We analyzed the genetic similarity between Cyperus ligularis L. and C. odoratus L. based on DNA fingerprinting and cytogenetics. Significative genetic differentiation (G ST = 0.363) and low gene flow (N m = 0.877) indicated a clear genetic distinction between the two species. Moreover, the clustering analysis showed two distinct genetic groups, suggesting a lack of evidence for hybridization. The phenogram revealed two different lineages, and although all individuals of C. odoratus were collected from plots close to each other, they possessed greater genetic diversity than that observed among individuals of C. ligularis, which were sampled over a wider...
